Objective:To construct recombinant survivin and CDK1 tandem shRNAs pU6-M4 ***:sequences carrying nucleotides of shRNA (targeting survivin or CDK1) and terminator codon and endonuclease cutting sites positioning at two sites were designed and *** plasmids of pU6-M4-Survivin-shRNA and pU6-M4-CDKI-shRNA were constructed and identified,*** constructed plasmids were digested with restriction endonucleases;digested 4.9kb large fragments of pU6-M4-Survivin-shRNA were connected with 400bp small fragments of CDK1-shRNA by T4 DNA *** ligated products were transformed into competent TH5a cells,and then the recombinant clones were identified by enzymes digestion and ***: sequencing proved that pU6-M4-Survivin-shRNA and pU6-M4-CDK1-shRNA were successfully *** cleave identification and sequencing proved that pU6-M4-Survivin-shRNA and pU6-M4-CDKI-shRNA were correctly inserted into pU6-M4 ***:recombinant survivin and CDK1 tandem shRNAs pU6-M4 plasmids were correctly constructed.
