Linc-RAM promotes muscle cell differentiation via regulating glycogen phosphorylase activity

Long non-coding RNAs(lncRNAs)are important regulators of diverse biological processes,especially skeletal muscle cell differentiation.Most of the lncRNAs identified to date are localized in the nucleus and play regulatory roles in gene expression.The cytoplasmic lncRNAs are less well understood.We previously identified a long intergenic non-coding RNA(linc-RNA)activator of myogenesis(Linc-RAM)that directly binds MyoD in the nucleus to enhance muscle cell differentiation.Here,we report that a substantial fraction of Linc-RAM is localized in the cytoplasm of muscle cells.To explore the molecular functions of cytoplasmic Linc-RAM,we sought to identify Linc-RAM-binding proteins.We report here that Linc-RAM physically interacts with glycogen phosphorylase(PYGM)in the cytoplasm.Knockdown of PYGM significantly attenuates the function of Linc-RAM in promoting muscle cell differentiation.Loss-of-function and gain-of function assays demonstrated that PYGM enhances muscle cell differentiation in an enzymatic activity-dependent manner.Finally,we show that the interaction between Linc-RAM and PYGM positively regulates the enzymatic activity of PYGM in muscle cells.Collectively,our findings unveil a molecular mechanism through which cytoplasmic Linc-RAM contributes to muscle cell differentiation by regulating PYGM activity.Our findings establish that there is crosstalk between lncRNAs and cellular metabolism during myogenic cell differentiation.

Effect of drought stress on male fertility restoration in A3 CMS-inducing cytoplasm of sorghum

Use of cytoplasmic male sterility (CMS) in hybrid breeding requires effective male fertility-restoring lines. In sorghum, very few restoring lines that can restore fertility in A_3 CMS have been reported. To identify the reasons for this deficiency, F_1 and F_2 hybrids of an A_3 CMS line crossed with the line IS1112C, a donor of fertility-restoring (Rf) genes for A_3 cytoplasm, and testcrosses of fertile plants to A_3CMS lines were grown under contrasting water availability regimes in dryland and irrigated field plots. In the irrigated plots the frequency of fertile plants in testcrosses was twice that in dryland plots (P cytoplasm.

An AFM Observation on Fossil cytoplasm

Fossil cytoplasm is a new research topic of interest in paleobotany. Atomic force microscope （AFM） is a new technology applied widely in physics and biology; however, it is rarely used in paleontology. Here we applied AFM for the first time to study fossil cytoplasm. The results indicate that the fossil cytoplasm is heterogeneous and full of ultrastructures, just like extant cytoplasm, and that the application of AFM, especially in combination with other techniques, can reveal the subcellular details of fossil plants with more confidence.

The male sterility(MS)line is a prerequisite for efficient production of hybrid seeds in rice,a self-pollinating species.MS line breeding is pivotal for hybrid rice improvement.Understanding the historical breeding tra-jectory will help to improve hybrid rice breeding strategies.Maternally inherited cytoplasm is an appro-priate tool for phylogenetic reconstruction and pedigree tracing in rice hybrids.In this study,we analyzed the cytoplasmic genomes of 1495 elite hybrid rice varieties and identified five major types of cytoplasm,which correspond to different hybrid production systems.As the cytoplasm donors for hybrids,461 MS lines were also divided into five major types based on cytoplasmic and nuclear genomic architecture.Spe-cific core accessions cooperating with different fertility-associated genes drove the sequence divergence of MS lines.Dozens to hundreds of convergent and divergent selective sweeps spanning several agro-nomic trait-associated genes were identified among different types of MS lines.We further analyzed the cross patterns between different types of MS lines and their corresponding restorers.This study system-atically analyzed the cytoplasmic genomes of rice hybrids revealed their relationships with nuclear ge-nomes of MS lines,and illustrated the trajectory of hybrid rice breeding and the strategies for breeding different types of MS lines providing new insights for future improvement of hybrid rice.

Since Wilson and Ross （1962） bred wheat male sterile line with T. timopheevi cytoplasm （G type） and its relative restoration line, with high utilization value, the progress has been accelerated in research of heterotic use of wheat male sterility. But so far they have not been used in large area production, because of the limited restorer resource, imperfect grains and insignificant population heterosis. Ae（?）ilops kotschyi and Ae. Variabilis types/（Sv type） overcame well some problems in G type, such as restorer resource, grain plumpness etc., but at the same time, other more difficult problems arose, e. g. their sterile line and hybrid F1 generation produced monoploids at a certain rate, hybrid growth was weak and restorer degree of sterility had a large variation scale. Identification of Target Ligands of CORYNE in Arabidopsis by Phage Display Library

CORYNE（CRN） plays important roles in stem cell division and differentiation of shoot apical meristem （SAM） in Arabidopsis thaliana.The cytoplasmic kinase domain of CRN has been cloned and expressed in Escherichia coli,and further purified by two consecutive steps of affinity chromatography.By using this purified CRN as a ligand,a 12-mer random-peptide library was used to determine the specific amino acid sequences binding with the recombinant CRN molecule.After four rounds of biopanning,positive phage clones were isolated and sequenced,and further tested by enzyme linked immunosorbent assay for their binding ability and specificity.Two positive clones that specifically bind to the intracellular protein kinase domain of CRN have been identified.Alignment of these peptides and the kinase-associated protein phosphatase（KAPP） shows high similarity,indicating that KAPP might interact with the cytoplasmic kinase domain of CRN and negatively regulate the CLV signal.Our current study would be helpful to better understand the CLV3 signal pathway.

