Relationship between atrial natriuretic peptide-immunoreactive cells and microvessels in rat gastric mucosa

Aim:To investigate the ultrastructural localization of atrial natriuretic peptide(ANP)-synthesizing cells and the relationship between ANP-synthesizing cellsand microvessels in rat gastric ***:Immunohistochemistry tech-niques and postembedding immunoelectron microscopy techniques were used tovalidate the findings regarding the expression of ANP-synthesizing cells and theultrastructural localization of ANP-synthesizing cells in the gastric ***-tochemistry techniques and the tannic acid-ferric chloride method(TA-Fe stain-ing method)were used to reveal microvessel density and the distribution of ANP-synthesizing cells in different regions of the ***:cells expressingANP were localized and ANP-synthesizing cells were identified as enterochroma-ffin(EC)cells in the gastric ***-synthesizing cells existed in differentregions of the *** percentage ANP-synthesizing cells in the mucosawas greatest in the fundus(46.7%e5.3%),intermediate in the antrum(40.1%±4.5%),and least in the body(21.6%±3.6%).There was a positive relationshipbetween the percentage of ANP-synthesizing cells and the density of microvesselsin the antral mucosa,but not in the fundus or body ***:ANP issynthesized by EC cells in rat gastric mucosa,and ANP-synthesizing cells aremost dense in the gastric *** may act not only as a regional autocrineand/or paracrine regulator,but also as an endocrine regulatory peptide in thegastrointestinal tract.

Midgut neuroendocrine tumor presenting with acute intestinal ischemia

Neuroendocrine tumors represent a heterogeneous group of neoplasms that arise from neuroendocrine cells and secrete various peptides and bioamines. While gastrointestinal neuroendocrine tumors,commonly called carcinoids,account for about 2/3 of all neuroendocrine tumors,they are relatively rare. Small intestine neuroendocrine tumors originate from intestinal enterochromaffin cells and represent about 1/4 of small intestine neoplasms. They can be asymptomatic or cause nonspecific symptoms,which usually leads to a delayed diagnosis. Imaging modalities can aid diagnosis and surgery remains the mainstay of treatment. We present a case of a jejunal neuroendocrine tumor that caused nonspecific symptoms for about 1 year before manifesting with acute mesenteric ischemia. Abdominal X-rays revealed pneumatosis intestinalis and an abdominal ultrasound and computed tomography confirmed the diagnosis. The patient was submitted to segmental enterectomy. Histopathological study demonstrated a neuroendocrine tumor with perineural and arterial infiltration and lymph node metastasis. The postoperative course was uneventful and the patient denied any adjuvant treatment.

The hyperpolarisation activated c AMP-gated cation channels(HCNs)belong to the cyclic nucleotide-gated(CNG)channel family and consist of four subtypes,*** channels are known to be expressed in the heart where they play important roles in the generation of the cardiac *** of HCNs are also expressed in non-cardiac *** have previously shown that HCN2 and HCN3 were differentially expressed in spinal and vagal primary afferent neurons of the small *** blockers attenuated the vagal component of the mesenteric afferent response to distension of the normal ***,HCNs appeared to play a role in priming the vagal but not spinal afferents in the normal *** this work we further investigated the possible expression and function of HCNs within the *** staining showed HCNs being distinctively expressed in the enterochromaffin(EC)cells of the mice,the rat and the human *** then investigated the possible role of HCNs in regulation of 5-HT release through real-time amperometric measurement in an isolated murine gut *** AMP was shown to increase 5-HT release and the mesenteric afferent nerve *** effect was reversible by HCN blockers,ZD7288 and Cs Cl,which could also inhibit the basal 5-HT release.5-HT3 receptor antagonist,tropisetron,also blocked the afferent nerve response to c *** cholera toxin(CT)is known to increase c AMP level and 5-HT release,we tested whether activation of HCNs might underlie CT-induced 5-HT release and afferent nerve *** expected,CT caused a slowly developing increase in 5-HT release and afferent nerve activity,which were reversed by ***,tropisetron was able to inhibit CT-induced afferent *** results suggest that HCNs drive serotonin release and afferent nerve activity in the *** propose HCNs might represent a novel therapeutic target for inflammatory or functional bowl diseases.
