Toxic cyanobacterial blooms constitute a threat to human safety because Microcystis sp. releases microcystins during growth, and particularly during cell death. Therefore, analysis of toxic and nontoxic Microcystis in natural communities is required in order to assess and predict bloom dynamics and toxin production by these organisms. In this study, an analysis combining fluorescence in situ hybridization (FISH) with flow cytometry (FCM) was used to discriminate between toxic and nontoxic Microcystis and also to quantify the percentage of toxic Microcystis present in blooms. The results demonstrate that the combination of FISH and flow cytometry is a useful approach for studying the ecology of Microcystis toxin production and for providing an early warning for toxic Microcystis blooms.

Science flow cytometry measurement of intensity is relative measur, ement. The standards are therefore necessary. There are two types of standards. One is the instrumental standard for adjustment FCM. Fluorescent microspheres or glutaraldehyde fixed chicken red blood cells are commonly used for this purpose. The other is the DNA internal standard of nuclear DNA ploidy analysis for correcting the changes in preparation and staining condition and instrument drift during measurement.

Objective:To investigate the role of water-soluble extract of Salvia fruticosa(Creek sage)(***) leaves in reducing both intrinsic cellular and H_2O_2-induced DNA oxidation in cultured human embryonic kidney 293 *** to the Eastern-Mediterranean basin,is widely used as a medicinal herb for treatment of various ***:Dried leaves of *** were extracted in phosphate buffer saline and purified using both vacuum and high pressure *** mL of the preparation contained(7.1±1.0)mg of ***-293 cells were incubated in one set with *** extract in the presence of 0.1 mmol/L H_2O_2,and in the other set with the addition of the extract *** DNA oxidation was measured using fluorescence upon fluorescein isothiocyanate derivarization of 8-oxoguanine *** fluorescence was measured using flow cytometry ***:Cells incubated 3 h with 150μL extract and exposed to 0.1 mmol/L H_2O_2 showed lower intensity of fluorescence,and thus lower DNA ***,cells incubated 3 h with 100μ*** the extract showed lower intensity of fluorescence,and thus lower intrinsic cellular DNA oxidation compared to control(without ***).Conchisions:The results from this study suggest that the water-soluble extract of *** leaves protects against both H_2O_2-induced and intrinsic cellular DNA oxidation in human embryonic kidney 293 cells.

Distribution of one group of marine viral particles along the Kuroshio Current and theadjacent area was investigated in June, 1998 using flow cytometry for the first time. Cytomics是一个特殊名词,她将cyto-与-omics连在一起,代表着一个崭新的领域。Cyto-主要来自分析细胞学(analyticalcytology或cytometry),而-omics主要来自蛋白质组学(proteomics)及其决定者基因组学(genomics)。经过三十余年的发展,分析细胞学可以通过分子探针将异质性的或混杂的细胞群体分为不同的细胞组份(cytomes),而基因组学和蛋白质组学的技术已能鉴定出某一物种的整套基因组和蛋白质组。因此,我们认为,细胞组学的任务是分离细胞组,并在基因组和蛋白质组层面确定其赖以存在和转变的分子基础。细胞组学的实施需要两大系列技术,一是细胞组分离技术,二是分离后再分析技术。这些技术的发展将使我们最终理解基因组并利用之,也必将拓展出更新的领域。

Spectral Compensation for Linear-Logarithmic Flow cytometry Acquisitions

Compensating for fluorescence overlap in multiparameter flow cytometry datasets, of which one parameter is linear distributed and at least one parameter is logarithmic distributed, leads usually to extreme high compensation values. We investigated this phenomenon with an adapted flow cytometry model, of which the two parameters can easily be converted from linear to logarithmic and vice versa. With the adapted model, spectral compensation was performed both for linear-logarithmic and linear-linear parameter distribution. The results of the flow cytometry model were validated with a real world example which was also compensated twice. The results of the two experiments show that the compensation values equal to the theoretically expected value when both parameters are linear distributed. However, the compensation value exceeds 100% when one of the two parameters is logarithmic distributed. In addition, we found that spectral compensation of differently distributed parameters leads to deformation of the compensated events. With the adapted flow cytometry model presented in this paper it is shown how to correctly compensate flow cytometry acquisitions with different distributed parameters.

We have previously demonstrated that allogeneic human bone marrow (BM) supports human islet function and longevity in vitro. We hypothesize that BM supporting human islets may include to increase β-cell in cultured islets. In this study, we developed a method to quantify insulin-producing β cells from cultured islets by using immunofluorescent staining and flow cytometry analysis to explore this possibility. The results show that human islets cocultured with BM for 39 days contained a significantly higher number of insulin-positive β cells (42.3% ± 4.5%) compared to the islet-only cultures (1.15% ± 0.78%), and increased insulin release levels evaluated by ELISA is consistent with increased β cells in same culture condition. Human islet culture with BM significantly increase β-cells while islet only culture lost β-cells in same culture period supports the possibility of BM increasing β-cells in cultured islets.

Background: Mature red blood cells lack protein synthesis and are unable to restore inactivated enzymes, damaged cytoskeleton and membrane proteins. An oxidation breakdown of band 3 is probably part of the mechanism leading to the generation of a senescent cell antigen. This specific signal serves for the clearance of RBCs by inducing the binding of autologous IgG and C3, leading to phagocytosis. In addition, phosphatidilserin molecules appear in the outer membrane and the CD47 expression diminishes. Methods: Erythrocytes of different ages from whole blood were studied by flow cytometry analysing light scatter proprieties, binding of autologous IgG, C3 complement deposits, externalization of phosphatidylserine and CD47 expression. Dot-plot analysis based on forward scatter versus side scatter parameters showed two RBCs populations of different sizes and density. RBCs were further incubated with Alexa 488 IgG, APC-anti-C3, PE-annexin-V and PE-CD47. The comparison of the values obtained for the different variables studied in SeRBC and YRBC populations was carried out by the Student t-test for matched samples or by the Wilcoxon test (after verification of the normality assumption). Results: The percentage of IgG and C3 positive cells was significantly higher in senescent red blood cells population. The fraction of annexin-V positive RBCs was also larger in SeRBCs while the CD47 expression was lower in this population. Conclusions: These results indicate that flow cytometry allow differenciation of erythrocytes populations of different ages, turning this tool into an useful alternative option to study erythrocyte aging process. These findings will contribute to a better understanding of the process and mechanisms involved in erythrocyte senescence process.

Flow cytometric (FCM) and microspectrophotometric (MSP) measurements of cell nuclear DNA content were made in 53 fresh gastric carcinoma specimens and in 30 gastric mucosal specimens with chronic gastritis. DNA aneuploidy was found in 32/53 (60%) of gastric carcinomas, and appeared more frequently in wellor moderately differentiated tubular adenocarcinomas (90%) than in undifferentiated and mucousa cell carcinomas (23.6%) (P<0.001). No aneuploidy was found in chronic gastritis samples, but their proliferative cell fractions were higher than in normal control gastric mucosa samples (P<0.01). A comparison was made between FCM and MSP analyses of DNA content in 29 cases of gastric carcinoma, a high correlation rate (r=0.90) was found. The advantages and limitations of both methods are discussed, they may be used in combination for more precise cytochemical analysis.

Flow cytometry is a versatile technique to study different aspects of the cell cycle from subpopulations of cells to detailed cell kinetic information. In this paper a basic review of cell kinetic parameters is presented followed by detailed descriptions of the different flow cytometric methodologies that can be used to extract pertinent information for a particular study. The methodologies range from simple DNA profile analysis, the use of bromodeoxyuridine to cell cycle-associated proteins such as the cyclins.
