Effect of Reelin signaling on pre-plate splitting during cerebral cortex development

The development of the 6-layered cerebral neocortex is one of the most important events during nervous system development, and disturbances could result in various malformations, causing clinically intractable diseases, such as epilepsy and cerebral palsy. Pre-plate splitting is the first developmental step of the cortical plate formation. Without correct pre-plate splitting, normal cerebral cortex structures are disturbed. The Reelin-Dabl molecular pathway plays a critical role during cerebral cortex development, and deficiencies in this pathway result in failed pre-plate splitting and an inverted cortical plate. This paper summarizes findings involving Reelin and pre-plate splitting and further explores the precise role of Reelin during pre-plate splitting.

Fluorescent histochemical observation and estimation of the content ofcatecholamine(CA)in cerebral cortex of twenty rats with unilateral occlusion ofmiddle cerebral artery were performed in this present *** d after localcerebral infarction,the CA granules which were of different fluorescent featurefrom those of normal cortex were seen in the marginal area of infarct. The contentof norepinephrine in bilateral cortexes was significantly reduced, but that ofdopamine was unchanged.

＜正＞Hereditary spastic paraplegias（HSPs） are human genetic disorders characterized by lower extremity spasticity and *** the HSPs,SPG3A is particularly notable for its early onset of spastic paraplegia. SPG3A is caused by mutations in atlastin-1,which belongs to the dynamin family of *** previous studies found that atlastin-1 is enriched in brain,little is known about their physiological functions,especially in neuronal ***,we report that expression of atlastin-1 mRNA was restricted in the deep layer of mouse cerebral cortex during early *** expression of atlastin-1 and R217Q,a mutant form defective of GTPase,did not affect neuronal generation,migration and axon growth in the postnatal brains of less than P7. However,when the cortices were examined at a late developmental stage of P14,the expression of atlastin-1 led to fewer and scattered labeled-neurons as compared to that expressing the control *** expressing the R217Q were present normally at P14,which suggests that the GTPase activity of atlastin-1 may be important for its early restricted *** study suggests a possibility that atlastin-1 might act as a neuropathogenic mechanism of hereditary spastic paraplegias if it expresses abnormally during the development.

Aims Depression is associated with significant morbidity and functional disability,and it is thus important to reveal the mechanism of depression.A variety of studies suggest an involvement of dopaminergic receptors in the pathophysiological mechanism of none-stress-associated depression-like behavior in ***,controversy exists about whether chronic stressors act on dopaminergic receptors in cerebral ***,we investigated the level of dopaminergic receptor-2(DRD2) protein and the possible mechanisms in a chronic unpredictable mild stress(CUMS) rat model of *** The chronic stressed depression rats were established by chronic unpredictable mild stress and separation after 28 *** tests were measured using forced swim test and sucrose preference test. Radioimmunoassay was utilized to detect the change of cAMP *** protein levels were measured by western blotting and Immunohistochemical *** The results showed that CUMS-induced depressive-like symptoms in mice characterized by reduced sucrose consumption, body weight,and increased duration of immobility in forced swimming ***,we found that chronic stressors,by down-regulated the expression of DRD2,could affect the cerebral cortex and consequently cause depression by decreasing the expression of protein kinase A(PKA), transcription factor cAMP response element-binding protein(CREB) and phospho-CREB (p-CREB) in cerebral ***,in rat model of depression,decreased cyclic adenine monophosphate(cAMP) levels was present at the same time,which is consistent with clinical findings in depressed *** Our findings suggested that dopaminergic system malfunction could play a central role in stress-related disorders such as depression.

A rat model of cerebral ischemia/reperfusion was established by suture occlusion of the left middle cerebral *** situ hybridization results showed that the number of brain-derived neurotrophic factor mRNA-positive cells in the ischemic rat cerebral cortex increased after cerebral ischemia/reperfusion *** frequency continuous wave electroacupuncture(frequency 2-6 Hz,current intensity 2 mA) stimulation of the brachial plexus trunk on the healthy(right) side increased the number of brain-derived neurotrophic factor mRNA-positive cells in the ischemic cerebral cortex 14 days after cerebral ischemia/reperfusion *** the same time,electroacupuncture stimulation of the healthy brachial plexus truck significantly decreased neurological function scores and alleviated neurological function *** findings suggest that electroacupuncture stimulation of the brachial plexus trunk on the healthy(right) side can greatly increase brain-derived neurotrophic factor mRNA expression and improve neurological function.

Objective Bromodomain-containing protein 2（Brd2） is one of four BET（bromodomain and extra-terminal domain） family members. Various functions have been ascribed to the Brd2, including cellular proliferation, apoptosis and transcription. However, little is known about the potential function of Brd2 in the development of cerebral cortex. Methods To understand further the precise function of Brd2 in cortical development, fluorescent in situ hybridization combined with immunofluorescent histochemistry were used to detect the expression and feature of Brd2 in the cerebral cortex. We used the technique of in utero electroporation and silenced Brd2 successfully in subsets of neural progenitors in the developing mouse cortex. Results Brd2 is most abundant in the cerebral cortex. Brd2 is highly expressed in neurons but is undetectable in glial cells, indicating an important role of Brd2 in neuronal function. Down regulating Brd2 by RNA interference in cerebral cortex induced cortical development abnormalities. Conclusion Our findings reveal an important role of Brd2 in the development of cerebral cortex.

＜正＞The mammalian neocortex consists of six distinct cellular layers of local circuit neurons and projection *** studies have discovered a number of transcription factors that control specific axonal projections and layer formation in the *** of Satb2（special AT-rich sequence binding protein） in the upper layers （Ⅱ-Ⅲ） determines the callosal neuronal projection to the contralateral *** in situ hybridization and immuofluorescence,we found that the expressions of mRNA and protein for Satb2 are not identical in the cortex,which suggests that micro RNAs may be involved in regulation of the cortical *** this end,layer V neurons were sorted by FACS from the Fezf2-eGFP mice that express GFP in the subcortical projection neurons of layer V. MicroRNA expression profiling found that MicroRNA-218 was enriched in the sorted layer V *** of MicroRNA-218 had more than twofold increase in the layer V neurons compared to that of non-fluorescent control as determined by real time PCR assay of the micro *** vivo expression of MicroRNA-218 significantly reduced the mRNA for Onecutl,Medga1,and Sox5 but not *** vitro expression of the microRNA inhibited the luciferase activity of a fragment containing the binding site of the microRNA in the Satb2 3’UTR,suggesting that the translation of Satb2 may be reduced by *** hypothesize that micro RNAs may regulate the expression of the layer specific transcription factor and modulate the development of the cerebral cortex in vivo.
