In vitro study of nonthermal atmospheric pressure plasma in improving the durability of the dentin–adhesive interface with an etch-and-rinse system

In this study, we employed a nonthermal atmospheric pressure plasma(NTAPP) jet to evaluate the effect of plasma treatment on the durability of resin–dentin bonding under a thermocycling challenge. Furthermore, we assessed the degradation resistance of plasma-treated collagen under a sodium hypochlorite(NaClO) challenge. We assessed the beneficial effect of NTAPP treatment on the acid-etched dentin–bonding interface by testing the micro-tensile bond strength and examining the morphology. We found that the immediate bonding strength of the dentin significantly increased after NTAPP treatment. Compared with the control group, NTAPP resulted in a more prominent effect on the bonding durability of the dentin–adhesive interface after treatment for 5 or 10 s. Simultaneously, the mechanical strength of dentin collagen under the NaClO challenge was improved. Our results indicate that, in optimal conditions, NTAPP could be a promising method to protect dentin collagen and to improve the bonding durability between dentin and etch-and-rinse adhesives.

Determination of elastic and plastic mechanical properties of dentin based on experimental and numerical studies

The aim of this study is to investigate the change of mechanical properties of human dentin due to aging and spatial variation. Sections of coronal dentin are made from human molars in three groups： young, mid-aged, and old patients. A nanoindentation test is conducted from regions near the pulp to the dentin-enamel junction （DE J） to evaluate the load-depth indentation response and determine Young＇s modulus and hardness. Based on the loading and unloading load-displacement curves in nanoindentation, a numerical model of plastic damage is used to study the plastic and the damage behaviors and the contribution to the degradation in the unloading stiffness. The experimental results show that Young＇s modulus of the inner dentin is significantly lower than that of outer dentin in each age group. Compared with the young dentin, the old dentin has greater hardness and Young＇s modulus with similar spatial variations. The magnitudes of the yield strength and the damage variable are also affected by aging and vary with spatial locations. In the same age group, the yield strength in inner dentin is lower than those in middle and outer dentin, more damage occurs with similar spatial variations, and the yield strength of young dentin is generally lower and causes more damage compared with those in both the mid-aged and old groups.

Region dependent fracture resistance behavior of human dentin based on numerical simulation

dentin has a hierarchical structure and is composed of numerous tubules whose diameters and densities vary with the distances to the dentin-enamel junction. The unique structure determines the mechanical performance of dentin. In this study, a multiscale model, which is based on the combination of the virtual multidimensional internal bond(VMIB) theory and the Monte Carlo method, is used to simulate the fracture behavior of human dentin. Numerical simulations reveal that human dentin exhibits a graded resistance curve(R-curve). Among the three regions of dentin, superficial dentin shows the strongest resistance to crack propagation, and deep dentin has the weakest resistance. In addition, the predictions of fracture toughness of middle dentin agree well with the experimentally reported values, suggesting that the proposed model can be used to characterize the fracture behavior of human dentin comprehensively and properly.

To verify the theoretical models of varying transversely isotropic stress-strain relations of dentin established in the preceding work(Part Ⅰ),we per- form a set of *** of the very fine tooth size,it usually seems to be difficult to directly measure the inhomogeneous and anisotropic parameters of *** this paper,by the digital speckle correlation method,tensile experiments are made on the small dentin samples either parallel or perpendicular to the dentin *** the theoretically predicted elastic stress-strain relations,an optimiza- tion method is proposed to fit the strain curve adapted to the experimental data. The results show that the theoretical elastic stress-strain relations coincides very well with the experimental *** determined Young's modulus and Poisson's ratio of dentin matrix are 29.5GPa and 0.44,respectively,in the optimization sense.

