Chromatin condensation but not DNA integrity of pig sperm is greater in the sperm-rich fraction

Background Protamination and condensation of sperm chromatin as well as DNA integrity play an essential role during fertilization and embryo development.In some mammals,like pigs,ejaculates are emitted in three separate fractions:pre-sperm,sperm-rich(SRF)and post sperm-rich(PSRF).These fractions are known to vary in volume,sperm concentration and quality,as well as in the origin and composition of seminal plasma(SP),with differences being also observed within the SRF one.Yet,whether disparities in the DNA integrity and chromatin condensation and pro-tamination of their sperm exist has not been interrogated.Results This study determined chromatin protamination(Chromomycin A3 test,CMA_(3)),condensation(Dibromobi-mane test,DBB),and DNA integrity(Comet assay)in the pig sperm contained in the first 10 m L of the SRF(SRF-P1),the remaining portion of the sperm-rich fraction(SRF-P2),and the post sperm-rich fraction(PSRF).While chromatin protamination was found to be similar between the different ejaculate fractions(P>0.05),chromatin condensation was seen to be greater in SRF-P1 and SRF-P2 than in the PSRF(P=0.018 and P=0.004,respectively).Regarding DNA integrity,no differences between fractions were observed(P>0.05).As the SRF-P1 has the highest sperm concentra-tion and ejaculate fractions are known to differ in antioxidant composition,the oxidative stress index(OSi)in SP,calcu-lated as total oxidant activity divided by total antioxidant capacity,was tested and confirmed to be higher in the SRF-P1 than in SRF-P2 and PSRF(0.42±0.06 vs.0.23±0.09 and 0.08±0.00,respectively;Pcondensation than the PSRF.This could be related to the OSi of each fraction.

Durable dropwise condensation of saturated vapor is of significance for heat transfer and energy saving in extensive industrial applications.While numerous superhydrophobic surfaces can promote steam condensation,maintaining discrete microdroplets on surfaces without the formation of a flooded filmwise condensation at high subcooling remains challenging.Here,we report the development of carbon nanotube array-embedded hierarchical composite surfaces that enable ultra-durable dropwise condensation under a wide range of subcooling(ΔT_(sub)=8 K–38 K),which outperforms existing nanowire surfaces.This performance stems from the combined strategies of the hydrophobic nanostructures that allow efficient surface renewal and the patterned hydrophilic micro frames that protect the nanostructures and also accelerate droplet nucleation.The synergistic effects of the composite design ensure sustained Cassie wetting mode and capillarity-governed droplet mobility(Bond numbercondensation heat transfer.Our design provides a feasible alternative for efficiently transferring heat in a vapor environment with relatively high temperatures through the tunable multiscale morphology.

Bcl10 phosphorylation-dependent droplet-like condensation positively regulates DNA virus-induced innate immune signaling

B-cell lymphoma 10(Bcl10) is a scaffolding protein that functions as an upstream regulator of NF-κB signaling by forming a complex with Mucosa-associated lymphoid tissue lymphoma translocation protein 1(Malt1) and CARD-coiled coil protein family. This study showed that Bcl10 was involved in type I interferon(IFN) expression in response to DNA virus infection and that Bcl10-deficient mice were more susceptible to Herpes simplex virus 1(HSV-1) infection than control mice. Mechanistically,DNA virus infection can trigger Bcl10 recruitment to the STING-TBK1 complex, leading to Bcl10 phosphorylation by TBK1.The phosphorylated Bcl10 undergoes droplet-like condensation and forms oligomers, which induce TBK1 phosphorylation and translocation to the perinuclear region. The activated TBK1 phosphorylates IRF3, which induces the expression of type I IFNs.This study elucidates that Bcl10 induces an innate immune response by undergoing droplet-like condensation and participating in signalosome formation downstream of the c GAS-STING pathway.

