Efficacy of active ingredients in Qingdai(Indigo Naturalis)on ulcerative colitis:a network pharmacology-based evaluation

OBJECTIVE:To elucidate the protective effect of Qingdai(Indigo Naturalis,QD)on ulcerative colitis(UC)by means of in silico and in vivo ***:A systems pharmacology analysis was performed to predict the active components of QD whereas the putative biological targets of QD against UC were obtained through target fishing,network cons-truction and enrichment ***,we examined the ameliorative effect of QD in a mouse model of dextran sulfate sodium(DSS)-induced *** the 10-day experiment,the control and diseased mice were given with oral gavages of QD(1.3 g raw herbs·kg^(-1)·d^(-1))or 5-aminosalicylic acid(5-ASA,100 mg·kg^(-1)·d^(-1))every *** underlying pharma-cological mechanisms of QD in UC were determined using polymerase chain reaction tests,histological staining,enzyme-linked immunoassays,and Western blotting ***:Searching from various network pharmacology databases,29 compounds were identified in *** to the screening criteria suggested by TCMSP(***≥30%and DL≥0.18),nine of them were considered the active ingredients that contribute to the ameliorative effects of QD on different mouse models of *** importantly,the protective effect of QD on DSS-induced colitis was significantly associated with modulations of the expression levels of glycogen synthase kinase 3-β(Gsk3-β)and forkhead box p3(Foxp3),which are widely considered as important regulators of excessive inflammatory ***:The results of this study provide solid scientific evidence for the use of QD or its core active components in the clinical management of UC.

OBJECTIVE:To investigate the effect of a decoction made from the Traditional Chinese Medicine Wumei pill,on regulatory T cells and interleukin-10(IL-10) in a rat model of ulcerative colitis induced by2,4,6-trinitrobenzene sulfonic acid(TNBS).METHODS:Rat ulcerative colitis was induced with *** modeled rats were randomly divided into six groups:normal control group;model group;sulfasalazine suppositories treatment group;and high,moderate,and low dosage of Jiaweiwumei decoction groups(12 rats each).Colon injury index was evaluated after 14 *** peripheral blood lymphocyte separation,CD4 + T cells and CD4+/CD25+ T cell percentage was detected by flow *** content of IL-10 in serum and intestinal mucosa tissue was detected by sandwich enzyme-linked immunosorbent ***:Colon injury indices in the decoction groups were effectively reduced,compared with the model group(P ulcerative colitis.

Effect of compound Sophorae Flavescentis Jiechangrongcapsule on expression of NF-κB p65 and STAT6 in theintestinal mucosa of patients with ulcerative colitis

The effects of compound Sophorae Flavescen-tis Jiechangrong capsule(CSFJC)on the expression of nuclear factor-κB p65(NF-κB p65)and signal transducer and activator of transcription 6(STAT6)in the intestinal mucosa of patients with ulcerative colitis and the possible mechanism were *** patients with ulcerative colitis were randomly divided into a traditional Chinese medicine(TCM)group(n=11)treated by CSFJC and a western medicine(WM)group(n=7)treated by Sulfasalazine *** treatment duration lasted eight *** and after the treatment,the symptoms and the physical signs were observed,and the routine stool test,the colonoscopy,and pathological examination were performed in the two *** expression levels of NF-κB p65 and STAT6 were detected by using *** results showed that the total effective rate of the curative effectiveness in TCM and WM groups was 100%and 71.4%,respectively,and the total effective rate of colonic mucosa lesion in TCM and WM groups was 90.9%and 71.4%,respectively,with the differences being signiﬁcant(all Pulcerative colitis,which is a possible mechanism for CSFJC treating ulcerative colitis.
