Bullous pemphigoid (BP) is the most prevalent autoimmune sub-epidermal blistering disease that affects mainly the elderly and could lead to serious morbidity. It has numerous risk factors and triggers, including an aging population with several comorbidities and drug exposure. In the published paper, we reported a case about a 32 years old male patient with unknown medical conditions who presented with erythematous patches and plaques on the scalp, face, and trunk with scattered blisters two weeks after initiating doxycycline treatment for folliculitis. The exact pathogenesis of the drug-reaction in drug- associated bullous pemphigoid (DABP) remains controversial. In conclusion, it is crucial for clinicians to be aware of DABP when prescribing doxycycline. The purpose of this case report is to raise awareness of the possible association between bullous pemphigoid and doxycycline.

doxycycline （DOX） exhibits anti-inflammatory and MMP inhibitory properties. The objectives of this study were to evaluate the effects of DOX on alveolar bone repair. Controls （CTL） and DOX-treated （10 and 25 mg. kg- 1） molars were extracted, and rats were killed 7 or 14 days later. The maxillae were processed and subjected to histological and immunohistochemical assays. Hematoxylin-eosin staining （7th day） revealed inflammation in the CTL group that was partly reversed after DOX treatment. On the 14th day, the CTL group exhibited bone neoformation, conjunctive tissue, re-epithelization and the absence of inflammatory infiltrate. DOX-treated groups exhibited complete re-epithelization, tissue remodelling and almost no inflammation. Picrosirius red staining in the DOXlO group （7th and 14th days） revealed an increased percentage of type I and III collagen fibres compared with the CTL and DOX25 groups. The DOXlO and DOX25 groups exhibited increases in osteoblasts on the 7th and 14th days. However, there were fewer osteoclasts in the DOXlO and DOX25 groups on the 7th and 14th days. Wnt-lOb- immunopositive cells increased by 130% and 150% on the 7th and 14th days, respectively, in DOX-treated groups compared with the CTL group. On the 7th day, Dickkopf （Dkk）-I immunostaining was decreased by 63% and 46% in the DOXlO and DOX25 groups, respectively. On the 14th day, 69% and 42% decreases in immunopositive cells were observed in the DOXlO and DOX25 groups, respectively, compared with the CTL group. By increasing osteoblasts, decreasing osteoclasts, activating Wnt lOb and neutralising Dkk, DOX is a potential candidate for bone repair in periodontal diseases.

Aim:To study the effect of doxycycline （DOX） on osteoclastogenesis,matureosteoclast fate and function,wear particles-induced osteoeolysis,and to providesome foundation for treating aseptic loosening and osteolysis after jointarthroplasty.Methods:Osteoclasts were generated from mouse bone marrowmonocytes with the receptor activator of NF-kB ligand and the macrophage colonystimulating factor.DOX at a concentration of 5,10,15,and 20 μg/mL was respec-tively added to the medium.Seven days later,the osteoclasts were determinedthrough tartrate-resistant acid phosphatase （TRAP） staining.Mature osteoclastswere isolated from newborn rabbits and cultured for 3 d in 24-well plates or onbone slices.DOX at a concentration of 5,10,15,and 20 μg/mL was respectivelyadded to the medium.After TRAP staining,the osteoclasts were counted,resorp-tion on bone slices was quantified,and the area was calculated after toluidine blueand Mayer-hematoxylin staining.Polymethyl methacrylate （PMMA） or ultra-highmolecular weight polyethylene （UHMWPE） particles were implanted on thecalvariae of C57BL/J6 mice.DOX,at a dose of 2 and 10 mg·kg-1·d-1,was respec-tively given intraperitoneally for 7 d.Seven days later,the calvariae were removedand processed for pathological analysis.Results:DOX treatment effectivelyinhibited in vitro osteoclastogenesis,affected the fate of mature osteoclasts,andinhibited mature osteoclasts,causing bone resorption.In vivo data indicated thatDOX strongly inhibited PMMA or UHMWPE-induced osteolysis andosteoclastogenesis.Conclusion:DOX can effectively inhibit osteoclastogenesisand affect mature osteoclast fate and suppress wear particles induced by osteoly-sis and osteoclastogenesis.DOX might be useful in the treatment or preventionof wear particles-induced osteolysis and aseptic loosening for its effect on osteo-clast generation and mature osteoclast fate and function.

Solvent exchange induced in situ forming gel(ISG) is the promising drug delivery system for periodontitis treatment owing to the prospect of maintaining an effective high drug level in the gingival crevicular fluid. In the present study, the influence of clove oil(CO) on the characteristics of doxycycline hyclate(DH)-loaded ISG comprising Eudragit RS(ERS) was investigated including viscosity/rheology, syringeability, in vitro gel formation/drug release, matrix formation/solvent diffusion and antimicrobial activities. CO could dissolve ERS and increase the viscosity of ISG and its hydrophobicity could also retard the diffusion of solvent and hinder the drug diffusion; thus, the minimization of burst effect and sustained drug release were achieved effectively. All the prepared ISGs comprising CO could expel through the 27-gauge needle for administration by injection and transform into matrix depot after exposure to the simulated gingival crevicular fluid. The antimicrobial activities against Staphylococcus aureus, Escherichia coli, Streptococcus mutans and Porphyromonas gingivalis were increased when the ratio of CO and N-methyl pyrrolidone(NMP) was decreased from 1:1 to 1:10 owing to higher diffusion of DH except that for C. albicans was increased as CO amount was higher.Therefore, CO could minimize the burst while prolonging the drug release of DH-loaded ERS ISG for use as a local drug delivery system for periodontitis treatment.

