Genome-wide identification of microRNAs involved in the regulation of fruit ripening and climacteric stages in melon(Cucumis melo)

Fruit ripening is influenced by multiple plant hormones and the regulation of genes.However,studies on posttranscriptional regulators(e.g.,miRNAs)of fruit growth and ripening are limited.We used miRNA sequencing and degradome methods to identify miRNAs and their target genes in melon(Cucumis melo cv.Hetao melon).A total of 61 conserved miRNAs and 36 novel miRNAs were identified from fruit growth,ripening,climacteric,and postclimacteric developmental stage samples,of which 32 conserved miRNAs were differentially expressed between developmental stage samples.Sixty-two target genes of 43 conserved miRNAs and 1 novel miRNA were identified from degradome sequencing.To further investigate miRNA influencing fruit ripening,transgenic melon plants overexpressing pre-cmemiR393(cme-miR393-OE)were generated and characterized.The results showed that fruit ripening was delayed in cme-miR393-OE transgenic lines compared to nontransgenic fruits.The target of cme-miR393 was also identified,and the expression of CmAFB2 was repressed in transgenic plants.These results provide evidence that miRNA regulates melon fruit ripening and provide potential targets to improve the horticultural traits of melon fruit.

Plant proton-translocating pyrophosphatases （H^＋-PPases） are generally thought of as tonoplast-localized proton pumps that utilize the energy released by hydrolysis of the phosphoanhydride bond of pyrophosphate （PPi） to assist in the acidification of the vacuole.It has been suggested that H^＋-PPases function as an additional acidification mechanism to complement the ubiquitous V-ATPase,especially in young,growing tissues where PPi is abundant and ATP is depleted （Shiratake et al.,1997）.

Based on the property of the discrete model entirely inheriting the symmetry of the continuous system,we present a method to construct exact solutions with continuous groups of transformations in discrete nonconservative systems.The Noether's identity of the discrete nonconservative system is obtained.The symmetric discrete Lagrangian and symmetric discrete nonconservative forces are defined for the system.Generalized quasi-extremal equations of discrete nonconservative systems are presented.Discrete conserved quantities are derived with symmetries associated with the continuous system.We have also found that the existence of the one-parameter symmetry group provides a reduction to a conserved quantity;but the existence of a two-parameter symmetry group makes it possible to obtain an exact solution for a discrete nonconservative system.Several examples are discussed to illustrate these results.

For El-Nabulsi's fractional Birkhoff system, Mei symmetry perturbation, the corresponding Mei-type adiabatic invariants and Noether-type adiabatic invariants are investigated in this paper. Firstly, based on El-NabulsiBirkhoff fractional equations, Mei symmetry and the corresponding Mei conserved quantity, Noether conserved quantity deduced indirectly by Mei symmetry are studied. Secondly, Mei-type exact invariants and Noether-type exact invariants are given on the basis of the definition of adiabatic invatiant. Thirdly, Mei symmetry perturbation, Mei-type adiabatic invariants and Noether-type adiabatic invariants for the disturbed El-Nabulsi's fractional Birkhoff system are studied.Finally, two examples, Hojman-Urrutia problem for Mei-type adiabatic invariants and another for the Noether-type adiabatic invariants, are given to illustrate the application of the results.

Roots are subterranean organs and normally follow the gravity vector to grow downward. However, in nature, roots can be exposed to sunlight in a negative phototropism response termed root light avoidance （Kutschera and Briggs, 2012）. Such tropic responses are characterized by organ curvature caused by asymmetric growth between different sides of the root, which generally correlates with an asymmetric distribution of the plant hormone auxin （Wang et al., 2015）.

The isolated liver nuclei and the mature sperm nuclei from Rana nigromaculata and Bufo bufo asiaticus had been investigated by indirect immunofluorescenee method with anti serum specific to the spindle fiber attachments (SFAs) from the scleroderma patients with CREST syndrome. It was found that in the isolated liver nuclei and the mature sperm nuclei of these animals there are discrete speckles which bind the antibody specific to the SFAs. The maximum number of these speckles counted in a single sperm nuclei of the frog corresponds with the haploid chromosome number of this animal (n=13). The haploid chromosome complement of the toad is 11 and the maximum number of the speckles counted in a single sperm nucleus of this species is also 11. These findings suggest that the scleroderma serum can be used to identify the SFAs of the amphibias and the antigens of SFAs are conserved in the mature sperms of these animals. The implication of these conclusions is discussed.

In the recent twenty years, the study on the Lie symmetries and conserved quantities of constrained mechanical systems has been making great progress[1-5]. Up to now, however, all the studies are limited to the mechanical systems with ideal bilateral constraints. This note further studies the Lie symmetries of mechanical systems with unilateral holonomic constraints, and two problems of Lie symmetries for the systems are put forward and solved, i.e. finding the conserved quantity from a Lie symmetric transformation of the system and finding the corresponding Liesymmetry from an integral of the systemi ? 梛 ? Jsymmetry from an integral of the system.
