doubled-HAPLOID TECHNOLOGY FACES A GREAT CHALLENGE FOR HYBRID BREEDING,Ensuring food security for the ever-growing population is a common mission and a great challenge for agricultural scientists worldwide.Historically,advances in crop breeding and management practices have contributed substantially to crop productivity.Indeed,the substantial increase in global grain yields over the last eight decades is largely due to the adoption of hybrids.However,the rate of increase of hybrid yields began to slow down in the early 2000s,and since then,it has reached a plateau for many crops and regions(https://***).Therefore,we must find solutions to accelerating genetic gain and boost hybrid development,for which developing new breeding technologies provides novel creative opportunities.

We study the electronic structure of double-twisted few-layer graphite(DTFLG),which consists of three few-layer graphites(FLGs),i.e.,an ABA-stacked graphene multilayer,stacked with two twist angles.We consider two categories of DTFLG,the alternately and chirally twisted cases,according to the rotation direction of the two twist angles.We show that,once the middle FLG of DTFLG is not thinner than the trilayer,both types of DTFLG can remarkably host two pairs of degenerate Moire flat bands(MFBs) at EF,twice that of the magic angle twisted bilayer graphene(TBG).The doubled MFBs of DTFLG lead to a doubled density of states(DOS) at the Fermi level EF,which implies much stronger correlation effects than TBG.The degeneracy of MFBs can be lifted by a perpendicular electric field,and the isolated MFBs have a nonzero valley Chern number.We also reveal the peculiar wave function patterns of the MFB s in DTFLG.Our results establish a new family of Moire systems that have a much higher DOS at EFand thus possibly much stronger correlation effects.

Dear Editor,Soybean(Glycine max[L.]Merr.)provides more than half of the oilseeds and more than a quarter of protein worldwide.It is estimated that the production of soybean has to be doubled by 2050 to meet the needs of the rapidly increasing consumption of soybean seeds along with a continuously increasing population(Ray et al.,2013).As such,development of a genotyping platform with high throughput,high efficiency and high precision but low-cost is urgently needed to accelerate soybean functional study and molecular design breeding.

The ability to generate(di)haploid plants has provided a strategy to significantly accelerate the crop-breeding process.The major advantage of doubled haploids to breeders lies in the simultaneous genetic fixation at every locus within a single generational step.Hence,each doubled haploid(DH)represents a pure line that can be further evaluated for the range of agronomically important traits.Haploids have been induced mainly through the generation of plants from cultivated gametophytic(haploid)cells,i.e.,in vitro haploid technologies,or through the selective loss of one parental chromosome set upon inter-or intraspecific hybridization,i.e.,in vivo haploid induction.However,in many crop species,efficient haploid technology is not yet available or only applicable to a limited number of genotypes(reviewed by Jacquier et al.,2020).Due to the limits and costs of current haploid technologies,plant breeders are highly interested in any methodological improvements.An efficient DH production system with low genotype dependency based on the in vivo approach exists so far only in maize.The haploid-inducing capacity of the inbred line Stock6,originally reported to cause haploid induction rates of 2.3%–3.2%(Coe,1959),was further increased to above 8%in various inducer breeding programs(reviewed by Jacquier et al.,2020).The underlying genetics of the in vivo haploid induction process is quite complex.However,in recent years significant advances have been made in elucidating the major players.

Dear Editor,Potato is a vital food security crop and is ranked as the world’s third most important food crop after rice and wheat.In 2011,the first genome assembly of a doubled monoploid potato DM1-3516 R44(DM)was released(Potato Genome Sequencing Consortium,2011),which has been widely used as one of the most popular reference genomes in the last decade and served as a valuable resource in plant genomics and potato genetics community(Leisner et al.,2018;Yang et al.,2020;Zheng et al.,2020).

Cardiovascular disease(CVD)has become the leading cause of death and disability in the global population and imposes a significant economic burden on global health systems.Prevalent cases of total CVD nearly doubled from 271 million in 1990 to 523 million in 2019,and the number of CVD deaths steadily increased from 12.1 million in 1990,reaching 18.6 million in 2019.The global trends for disabilityadjusted life years and years of life lost also increased significantly,and years lived with disability doubled from 17.7 million to 34.4 million over that period.

Crop breeding aims to generate pure in bred lines with multiple desired traits. doubled haploid (DH) and genome editing using CRISPR/Cas9 are two powerful game-changing technologies in crop breeding. However, both of them still fall short for rapid generation of pure elite lines with integrated favorable traits. Here, we report the development of a Haploid-Inducer Mediated Genome Editing (IMGE) approach, which utilizes a maize haploid inducer line carrying a CRISPR/Cas9 cassette targeting for a desired agronomic trait to pollinate an elite maize in bred line and to generate genome-edited haploids in the elite maize background. Homozygous pure DH lines with the desired trait improvement could be generated within two generations, thus bypassing the lengthy procedure of repeated crossing and backcrossing used in conventional breeding for integrating a desirable trait into elite commercial backgrounds.
