Objective：To observe the time-effect relation of extracts from ginseng,notoginseng and chuanxiong on angiotensin II（AngⅡ）-induced senescence of vascular endothelial cells and explore the feature of Chinese medicine against vascular diseases.Methods：Human umbilical vein endothelial cells（HUVECs）cultured in vitro were stimulated with 10^-6 mol/L Ang Ⅱ to induce cell senescence,which were divided into 4groups,the blank control group,the Ang Ⅱ model group,the extracts group and the telmisartan group.The（J-gal was used to identify senescence of cells,the cell counting kit-8 method was applied to assess the cell viability,the cell function was examined with the level of endothelial nitric oxide synthase（eNOS） and the flow cytometry was used for analyzing the cell cycle changes.Results：Compared with the control cells,the cells positive for β-gal staining was significantly increased in the Ang Ⅱ model group,and showed cell cycle arrest at G0/G1 phase with decreased S and G2/M phase ceil percentage,eNOS expression and cell viability（P〈0.05）.The extracts and telmisartan treatment of Ang II-induced cells resulted in decreased β-gal positive cells with a reduction in G0/G1phase cells and an increasing in S,G2/M phase ceils and eNOS expression（P〈0.05）.At 24 h,the extracts were more effective than telmisartan（P〈0.05）;while telmisartan was more effective at 48 h（P〈0.05）.Conclusion：Extracts from ginseng,notoginseng and chuanxiong can delay Ang II-induced aging of HUVECs and may play an important role in early senescence.

优化川芎(Ligusticum chuanxiong Hort.)中阿魏酸的常温超高压提取条件,应用高效液相色谱法测定提取液中的阿魏酸含量,以阿魏酸提取率为评价指标进行L9(34)正交试验。结果表明,常温超高压提取川芎中阿魏酸的最优工艺为乙醇体积分数70%,压力300 MPa,固液比1∶50(m∶V,g∶mL),时间3.0 min,提取1次。超高压提取技术作为一种新的提取方法,提取川芎中阿魏酸稳定、可行。

Objective:To observe the clinical efficacy of chuanxiong Hualiu Mixture in the treatment of blood stasis and phlegm stagnation type of ovarian cyst and the effects on physical and chemical indicators.Methods: A total of 80 patients with blood stasis and phlegm stagnation type of ovarian cyst were randomly divided into two groups, research group (n=40) took chuanxiong Hualiu Mixture, control group (n=40) took Guizhi Fuling Capsule, 3 months was a course of treatment, and the curative effect was compared between two groups.Results:(1) The overall response rate of chuanxiong Hualiu mixture group after 3 months of treatment was 82.5%, that of Guizhi Fuling Capsule group was 62.5%, the overall response rate of chuanxiong Hualiu mixture group was higher than that of control group, and the difference was statistically significant;(2) before and after treatment, sex hormone and hemorheology index E2 of both groups decreased significantly, whole blood viscosity, whole blood reduced viscosity and plasma viscosity decreased significantly, the results were statistically significant, but there was no difference between two groups;(3) rank sum test of TCM symptom efficacy between two groups before and after treatment showed that the overall response rate of chuanxiong Hualiu mixture group was 95%, the overall response rate of Guizhi Fuling Capsule group was 85%, the overall response rate of research group was better than that of control group, and the difference was statistically significant.Conclusion:chuanxiong Hualiu mixture has good curative effect for the treatment of ovarian cyst, it can effectively improve the patients' blood stasis and phlegm stagnation constitution, and its mechanism of action is associated with lowering estrogen levels and improving hemorrheology.

为了解川芎(Ligusticum chuanxiong Hort.)阿魏酸生物合成相关基因的密码子使用偏好性特点,为运用基因工程技术实现阿魏酸的异源生物合成提供理论依据,对川芎转录组中共50 108条Unigenes使用Codon W、Cusp和Chips进行在线分析。结果表明,总GC含量为41.4%,有效密码子占总数的16.17%,最优密码子偏好以A/U结尾,表明川芎转录组Unigenes密码子偏好程度整体水平不高。比较分析了川芎转录组中阿魏酸生物合成相关基因(PAL、C4H、C3H与COMT)与不同模式生物的稀有密码子,表明与大肠杆菌基因组密码子使用频率差值较大的有4个,与酵母、烟草和拟南芥基因组差值较大的均有3个,这预示着川芎阿魏酸生物合成相关基因在酵母、烟草和拟南芥中的表达效率较高。

[目的]建立和优化适合川芎的ISSR-PCR反应体系。[方法]以川芎叶片为材料,利用改良CTAB法提取了叶片基因组DNA,利用单因素试验分析了DNA模板、Mg2＋、dNTP、引物和TaqDNA聚合酶的浓度对ISSR-PCR反应的影响,对适合川芎的ISSR-PCR反应条件进行了优化。[结果]确定了适合川芎的ISSR-PCR反应体系为25.0μl,其中含2.5μl10×TaqDNA聚合酶缓冲液、2.1mmol/LMgCl2、300μmol/LdNTP、0.4μmol/L引物、1.0UTaqDNA聚合酶和20~40ng模板DNA。[结论]为进一步对全国17个不同分布区的川芎资源进行遗传多样性研究奠定了基础。

Comparative Study of chuanxiong and Dextran 40 in the Treatment of Acute Cerebral Infuriction

This paper reports the results of a double-blind trial in 220 patients with acute cerebral in-farction evidenced by computerized tomography, who were randomly divided into a chuanxiong group(134 cases) and a low molecular weight dextran group (86 cases) . A weighted scoring system was adopt-ed to evaluate the neurological function and living capability. The results showed that the total therapeuticefficacy rate in the chuanxiong group and in the dextran 40 group were 86. 6% and 62. 8% respectively.The effects of chuanxiong on the treatment of acute cerebral infarction was superior to the low molecularweight dextran, and the difference between the two groups was statistically significant (P

The authors established experimental atherosclerosis in rabbits by means of combininghyperlipidemia and repeatedly administered intravenously with serum ot other origins to make the im-munopathologic injuries of arterial endothelia in order to observe the preventive and therapeutic effects otchuanxiong on experimental atherosclerosis. The authors observed that chuanxiong could significantly de-crease the levels of the serum cholesterol and low density lipoprotein (Pchuanxiong could reduce the red cell deformability. This studysuggests that chuanxiong plays an active role in preventive and therapeutic effect on atherosclerosis.

两面针、川芎、生首乌内生真菌的分离纯化及抑菌性研究

采用组织块法从两面针(Zanthorμlum nitidum(Roxb.)DC)、川芎(Ligusticum chuanxiong Hort)生首乌(Fallopia multiflora)组织中分别分离得到7、11、6株内生真菌,以大肠杆菌(Escherichia coli)、肺炎球菌(Diplococcus pneumoniae)、金黄色葡萄球菌(Staphylococcus aureus)绿脓杆菌(Pseudomonas aeruginosa)、地衣杆菌为供试菌种,对所有两面针、川芎生首乌内生真菌进行抑菌性试验,结果显示,两面针3株内生真菌至少对1株测试病原菌有抑制作用。