Expression of Phospho-MeCP2s in the Developing Rat Brain and Function of Postnatal MeCP2 in Cerebellar Neural Cell Development

Abnormal expression and dysfunction of methyl-CpG binding protein 2 （MeCP2） cause Rett syndrome （RTT）. The diverse phosphorylation modifications modulate MeCP2 function in neural cells. Using western blot and immunohistochemistry, we examined the expression patterns of MeCP2 and three phospho-MeCP2s （pMeCP2s） in the developing rat brain. The expression of MeCP2 and phospho-S80 （pS80） MeCP2 increased while pS421 MeCP2 and pS292 MeCP2 decreased with brain maturation. In contrast to the nuclear localization of MeCP2 and pS80 MeCP2, pS421 MeCP2 and pS292 MeCP2 were mainly expressed in the cytoplasmic com- partment. Apart from their distribution in neurons, they were also detected at a low level in astrocytes. Postnatallyinitiated MeCP2 deficiency affected cerebellar neural cell development, as determined by the abnormal expression of GFAP, DCX, Tuj 1, MAP-2, and calbindin-D28k. Together, these results demonstrate that MeCP2 and diverse pMeCP2s have distinct features of spatio-temporal expression in the rat brain, and that the precise levels of MeCP2 in the postnatal period are vital to cerebellar neural cell development.

Model acupuncture point:Bone marrow-derived stromal stem cells are moved by a weak electromagnetic field

AIM To show the existence of a structural formative role of magnetic fields(MFs) with respect to biological objects by using our proposed model of an acupoint.METHODS We introduced a magnetised 10-100 μT metal rod(needle) into culture dishes with a negatively charged working surface and observed during 24 h how cells were arranged by MFs and by electrical fields(EFs) when attached. Rat and human bone marrow-derived stromal stem cells(r BMSCs and h BMSCs), human nonadherent mononuclear blood cells, NCTCs and A172 cells, and Escherichia coli(E. coli) were evaluated. The dish containing BMSCs was defined as the model of an acupoint. r BMSCs proliferative activity affected by the needle was investigated. For investigating electromagnetic field structures, we used the gas discharge visualisation(GDV) method.RESULTS During 24 h of incubation in 50-mm culture dishes, BMSCs or the nonadherent cells accumulated into a central heap in each dish. BMSCs formed a torus(central ring) with an inner diameter of approximately10 mm only upon the introduction of the needle in the centre of the dish. The cells did not show these effects in 35- or 90-mm culture dishes or hydrophobic dishes or rectangular cuvettes. NCTCs and A172 cells showed unstable the effects and only up to two weeks after thawing. Moreover, we observed that the appearance of these effects depended on the season. In winter, BMSCs showed no the effects. GDV experiments revealed that the resonant annular illumination gradually formed from 10 to 18-20 s in polar solutions with and without cell suspension of BMSCs, NCTCs and E. coli when using circular 50-mm dishes, stimulation at 115 V and switching of the electrode poles at 1 kH z. All these data demonstrate the resonant nature of the central ring. Significant influence of MFs on the rB MSC proliferation rate was not observed.CONCLUSION BMSCs can be moved by MFs when in the presence of a constant EF and MF, when the cells are in the responsive functional state, and when there is a resonant relationship between them.

cytoplasm Has No Effect on the Yield and Quality of Biomass Sorghum Hybrids

The development of the biomass sorghum hybrid seed industry is contingent on the use of cytoplasmic male sterility. Within sorghum, there are several different cytoplasmic male sterility systems and it is important to determine early in development if cytoplasm will affect agronomic performance or composition characters. Thus, if there is a difference, then the best system can be deployed. The purpose of this study was to determine if cytoplasm per se influences agronomic performance of biomass sorghum using a set of iso-cytoplasmic hybrids. Three hybrid genotypes were produced in three different cytoplasms (A1, A2, and A3) for a total of nine hybrids. These hybrids were evaluated for plant height, biomass yield, and biomass composition in three Texas environments (Weslaco, College Station, and Halfway) in 2010. Across environments, significant differences existed among hybrids for both agronomic and compositional traits, but cytoplasm per se had no effect on any measured trait. Since cytoplasm did not effect on hybrid performance, any of the tested cytoplasms (A1, A2, and A3) can be deployed in hybrid biomass sorghums.