The superfluorescence effect has received extensive attention due to the many-body physics of quantum correlation in dipole gas and the optical applications of ultrafast bright radiation field based on the cooperative quantum state.Here,we demonstrate not only to observe the superfluorescence effect but also to control the cooperative state of the excitons ensemble by externally applying a regulatory dimension of coupling light fields.A new quasi-particle called cooperative exciton-polariton is revealed in a light-matter hybrid structure of a perovskite quantum dot thin film spin-coated on a Distributed Bragg Reflector.Above the nonlinear threshold,polaritonic condensation occurs at a nonzero momentum state on the lower polariton branch owning to the vital role of the synchronized excitons.The phase transition from superfluorescence to polariton condensation exhibits typical signatures of a decrease of the linewidth,an increase of the macroscopic coherence as well as an accelerated radiation decay rate.These findings are promising for opening new potential applications for super-brightness and unconventional coherent light sources and could enable the exploitation of cooperative effects for quantum optics.

Supersonic expansion and condensation characteristics of hydrogen gas under different temperature conditions

This paper introduced supersonic expansion liquefaction technology into the field of hydrogen liquefaction. The mathematical model for supersonic condensation of hydrogen gas in a Laval nozzle model was established. The supersonic expansion and condensation characteristics of hydrogen gas under different temperature conditions were investigated. The simulation results show that the droplet number rises rapidly from 0 at the nozzle throat as the inlet temperature increases, and the maximum droplet number generated is 1.339 x 1018 kg-1 at inlet temperature of 36.0 K. When hydrogen nucleation occurs, the droplet radius increases significantly and shows a positive correlation with the increase in the inlet temperature, and the maximum droplet radii are 6.667 x 10-8 m, 1.043 x 10-7 m, and 1.099 x 10-7 m when the inlet temperature is 36.0 K, 36.5 K, and 37.0 K, respectively. The maximum nucleation rate decreases with increasing inlet temperature, and the nucleation region of the Laval nozzle becomes wider. The liquefaction efficiency can be effectively improved by lowering the inlet temperature. This is because a lower inlet temperature provides more subcooling, which allows the hydrogen to reach the thermodynamic conditions required for large-scale condensation more quickly. Background:Genetic selection in cattle has been directed to increase milk production.This,coupled to the fact that the vast majority of bovine artificial inseminations(AI)are performed using cryopreserved sperm,have led to a reduction of fertility rates over the years.Thus,seeking sensitive and specific sperm biomarkers able to predict fertility rates is of vital importance to improve cattle reproductive efficiency.In humans,sperm chromatin condensation evaluated through chromomycin A3(CMA3)has recently been purported to be a powerful biomarker for sperm functional status and male infertility.The objectives of the present study were:a)to set up a flow cytometry method for simultaneously evaluating chromatin condensation and sperm viability,and b)to test whether this parameter could be used as a predictor of in vivo fertility in bulls.The study included pools of three independent cryopreserved ejaculates per bull from 25 Holstein males.Reproductive outcomes of each sire were determined by non-return rates,which were used to classify bulls into two groups(highly fertile and subfertile).Results:Chromatin condensation status of bovine sperm was evaluated through the combination of CMA3 and Yo-Pro-1 staining and flow cytometry.Sperm quality parameters(morphology,viability,total and progressive motility)were also assessed.Pearson correlation coefficients and ROC curves were calculated to assess their capacity to predict in vivo fertility.Sperm morphology,viability and total motility presented an area under the ROC curve(AUC)of 0.54,0.64 and 0.68,respectively(P>0.05),and thus were not able to discriminate between fertile and subfertile individuals.Alternatively,while the percentage of progressively motile sperm showed a significant predictive value,with an AUC of 0.73(P=0.05),CMA3/Yo-Pro-1 staining even depicted superior results for the prediction of in vivo fertility in bulls.Specifically,the percentage of viable sperm with poor chromatin condensation showed better accuracy and precision to predict in vivo fertility,wi

In Tube condensation:Changing the Pressure Drop into a Temperature Difference for a Wire-on-Tube Heat Exchanger

