OBJECTIVE: To establish a quantitative model for the online analysis of hesperidin content in the extraction process of Huatan Qushi decoction for individuals with phlegm-dampness constitution by using near-infrared(NIR) spectroscopy and partial least squares(PLS) ***: The reference content of hesperidin, an effective ingredient of Huatan Qushi decoction,was determined using high-performance liquid chromatography(HPLC). The modeling band was selected using synergy interval PLS(Si PLS) algorithms, and a PLS model demonstrating the relationship between NIR predictive values and HPLC reference values was established. The optimal modeling route was selected through global optimization of process trace ***: The root mean square error of cross-validation was 9.8410. The coefficients of determination of the calibration set and prediction set were0.9860(R2 cal) and 0.9458(R_(cal)~2), respectively. The residual predictive deviation was 3.0115. The parameters demonstrated high predictive capability and reliability. The band was selected via the Si PLS method. Using questionnaire is a common, efficient and cheap way to collect data in population-based epidemiological studies. However, there is a general trend of declining response rates leading to a higher risk for bias affecting the validity of results. Use of anonymous questionnaires and/or other ways of distributions, e.g. via schools instead of postal, may increase response rates, but can also lead to other types of systematic errors. This study indicates that results from cross-sectional questionnaire studies differ depending on distribution method and whether the survey is anonymous or not. Also the official purpose of the survey may affect the outcome. When studying well-known risk-factors and health effects, potential methodological problems like reporting bias must be considered in cross sectional studies.

Indoor Exposure to Mould and dampness in Infancy and Its Association to Persistent Atopic Dermatitis in School Age: Results from the Greek ISAAC II Study

Introduction: The presence of mould as a source of perennial allergens and bacteria products has been related to the appearance of respiratory symptoms in several studies. Yet, its role in eczema has not been elucidated. The aim of this study was to investigate the association between exposure to indoor visible molds/dampness and the manifestation of eczema in children. Methods: The study is part of the Greek contribution to ISAAC IΙ that includes 2023 students of randomly selected public primary schools in Athens and Thessaloniki, aged 9 - 10 years old. The children represented a general population sample and were evaluated according to ISAAC II questionnaire, validated for Greek language. Additionally, skin prick tests to aero-allergens were performed and children were examined for active skin lesions. Results: 13% had suffered from eczema in the past, 9% had current and 2% had atopic eczema (positive at least one skin prick test). Out of the children examined, half reported that eczema first appeared after the age of five years old whereas 70% mentioned persistence of eczema. dampness was reported in 10.8% and visible mould in 6.4% of all cases during infancy, while continued exposure until the age of 10 years old was reported in 38% and 33% out of them respectively. 10.8% of the sensitized children were positive to house dust mites and Alternaria, however, sensitization was not related to indoor exposure. In logistic regression analysis evaluating 20 environmental risk factors, a significant association was noted between the presence of indoor visible mold and dampness in infancy, and the presence of current eczema OR 1, 89 (95%CI 1.18 - 3.03). This association remained significant irrespective of the family history of eczema and sensitization. Conclusions: Frequently eczema first appears at early school age. Thirty rats were divided into three groups: Damp group ( relative humidity was over 90% andtemperature 18 ￣25 ) , Cold group ( T : 2 2 , RH : 54 4% ) and normal control group ( T : 18 ￣ 25 , RH :54 4% ) . The results showed that number of Escherichia coli increased greatly while number of bifidobac-terium reduced in high moisture. In Damp group, Ievel of secretory IgA was much higher as compared withthat of the normal controlled and Oold group. Th/i and Ts/c in Cold group and in Damp group were slighttylower than that in the normal group. Light microscopic examinations showed that there were intiltration ot in-flammatory colls and low-grade hyperemla in the phalangeal joints' synovium of rats of the Damp group inpathological section. Meanwhile there were low-grade erosion, degeneration, necrosis and exfoliation in ep-ithelium mucosa of small intestine.

OBJECTIVE: To investigate the expression of and interleukin-12(IL-12) and interferon-γ(IFN-γ) in relation to the pathology of damp-heat of spleen-stomach syndrome(DHSS) induced by Helicobacter pylori(H. pylori) ***: In total, 114 individual gastric mucosal specimens including 83 DHSS, 19 spleen-qi deficiency syndrome(SQD) and 12 from healthy volunteers(CON) were collected by gastroscopy. To explore the relationship between the two syndromes and H. pylori infection, individual samples were tested using rapid urease and methylene blue tests. Hematoxylin and eosin stained sections were examined to grade for the degree of inflammation and inflammatory activity, and expression of IL-12 and IFN-γ was investigated by ***: Statistically significant differences in the degree of inflammation and inflammatory activity were observed between the groups of specimens:DHSS, SQD and CON(P 0.05) groups. Intriguingly, in gastric specimens exhibiting IM and dysplasia, IL-12 translocated from the nucleus into the ***: Our findings suggest that IL-12 and IFN-γ are involved in DHSS pathology, but not in SQD, acting as healthy-Qi. DHSS is not just the consequence of those two cytokines but results from the cross-talk between a number of cytokines and/or other proteins, which may warrant further investigation in DHSS patients infected with H. pylori.

Objective: to explore the mechanism of transportation and transformation of dampness by the way of the expression of organic anion transporting polypeptide (oatp) superfamily member 2a1 (oatp2a1) mRNA in rat with spleen deficiency syndrome and the significance in transportation and transformation of dampness. Methods: 32 wistar male rats were divided randomly into four groups: normal group (n = 6), normal + AA group (n = 6), spleen deficiency group (n = 10), Spleen deficiency + AA group (n = 10). After reserpine-induced spleen deficiency model was made, intragastric administration of aristolochic acid (AA) was adopted for three days, the expression of oatp2a1 mRNA were detected in the tissues of lung, liver, kidney, stomach, small intestine and large intestine in four groups by using Fluorescent Quantitative-Polymerase Chain Reaction (FQ-PCR). Results: the expression of oatp2a1 mRNA in above six tissues could be detected. The ex-pression of oatp2a1 mRNA in liver tissue of rat with spleen deficiency syndrome was up-regulated compared to normal group (P = 0.035, P dampness. The changes of expression of oatp2a1 mRNA in small intestine, liver tissue suggests that small intestine, liver might play an important role in the transportation and transformation of dampness in the state of spleen deficiency. We further concluded that the function of spleen’s governing transportation and transformation of dampness was not only including the function of the gastrointestinal, but also part of the liver function in some degree, which needs to be further studied.