Regardless of the substantial progress in designing titanium-based dental implants and aseptic techniques,infection remains as the most common complication after implantation surgeries.Although,having a weakened immune system or systematic diseases is not seen as contraindicated for dental implants anymore,controlling the immune system is required to avoid surgical site infections after implantation.These patients have to control the surgical site infections by taking a high daily dose of oral antibiotics after dental implantation.The antibiotics oral administration has many side effects such as gastrointestinal symptoms,skin rashes and thrush.Coating antibiotics on the biomaterials surface could be a promising solution to reduce these disadvantages through locally releasing antibiotics in a controlled manner.The aim of this study was to investigate the effects of doxycycline coating layer on titanium-zirconium alloy surfaces in vitro and in vivo.In our previous studies,we demonstrated the chemical presence of doxycycline layer in vitro.In this study,we examined its physical presence using field emission scanning electron microscope and confocal microscope.We also analyzed its controlled released manner using Nano-Drop UV Vis spectrometer.After in vitro characterization of the coating layer,we evaluated its effects on the implant osseointegration in dogs and rabbits.The histological and histomorphometrical results exhibited no significant difference between doxycycline coated and uncoated groups regarding the implants osseointegration and biocompatibility for dental applications.Therefore,coating a doxycycline layer on TiZr implants could be favorable for reducing or removing the antibiotics oral administration after the implantation surgery.

AIM: To compare the effectiveness of topical cyclosporine A emulsion with that of oral doxycycline for rosacea associated ocular changes and dry eye complaints.METHODS: One hundred and ten patients with rosacea were screened. Thirty-eight patients having rosacea associated eyelid and ocular surface changes and dry eye complaints were included in the study. Patients were randomly divided into two groups: nineteen patients were given topical cyclosporine twice daily and nineteen patients were given oral doxycycline 100 mg twice daily for the first month and once daily for the following two months. Symptom and sign scores, ocular surface disease index questionnarie and tear function tests were evaluated at baseline and monthly for 3mo. Three months after results were compared with that of baseline.RESULTS: Mean values of symptom, eyelid sign and corneal/conjunctival sign scores of each treatment group at baseline and 3mo after treatments were compared and both drugs were found to be effective on rosacea associated ocular changes(P doxycycline treatment group(P doxycycline. In addition ocular rosacea as a chronic disease requires long term treatment and doxycycline has various side effects limiting its long term usage.

Oxide-polymer coatings were formed on the surface of the vanadium-free Ti–15Mo titanium alloy.The Ti alloy surface was modified by the plasma electrolytic oxidation process,and then,the polymer layer of a poly(D,Llactide-co-glycolide)with doxycycline was formed.The polymer evenly covered the porous oxide layer and filled some of the pores.However,the microstructure of the polymer surface was completely different from that of the PEO layer.The surface morphology,roughness and microstructure of the polymer layer were examined by scanning electron microscopy(SEM)and a confocal microscope.The results confirmed the effectiveness of polymer and doxycycline deposition in their stable chemical forms.The drug analysis was performed by highperformance liquid chromatography.The 1H NMR technique was used to monitor the course of hydrolytic degradation of PLGA.It was shown that the PLGA layer is hydrolysed within a few weeks,and the polyglycolidyl part of the copolymer is hydrolysed to glycolic acid as first and much faster than the polylactide one to lactic acid.This paper presents influence of different microstructures on the biological properties of modified titanium alloys.Cytocompatibility and bacterial adhesion tests were evaluated using osteoblast-like MG-63 cells and using the reference S.aureus and S.epidermidis strains.The results showed that the optimum concentration of doxycycline was found to inhibit the growth of the bacteria and that the layer is still cytocompatible.

BACKGROUND Multidrug-resistant Acinetobacter baumannii(MDRAB) has emerged as an increasingly important pathogen that causes nosocomial meningitis. However,MDRAB-associated nosocomial meningitis is rarely reported in children.CASE SUMMARY We report the case of a 1-year-old girl with a choroid plexus papilloma, who developed postoperative nosocomial meningitis due to MDRAB. The bacterial strain was sensitive only to tigecycline and colistin, and showed varying degrees of resistance to penicillin, amikacin, ceftriaxone, cefixime, cefotaxime,ciprofloxacin, levofloxacin, gentamicin, meropenem, imipenem, and tobramycin.She was cured with intravenous doxycycline and intraventricular gentamicin treatment.CONCLUSION doxycycline and gentamicin were shown to be effective and safe in the treatment of a pediatric case of MDRAB meningitis.

Determination of doxycycline by Europium Ion Fluorescence Probe Sensitized by Surfactant

The fluorescence property of europium(Ⅲ)-doxycycline(DC) complex and the effect of reaction condition were studied. It is found that in the buffer solution of Na_2B_4O_7-NaOH at pH 9.63 europium can react with doxycycline to form a complex. After intramolecular energy transfer, the system shows the characteristic fluorescence of Eu 3+. The fluorescence intensity can be enhanced again in addition of non-ionic surfactant Triton X-100 with λ_ ex=375 nm, λ_ em=617 nm. Based on this, a new method using europium ion as fluorescent probe for the determination of trace doxycycline was proposed. The fluorescence intensity is linearly related to the doxycycline concentration in the range of 2.8×10 -6～2.46×10 -5 mol·L -1 with correlation coefficient 0.9973. The detection limit was 1.35×10 -8 mol·L -1. It is applied to the determination of doxycycline in pharmaceutical preparations and in serum satisfactorily. The recovery is within the range of 93%～103%.
