In vivo antianxiety and antidepressant activity of Murraya paniculata leaf extracts

OBJECTIVE： Different parts of Murraya paniculata have been used traditionally for treating several ailments including mental disorders. The present study was designed to evaluate the antianxiety and antidepressant potential of M. paniculata leaves using elevated plus maze model and forced swim test, respectively. METHODS： Extracts of M. panicu/ata made with petroleum ether （60-80 ℃）, chloroform, ethanol and water were evaluated for antianxiety and antidepressant activity. The anxiolytic chloroform extract was subjected to column chromatography, yielding five fractions （F1-F5）. Fraction F5 （100 mg/kg）, which showed notable anxiolytic activity, was further chromatographed to get four subfractions （F5.1-F5.4）. Simultaneously, the ethano extract was partitioned with ethyl acetate to obtain ethyl acetate soluble fraction （EASF） and ethyl acetate insoluble fraction. Phytochemical screening of bioactive extracts/fractions and detection of mahanimbine in M paniculata leaf extract by thin-layer chromatography was also carried out. RESULTS： Fraction F5.3 （25 mg/kg） and EASF （20 mg/kg） showed significant anxiolytic and antidepressant activity, respectively. Thin-layer chromatography and phytochemical screening demonstrated the absence of mahanimbine in M. paniculata leaves. coumarins were observed to be responsible for the anxiolytic activity. CONCLUSION： The results validate the traditional use of M. paniculata leaves in the treatment of menta disorders.

A facile approach for the first synthesis of ocimarin, naturally occurring coumarins, was developed by employing a cerium （III） chloride heptahydrate-catalyzed Pechmann condensation of phenols and β-keto esters in a solvent-free system as key step, by which ocimarin was achieved by three steps starting from acetylacetic ether and resorcinol with total yield 23.2%. All structures of new compounds were confirmed by IR, ^1H NMR and MS.

Ten coumarin derivatives containing（E）-methyl 2-（methoxyimino）-2-phenylacetate were synthesized and *** compounds were identified by IR,~1H NMR and elemental *** test results indicated that compound 5j（R^1 is methyl and R^2 is n-C_6H_（13）） was the optimal structure in this paper with good fungicidal activity against CDM（85%） at 6.25 mg/L concentration.

Inspired from occurrence of anti-inflammatory activity of 3-substituted coumarins and antiulcer activity of various 2-substituted benzimidazoles,novel compounds have been designed by coupling coumarin derivatives at 3-position directly or through amide linkage with benzimidazole nucleus at *** resultant compounds are expected to exhibit both anti-inflammatory and antioxidant activities along with less gastric toxicity *** series of coumarin–benzimidazole derivatives(4a–e and 5a–e)were synthesized and evaluated for anti-inflammatory activity and antioxidant *** 4c,4d and 5a displayed good anti-inflammatory(45.45%,46.75%and 42.85%inhibition,respectively,versus 54.54% inhibition by indomethacin)and antioxidant(IC_(50) of 19.7,13.9 and 1.2 mmol/L,respectively,versus 23.4 mmol/L for butylatedhydroxytoluene)*** of ulcer index and in vivo biochemical estimations for oxidative stress revealed that compounds 4d and 5a remain safe on gastric mucosa and did not induce oxidative stress in *** of various molecular properties suggests the compounds to be sufficiently bioavailable.

A quality control （QC） strategy for quantitative and qualitative analysis of “common peaks” in chemical fingerprint was proposed to analyze Yuanhu Zhitong tablet （YZT）, using high performance liquid chromatography with diode array detector and tandem mass spectrometry （HPLC-DAD-MS/MS）. The chromatographic separation was achieved on an Agilent Eclipse plus C18 column with a gradient elution using a mixture of 0.4‰ ammonium acetate aqueous （pH 6.0 adjusted with glacial acetic acid） and acetonitrile. In chemical fingerprint, 40 peaks were assigned as the “common peaks”. For quantification of “common peaks”, the detection wavelength was set at 254 nm, 270 nm, 280 nm and 345 nm, respectively. The method was validated and good results were obtained to simultaneously determine 10 analytes （protopine, jatrorrhizine, coptisine, palmatine, berberine, xanthotoxin, bergapten, tetrahydropalmatine, imperatorin and isoimperatorin）. For qualification of “common peaks”, 33 compounds including 10 quantitative analytes were identified or tentatively characterized using LC-MS/MS. These results demonstrated that the present approach may be a powerful and useful tool to tackle the complex quality issue of YZT.

Hormone Receptor positive (HR+) breast cancer is the most common malignancy in women. New strategies in the treatments have targeted the estrogen biosynthesis pathways including the inhibition of the aromatase and 17β-HSD1 enzymes. The present work, describes the study of a new family of 9 hybrid compounds derived from estrone attached to a coumarin fragment, linked through different lengths of hydrocarbon chains. The activity of these compounds was evaluated by molecular docking with two relevant enzymes in breast cancer (HR+). It has been proposed nine compounds as 17β-HSD1 inhibitors and six as aromatase inhibitors. We found important interactions with key amino acids at the orthosteric site of each enzyme and their score values compared to the crystallographic ligand. The in silico analysis showed good score values in the proposed compounds, where the steroidal portion presented important interactions with Met374 and Tyr155 in aromatase and in 17β-HSD1 respectively. Highlighting Compounds 2, 5 and 8 with an aromatic ring at the C4 position of the coumarin moiety, which favored arene-H type interactions essential for protein-ligand recognition. In addition, the results related to the 17β-HSD1 enzyme demonstrated how the length of the linker influences the interaction;the best score was found for derivative 8 with a chain of 8 methylenes.

This review provides a comprehensive overview of the phytochemistry, medicinal properties, and biological activities of the Artemisia species. Artemisia species exhibit a wide range of morphological and phytochemical diversity, showing promise for the treatment of various diseases in humans, plants, and animals. Phytochemicals found in Artemisia spp. include terpenoids, flavonoids, coumarins, caffeoylquinic acids, sterols, and acetylenes. Dietary flavonoids have been linked to reduced cell aging, lipid peroxidation, and cancer risks. Sterols, including plant sterols, play a significant role in reducing the plasma cholesterol levels. Essential oils from Artemisia plants contain volatile chemicals with antibacterial, antiviral, and antifungal properties that contribute to their biological activity. This study aimed to elucidate the effective compounds of Artemisia plants and their medicinal benefits and biological activities.

In the present study, we isolated 18 known compounds from Tirpitzia ovoidea （Linaceae）, including seven terpenoids （1-7）, three simple phenylproponoids （8-10）, three coumarins （11 13） and five other compounds （14 18）. The structures of these compounds were identified by spectroscopic methods and compared with those reported in the literature. Compounds 1-16 were isolated from this plant as well as the Linaceae family for the first time.
