Comparison of Two Chemically-Induced Colitis-models in Adult Zebrafish, Using Optical Projection Tomography and Novel Transcriptional Markers

Crohn’s disease and ulcerative colitis—inflammatory bowel diseases (IBD)—are chronic conditions with an inadequately understood pathogenesis. Employing a set of novel molecular markers in a gene expression assay (qPCR), we have used adult zebrafish to investigate two acute inflammatory models, induced by the haptenizing agents oxazolone and TNBS. In addition, goblet cell dynamics in the scales and intestine and 5-HT (serotonin) in intestinal tissues were investigated through optical projection tomography. Gene expression studies revealed a distinct and significant upregulation of proinflammatory cytokines, acute-phase reactants and metalloprotease 9 in both chemical models, primarily after 72 hours. In comparison, transcription factors and cytokines associated with Th1 and Th17 (Crohn’s) and Th2 (ulcerative colitis) were mainly not affected in this acute setting. However, elevated transcript levels were detected in Foxp3, IL-10 and T-bet, which are linked with tolerance and Tregs in mammals. Goblet cells in scales were depleted in both chemical models and in the intestine of oxazolone-treated fish. A marked 5-HT signal was noted in intestinal tissue of some chemically treated zebrafish. In conclusion, a distinct acute inflammatory reaction was induced in both chemical models. Further, oxazolone and TNBS did not result in clear-cut Th2 and Th1/Th17 pathway signaling at this early timepoint, but the applied molecular tools may provide further insight to the IBD pathogenesis and translational value of the IBD zebrafish model.

Three-dimensional(3D)bioprinting has emerged as a promising approach for engineering functional tissues and organs by layer-by-layer precise positioning of biological materials,living cells,and biochemical *** with nonbiological printing,3D bioprinting involves additional complexities and technical challenges owing to the processing of living cells,such as the appropriate biomaterials that fulfill the requirements for both printability and *** this review,we first introduce the development course of 3D bioprinting,highlighting innovative forms of living building blocks and advances in enabling techniques of 3D *** then summarize the state-of-the-art advancements in 3D bioprinting for biomedical applications,including macroscale tissue or organ bioprinting,disease modeling,microphysiological systems,biobots,and bioprinting in *** the rapid development of 3D bioprinting over the past decades,most 3D bioprinted tissue or organ constructs are still far from being suitable for clinical translation,and it is necessary for the field of bioprinting to shift its focus from shape mimicking towards functionality ***,we provide our perspectives on this burgeoning field with an emphasis on functional maturation post printing and translational applications at the bedside.

Comparison of Different Modes of Molding in Canine Sepsis

Sepsis can cause a series of damages to various organs of the body,so it has always been regarded as a hot research topic in veterinary *** at the present situation of high morbidity and mortality of canine sepsis,in order to further explore the pathogenesis of this disease,it need to establish a stable and repeatable canine sepsis model that is in line with the clinical characteristics of the *** study selected 12 local dogs and randomly divided into three groups:rapid bolus injection group(Group A),continuous infusion group within 30 min(Group B)and continuous infusion group(Group C).Then,the lipopolysaccharides(LPS)with 2 mg·kg^-1 were injected through the brachial vein in different modes of administration,thus the model was fully *** the modeling period,body temperature(T),respiratory rate(RR),heart rate(HR)and mean arterial pressure(MAP)were monitored at 0,10,20,30,40,50 min and 1,2,3,4,5,6,7,8,9,12 and 24 *** was collected from the canine brachial vein at 0,1,2,3,4,5,6,7,8,9,12 and 24 h,respectively,for the detection of white blood cells(WBC).The test showed that the values of T,RR,HR,MAP,WBC of the dogs in group A all changed but did not exceed the normal range,and the clinical symptoms were not *** were no significant changes in the values of RR,HR,T,MAP and WBC of the dogs in Group B,and the clinical symptoms were not *** value of T of the dogs in Group C were significantly increased at 40 min(p<0.05),which reached the fever standard and lasted for 7 h;the value of RR increased significantly at 20 min(p<0.05),and a downward trend could be observed at 12 h,then it returned to normal at 24 h;the value of HR increased significantly at 50 min(p<0.05)and recovered at 8 h;the value of HR decreased significantly at 20 min(p<0.05),which remained at 12 h(p<0.05),and returned back to normal at 24 h;the value of WBC decreased significantly from 1 h to 4 h(p<0.05),which was lower than the normal value,and i

A NEW METHOD OF PRODUCING PORTAL HYPERTENSION IN DOGS

In order to rind a simple but satisfying method to make animal model with portal hypertension, a new method is designed by constricting half of the POrtal lumen and embolizing the portalvein chronically with silk thread. Twenty-two dogs were used in the experiment, among which onedied, and the others' portal venous pressure went up to (2. 83±0. 66) kpa 3～4 weeks arter the operation (1. 33±0.28) kpa before the operation (P < 0. 001 ). The results or gastrofiberscopic, portoveuographic and pathological examination showed that 17 dogs had esophagogastric varices with aformation rate or 80. 97 %. The 14 or which were achieved with a 100% formation rate as the methodwas improved. 12 dogs suffered portal hypertentive gastropathy (the rate of formation was57. 14% ). One dog was observed for 4 months arter the operation, still with portal hypertension. Allthis Indicates that the method is simple and time-saving, with high success rate, reliable quality andlong duration.

Neurons from the brains of Alzheimer’s disease（AD） and related tauopathy patients contain neurofibrillary tangles composed of hyperphosphorylated tau *** normally stabilizes microtubules（MTs）;however,tau hyperphosphorylation leads to loss of this function with consequent MT destabilization and neuronal ***,MT-stabilizing drugs such as paclitaxel and epothilone D have been shown as possible therapies for AD and related ***,MT-stabilizing drugs have common side effects such as neuropathy and *** find previously undescribed suppressors of tau-induced MT defects,we established a Drosophila model ectopically expressing human tau in muscle cells,which allow for clear visualization of the MT *** tau was hyperphosphorylated and resulted in decreased MT density and greater fragmentation,consistent with previous reports in AD patients and mouse *** a genetic screen,we found that a histone deacetylase 6（HDAC6） null mutation rescued tau-induced MT defects in bom muscles and *** and pharmacological inhibition of the tubulin speci-fic deacetylase activity of HDAC6 indicates that the rescue effect may be mediated by increased MT *** findings reveal HDAC6 as a unique potential drug target for AD and related tauopathies.
