Human cyclophilin 33 (hCyP33), found in 1996, consists of an RNA-binding domain in N-terminus, a cyclophilin domain in C-terminus and a connected part between the two domains. RNA-binding proteins concern functions, such as splicing, modification and transport, after transcription in eukaryotic cells. cyclophilins (CyPs) possess enzymatic activity, namely peptidyl-proryl cis-trans isomerase (PPIase). They are involved in folding, transport and interaction of proteins. Cyclosporin A (CsA), an immunosuppressant used by organ transplantation, binds to CyPs and suppresses their enzymatic activity. However, up to now it is unknown that which cellular and physiological roles hCyP33, which possesses the above-mentioned both functions, plays. In this paper the binding specificity of hCyP33 to different cellular RNA is investigated by means of ion-exchange chromatography and affinity adsorption. The results show that it binds specifically to poly(A) tailed mRNA, namely poly(A)+RNA.

Aim: To investigate methods for identifying specific cyclophilin D （CypD） inhibitors derived from quinoxaline, thus developing possible lead compounds toinhibit mitochondrial permeability transition （MPT） pore opening. Methods: Kinetic analysis of the CypD/inhibitor interaction was quantitatively performed byusing surface plasmon resonance （SPR） and fluorescence titration （FT） ***50values of these inhibitors were determined by PPIase inhibition activity ***: All the equilibrium dissociation constants （KD） of the seven compoundsbinding to CypD were below 10 μmol/L. The IC50values were all consistent withthe SPR and FT results. Compounds GW2, 5, 6, and 7 had high inhibition activitiesagainst Ca2+-dependent rat liver mitochondrial swelling and Ca2+uptake/*** GW5 had binding selectivity for CypD over CypA. Conclusion: Theagreement between the measured IC50values and the results of SPR and FT suggests that these methods are appropriate and powerful methods for fyingCypD inhibitors. The compounds we screened using these methods （GW1-7） arereasonable CypD inhibitors. Its potent ability to inhibit mitochondrial swelling and the binding selectivity of GW5 indicates that GW5 could potentially be used for inhibiting MPT pore opening.

本研究克隆和表达了日本血吸虫cyclophilin B(Sj CyPB)编码基因的cDNA,分析其在日本血吸虫不同发育阶段虫体的表达情况,评估该重组抗原在小鼠体内诱导的抗血吸虫免疫保护效果。本研究以日本血吸虫童虫cDNA为模板,RT-PCR扩增其基因全长cDNA,提交序列到NCBI,登录号为GQ403665。荧光实时定量PCR分析该基因在日本血吸虫不同发育阶段虫体的表达情况,构建重组表达质粒,表达纯化重组蛋白。利用Western blotting检测重组蛋白的抗原性。以重组抗原免疫小鼠,评估其对小鼠诱导的免疫保护效果。结果表明,RT-PCR获得了Sj CyPB编码基因的全长cDNA,其开放阅读框为672bp。经分析确定其为CyPs家族中的CyPB基因,命名为Sj CyPB。荧光实时定量PCR分析表明,该基因在18d童虫期表达量最高,32d次之。构建了重组表达质粒pGEX-6P-1-SjCyPB,并在大肠杆菌中成功表达,表达产物分子量为49.5kDa。Western blotting试验显示该重组蛋白具有良好的抗原性,在小鼠免疫试验中,与空白对照组比较,免疫组小鼠获得31.5%的减虫率和41.01%的肝脏减卵率。本研究获得了日本血吸虫童虫期高表达的Sj CyPB基因的全长cDNA,成功构建了Sj CyPB原核重组表达质粒,并在大肠杆菌中成功表达,证实该重组抗原在小鼠体内诱导产生了部分免疫保护效果。

cyclophilin A(简称CypA)由PPIA(Peptidylprolyl isomerase A)基因编码,是典型的cyclophilin家族蛋白,具有肽基脯氨酰顺反异构酶活性,在蛋白质的折叠和转运、信号转导、炎症、免疫调节、细胞凋亡及病毒复制等生物学过程中发挥着重要作用。本研究重点探讨了不同脊椎动物CypA的肽基脯氨酰顺反异构酶活性及其遗传变异。根据Gen Bank数据库PPIA基因的序列信息,克隆了脊椎动物的PPIA基因并构建其原核表达载体,其中鼠耳蝠(Myotis davidi)和绿头鸭(Anas platyrhynchos)的PPIA基因序列为首次报道。利用大肠杆菌表达GST-Cyp A融合蛋白并进行亲和层析,切除GST标签后再进行分子筛层析,获得纯化的CypA蛋白。利用胰糜蛋白酶偶联法测定CypA的肽基脯氨酰顺反异构酶活性,发现12种脊椎动物CypA的肽基脯氨酰顺反异构酶活性无显著差异。同时,通过一系列遗传变异和分子进化分析,发现12种脊椎动物CypA的酶活位点、CsA结合位点等重要功能域的氨基酸序列完全一致,而且其结构和在染色体中的基因定位也非常保守。研究结果表明,脊椎动物CypA的关键功能域高度保守,这是CypA维持其酶活及生物学功能的有力保障。