A theoretical study based on the Penalty factor(PF)method by Cavallini et al.is conducted to show that the pressure drop occurring in a wire-on-tube heat exchanger can be converted into a temperature difference for two types of refrigerants R-134a and R-600a typically used for charging refrigerators and freezers.The following conditions are considered:stratiﬁed or stratiﬁed-wavyﬂow condensation occurring inside the smooth tube of a wire-on-tube condenser with diameter 3.25,4.83,and 6.299 mm,condensation temperatures 35℃,45℃,and 54.4℃ and cover refrigerant massﬂow rate spanning the interval from 1 to 7 kg/hr.The results show that the PF variation is not linear with vapor quality and attains a maximum when the vapor quality is 0.2 and 0.18 for the R-134a and R-600a refrigerants,respectively.The PF increases with the refrigerant massﬂow rate if the inner diameter and saturation temperature constant,and it decreases on increasing the inner diameter to 6.299 mm for constant refrigerant massﬂow rate and saturation temperature.The PF for R-600a is higher than that for R-134a due to the lower saturation pressure in theﬁrst case.Furthermore,a stratiﬁedﬂow produces higher PF in comparison to the annularﬂow due to the effect of the surface tension.

Effect of crystallization on purity of volatile metallic magnesium prepared from a one-step multi-region condensation process under vacuum condition

We hereby report a green and simple volatilization-condensation process to prepare high-purity magnesium in different crystal forms under vacuum condition. This method can be also used for the analysis of other metals, which can be separated by the same concept. In addition, Mg condensation is a very fast process, and the corresponding easy growth of the Mg crystals is promoted when the concentration of magnesium vapor is raised. From the view of crystallization, ultrafast crystal growth of high-purity magnesium is a barrier-free and orderly process. Interestingly, different condensed forms of Mg obtained via this ultrafast volatilization-condensation process are showing different purities. The distinct morphological formation of Mg in different condensed form is probably attributed to the differences of surface energy change of Mg during the condensation process.

Polariton-hybridization of light-matter oscillations can emerge from various quasiparticles,such as phonon,plasmon,exciton and magnon.Particularly,exciton polaritons are bosonic quasiparticles with half-light,half-matter nature,which are originated from strong coupling between excitons and microcavity photons.The half-light nature results in extremely small effective mass,making it feasible to achieve high temperature even room-tempera-ture Bose-Einstein condensation(BEC).Meanwhile,the half-matter nature leads to strong nonlinear interaction,which is missing between photons and can promote the polaritons relaxation to ground state and give rise to low threshold polariton lasing,compared to photonic lasing.The exciton polaritons are of great importance in applications of quantum simulation,topological quantum optics,ultrafast optical switch and low threshold lasers.

Evolution in physiochemical and cloud condensation nuclei activation properties of crop residue burning particles during photochemical aging

As a main form of biomass burning in agricultural countries, crop residue burning is a significant source of atmospheric fine particles. In this study, the aging of particles emitted from the burning of four major crop residues in China was investigated in a smog chamber.The particle size distribution, chemical composition and cloud condensation nuclei(CCN)activity were simultaneously measured. The properties of crop residue burning particles varied substantially among different fuel types. During aging, the particle size and mass concentration increased substantially, suggesting condensational growth by formation of secondary aerosols. The particle composition was dominated by organics. Aging resulted in considerable enhancement of organics and inorganics, with enhancement ratios of 1.24–1.44 and 1.33–1.76 respectively, as well as a continuous increase in the oxidation level of organics. Elevated CCN activity was observed during aging, with the hygroscopicity parameter κ varying from 0.16 to 0.34 for fresh particles and 0.19 to 0.40 for aged particles.Based on the volume mixing rule, the hygroscopicity parameter of organic components(κorg) was derived. κorgexhibited an increasing tendency with aging, which was generally consistent with the tendency of the O:C ratio, indicating that the oxidation level was related to the hygroscopicity and CCN activity of organic aerosols from crop residue burning. Our results indicated that photochemical aging could significantly impact the CCN activation of crop burning aerosols, not only by the production of secondary aerosols, but also by enhancing the hygroscopicity of organic components, thereby contributing to the aerosol indirect climate forcing.
