No DCX-positive neurogenesis in the cerebral cortex of the adult primate

Whether endogenous neurogenesis occurs in the adult cortex remains *** increasing number of reports suggest that doublecortin(DCX)-positive neurogenesis persists in the adult primate cortex,attracting enormous attention *** this study,different DCX antibodies were used together with NeuN antibodies in immunohistochemistry and western blot assays using adjacent cortical sections from adult *** adsorption,antigen binding,primary antibody omission and antibody-free experiments were used to assess specificity of the *** found either strong fluorescent signals,medium-weak intensity signals in some cells,weak signals in a few perikarya or near complete lack of labeling in adjacent cortical sections incubated with the various DCX *** putative DCX-positive cells in the cortex were also positive for NeuN,a specific marker of mature ***,further experiments showed that most of these signals were either the result of antibody cross reactivity,the non-specificity of secondary antibodies or tissue *** confirmed DCX-positive cells were detected in the adult macaque cortex by *** findings show that DCX-positive neurogenesis does not occur in the cerebral cortex of adult primates,and that false-positive signals(artefacts)are caused by antibody cross reactivity and *** experimental protocols were approved by the Institutional Animal Care and Use Committee of the Institute of Neuroscience,Beijing,China(approval ***-AMMS-2014-501).

Inducement of Specific CTLs by Antigen-Peptides from Human Leukemia Cells and Their Cytotoxicity to Leukemia Cells

To investigate the inducement of cytotoxic T lym phocytes(CTL s) by antigen peptides m ixture from different leukemia cells and the cross- reaction of the m ixtures from different cell lines,antigen peptides m ixtures were prepared from different leukemia cell lines respectively and then bound with Hsp70 in *** and proliferation of PBMC were observed after stim u- lation with different Hsp70 - peptide *** ratio of CD8+ in proliferative cells was ana- lyzed by flow *** cytotoxicity of the activated PBMC to different target cells was as- *** results showed that the antigen peptides from different leukemia cell lines,bound with Hsp70 ,could activate PBMC effectively,and stimulate the activated PBMC to *** proliferative PBMC had specific cytotoxicity to corresponding leukem ia ***8+ cells,account- ing for a high proportion in proliferative cells,had a specific cytotoxicity to leukemia cells from which antigen peptides were prepared,suggesting that these CD8+ cells were CTL s specific to leukemia *** s activated by Hut78- peptides or Molt4 - peptides had a significantly stronger cytotoxicity to Hut78cells,Molt4 cells and Jurkat cells than that of CTL s activated by HL - 6 0 - peptides(Pcross- reactivity among antigen peptides m ixtures from different cell lines of the sam e type leukemia and more cross- reactive antigen peptides could be obtained from m ore cell lines,suggesting that antigen peptides m ixture with broad antigenic spectrum could be prepared by using multiple leukemia cell lines.

A Nonglycosylated 27 KDa Molecule as Common Antigen between Human Breast Cancer and Echinococcus granulosus Hydatid Cyst Wall

Background: Hydatid cyst, which has anti-cancer activities, is outwardly covered with the cyst wall. It is in close contact with the host tissues and its molecules presented to the immune system. In this work immunological reaction of the sera of breast cancer patients with the hydatid cyst wall antigens has been investigated. Method: For this purpose, sera of patients with breast cancer, hydatid cyst and normal human sera were collected and their reaction with hydatid cyst wall antigens was tested using western immunoblotting technique. Results: All sera of patients with breast cancer, hydatid cyst and also human normal sera reacted with a band in western immunoblotting. However, sera of patients with breast cancer showed reaction with a 27 KDa band. Results of this work also revealed that this band was not glycosylated and may express only in some stages of breast cancer development. Conclusion: Sera of patients with breast cancer cross reacted with a nonglycosylated antigen of hydatid cyst wall. However, more work is recommended to find if this band involves in anticancer activity of the hydatid cyst.

The aberrant carbohydrate structure expressing on the surface of cancer cells known as tumor-associated carbohydrate antigens（TACAs）, provide a target for cancer vaccines. Due to the low immunogenicity nature of carbohydrate, typical vaccines often contain a protein or peptide component to effectively improve its immunogenicity but cause problems in synthesis and in vivo application. Gold nanoparticles（GNPs） are promising alternatives to protein or peptide carriers for their controllable synthesis and special immunological properties. Herein we synthesized a series of fluorine substituted carbohydrate mimics with improved immunogenicity and bioavailability by assembling them to GNPs via different linkers. These T-antigens might produce antisera cross reaction as reported fluorine substituted analogues. Further antisera assays are undergoing to identify some modified GNPs as potential cancer vaccines.