目的检测氧化应激下人视网膜色素上皮(retinal pigment epithelium,RPE)细胞内cyclophilin D的表达,初步探索RPE细胞氧化损伤保护新靶点。方法培养细胞系ARPE19及人原代RPE细胞,相差显微镜下观察细胞形态,应用激光共聚焦显微镜免疫荧光法鉴定细胞。不同浓度H_2O_2(0μmol·L^(-1)、100μmol·L^(-1)、500μmol·L^(-1)、1 mmol·L^(-1))处理细胞24 h后,应用RT-PCR检测cyclophilin D在细胞内的表达。随后预先在部分细胞中加入3μmol·L^(-1)环孢素A(cyclosporin A,Cs A)处理30 min后再加入不同浓度H_2O_2(80μmol·L^(-1)、160μmol·L^(-1)、320μmo·L^(-1))2 h,即Cs A+H_2O_2组,应用乳酸脱氢酶(lactic dehydrogenase,LDH)法检测细胞致死率,并与H_2O_2组的LDH释放量相比较。结果培养的ARPE19和人原代RPE细胞均表达RPE细胞特异性抗体RPE65。100μmol·L^(-1)H_2O_2处理细胞24 h后cyclophilin D表达量明显升高,当H_2O_2浓度增加到500μmol·L^(-1)时,cyclophilin D表达量降低,1 mmol·L^(-1)H_2O_2处理时,cyclophilin D表达水平未见增加。在各H_2O_2组组间,LDH的释放量随着H_2O_2浓度的升高而增加;与H_2O_2组相比,Cs A+H_2O_2组LDH的释放水平明显降低(Pcyclophilin D的表达升高,该蛋白表达的增加可能进一步导致氧化应激下细胞的死亡,Cs A可能成为RPE细胞保护的新策略。

目的探讨人脐静脉内皮细胞凋亡模型中cyclophilin—D蛋白的表达及意义。方法分别应用100、300、500μmol／LH2O2按时间依从处理人脐静脉ECV304细胞，对照组单纯磷酸盐缓冲液（PBS）处理。检测细胞凋亡率、cyclophilin—D蛋白及其编码基因ppif的表达量的变化。结果500μmol／LH2O2处理组的ECV304细胞凋亡率、cyclophilin—D蛋白表达量和cyclophilin—D编码基因ppif的表达量均高于对照组（P〈0．01）。cyclophilin—D表达量与H2O2作用时间和凋亡率呈显著正相关，r分别为0．967（P〈0．01）和0．971（P〈0．01）。结论H2O2可成功诱导血管内皮细胞氧化损伤与凋亡，cyclophilin—D蛋白可作为细胞凋亡严重程度的预测因子。

Aim: The objects of this study originated from the experimental observations, whereby the HIV -1 gp120 V3 loop is a high-affinity ligand for immunophilins, and consisted in generating the structural complex of cyclophilin (Cyc) B belonging to immunophilins family with the virus subtype A V3 loop (SA-V3 loop) as well as in specifying the Cyc B segment forming the binding site for V3 synthetic copy of which, on the assumption of keeping the 3D peptide structure in the free state, may present a forwardlooking basic structure for anti-AIDS drug development. Methods: To reach the objects of view, molecular docking of the HIV-1 SA-V3 loop structure determined previously with the X-ray conformation of Cyc B was put into practice by Hex 4.5 program (http://***/~ritchied/ hex/) and the immunophilin stretch responsible for binding to V3 (Cyc B peptide) was identified followed by examination of its 3D structure and dynamic behavior in the unbound status. To design the Cyc B peptide, the X-ray conformation for the identical site of the native protein was involved in the calculations as a starting model to find its best energy structural variant. The search for this most preferable structure was carried out by consecutive use of the molecular mechanics and simulated annealing methods. The molecular dynamics computations were implemented for the Cyc B peptide by the GROMACS computer package (http:// ***/). Results: The overmolecular structure of Cyc B with V3 was built by computer modeling tools and the immunophilinderived peptide able to mask effectively the structurally invariant V3 segments embracing the functionally crucial amino acids of the HIV-1 gp120 envelope protein was constructed and analyzed. Conclusions: Starting from the joint analysis of the results derived with those of the literature, the generated peptide was suggested to offer a promising basic structure for making a reality of the protein engineering projects aimed at developing the anti-A

Objective:To study the correlation of serum cyclophilin A (CyPA) and monocyte chemoattractant protein-1 (MCP-1) levels with carotid atherosclerosis in patients with acute cerebral ***: 106 patients with acute cerebral infarction who were hospitalized in our hospital between July 2011 and August 2015 were selected as observation group, and 50 cases of healthy persons who received physical examination in our hospital during the same period were selected as normal control group. The serum CyPA and MCP-1 contnets in two groups were determined. According to the median of CyPA and MCP-1 contents in observation group, they were divided into high CyPA group and low CyPA group as well as high MCP-1 group and low MCP-1 group, 53 cases in each group. Contents of lipid metabolism indexes and carotid atherosclerosis illness-related indicators were compared between acute cerebral infarction patients with different CyPA and MCP-1 ***:Serum CyPA and MCP-1 contents in observation group were significantly higher than those in control group. Serum TC, LP(a) and LDL-C contents in high CyPA group and high MCP-1 group were higher than those in low CyPA group and low MCP-1 group while HDL-C contents were lower than those in low CyPA group and low MCP-1 group. Serum CysC, Hcy and UA contents in high CyPA group and high MCP-1 group were higher than those in low CyPA group and low MCP-1 ***: Serum CyPA and MCP-1 contents in patients with acute cerebral infarction are higher than those in normal population, and the contents of CyPA and MCP-1 are positively correlated with the degree of carotid atherosclerosis.