Glycosylation of dentin matrix protein 1 is critical for fracture healing via promoting chondrogenesis

Fractures are frequently occurring diseases that endanger human health. Crucial to fracture healing is cartilage formation, which provides a bone-regeneration environment. Cartilage consists of both chondrocytes and extracellular matrix (ECM). The ECM of cartilage includes collagens and various types of proteoglycans (PGs), which play important roles in maintaining primary stability in fracture healing. The PG form of dentin matrix protein 1 (DMP1-PG) is involved in maintaining the health of articular cartilage and bone. Our previous data have shown that DMP1-PG is richly expressed in the cartilaginous calluses of fracture sites. However, the possible significant role of DMP1-PG in chondrogenesis and fracture healing is unknown. To further detect the potential role of DMP1-PG in fracture repair, we established a mouse fracture model by using a glycosylation site mutant DMP1 mouse (S89G-DMP1 mouse). Upon inspection, fewer cartilaginous calluses and down-regulated expression levels of chondrogenesis genes were observed in the fracture sites of S89G-DMP1 mice. Given the deficiency of DMP1-PG, the impaired IL-6/JAK/STAT signaling pathway was observed to affect the chondrogenesis of fracture healing. Overall, these results suggest that DMP1-PG is an indispensable proteoglycan in chondrogenesis during fracture healing.

Importance of aging to dehydration shrinkage of human dentin

There is an increase in the mineral content of human dentin with aging. Due to the consequent changes in the mineral to the collagen ratio, this process may influence the degree of hydrogen bonding that occurs with the loss of water and the extent of shrinkage as a result of dehydration. Thus, the objective of this investigation is to quantify the differences in the dehydration shrinkage of human dentin with patient age. Specimens of coronal dentin are prepared from the molars of young （23 ~〈 age 34） and old （52 ~ age ~〈 62） patients, and then maintained in storage solutions of water or hanks balanced salt solutions （HBSS）. Dimensional changes of the dentin specimens occurring over periods of free convection are evaluated by using the microscopic digital image correlation （DIC）. The results distinguish that the shrinkage of the young dentin is significantly larger than that of the old dentin, regardless of the orientation and period of the storage. The strains parallel to the tubules increase with proximity to the dentin enamel junction （DE J）, whereas the shrinkage largest in the deep dentin （i.e., near the pulp）. increases from the pulp to the DE J, and is the strains in the transverse direction axe the The degree of anisotropy in the shrinkage largest in the young dentin.

The Crystal Characteristics of Enamel and dentin by XRD Method

Crystal characteristics of tooth enamel and dentin were investigated using XRD, SEM, and EPMA methods. The results show that the mineral phase in enamel is HA and in dentin is HA and minor whitlockites. The dentin HA and the enamel HA have different crystallinity, the crystallinity of enamel HA is much higher than that of dentin HA. The average particle size of the enamel HA and dentin HA are 897 A and 309 A, respectively. The HA in enamel is regularly arranged, and in dentin the arrangement of HA is different from the enamel HA in the same section. Both the enamel and the dentin are mainly. consisted of Ca, P, O, and C, and the trace elements Mg, Sr, Al, Na, and K. The dentin contains more trace elements than the enamel. However, the incorporation of trace elements in both dentin and enamel are very limited. Other impurities such as F and Cl are less than their detection limit. The a and c values of enamel HA are 9.433 A and 6.896 A, and those of the dentin HA are 9.498 A and 6.896 A, respectively. The expansion in a value results from those the larger size of [ CO3 ]^2- group substituing for the smaller [ OH ] ^- group in the channel, and replacement of [ OH ]^ - by [ CO3 ]^2- dominates the change in cell parameter, taking into account of other trace elements.

Synthesis of Fluoridated Hydroxyapatite Containing Potassium and Its Effect on dentin

The aim of this work was to produce a nanoscale fluoridated and potassium containing hydroxyapatite via sol-gel method and to observe the effect of occlusion of the dentin tubule under scanning electron microscopy. HF was selected as fluorine-containing rengent which was added into mixed Ca （ NO3 ）2-P2O5 ethanol solutions to obtain dipping sols. All of the resulting samples showed apatite structure in their X- ray diffraction pattern. The lattice parameter a and the crystallite size were calculated to demonstrate the crystalline structure of fluoridated and potassium containing HA. Extracted paired premolars were chosen to obtain a fiat surface with opened tubules. It was observed that the tubules were covered by a 10 μm layer of fluoridated and potassium containing HA under the SEM. As the experimental results show the nanoscale fluoridated and potassium containing hydroxyapatite occludes the dentin tubules.
