Environmentally Friendly Tannic Acid-Furfuryl Alcohol-Soybean Isolate/casein Composite Foams Reinforced with Wood Fibers

In this study,two series of foams based on tannic acid(TA),furfuryl alcohol(FA),soybean protein isolate(SPI),and casein(CA),namely TA–FA–SPI(TS series)and TA–FA–CA(TC series)were developed,and their properties were enhanced by adding poplar fibers(WF).From the samples produced,a complete set of characterization was performed including possible crosslinking reactions,morphology,mechanical properties,flame retardancy,thermal insulation and thermal ***-transform infrared spectroscopy(FTIR)revealed possible covalent crosslinking among the components and hydrogen bonding between WF and the *** results indicated that lower prepolymer viscosity led to lower apparent density,while WF addition reduced even more the *** tests showed that the maximum compressive strengths were good,while WF improved the compressive strength by up to 56%.Scanning electron microscopy(SEM)showed uniform cell structures,but small open pores were ***-dimensional(2D)CT scan images confirmed the good compatibility between WF and the matrix,with low anisotropy in the *** tests indicated that WF decreased the pulverization ratio of the materials by up to 42%.Thermogravimetric analysis(TGA)showed good thermal stability of the materials up to 328°*** burning tests showed that the materials were self-extinguishing without residue(dripping).The lowest thermal conductivity was 0.04 W/m·*** results suggest that these novel formaldehyde-free,high biomass content(95%–96%)foams and composite foams have high potential to replace traditional phenolic foams(PF)in applications such as construction,transportation,packaging,and thermal insulation.

Chemotherapy outcomes for the treatment of glioma remains unsatisfactory due to the inefficient drug transport across the blood–brain barrier(BBB) and insufficient drug accumulation in the tumor region. Although many approaches, including various nanosystems, have been developed to promote the distribution of chemotherapeutics in the brain tumor, the delivery efficiency and the possible damage to the normal brain function still greatly restrict the clinical application of the ***, it is urgent and necessary to discover more safe and effective BBB penetration and gliomatargeting strategies. In the present study, menthol, one of the strongest BBB penetration enhancers screened from traditional Chinese medicine, was conjugated to casein, a natural food protein with brain targeting capability. Then the conjugate self-assembled into the nanoparticles to load anti-cancer *** nanoparticles were characterized to have appropriate size, spheroid shape and high loading drug capacity. Tumor spheroid penetration experiments demonstrated that penetration ability of mentholmodified casein nanoparticles(M-CA-NP) into the tumor were much deeper than that of unmodified nanoparticles. In vivo imaging further verified that M-CA-NPs exhibited higher brain tumor distribution than unmodified nanoparticles. The median survival time of glioma-bearing mice treated with HCPT-MCA-NPs was significantly prolonged than those treated with free HCPT or HCPT-CA-NPs. HE staining ofthe organs indicated the safety of the nanoparticles. Therefore, the study combined the advantages of traditional Chinese medicine strategy with modern delivery technology for brain targeting, and provide a safe and effective approach for glioma therapy.

Dynamic ultra-high pressure homogenization(UHPH)is a novel high-pressure processing *** this study,the effects of dynamic UHPH on the structure and functional properties of casein were systematically *** was found that the functional properties of casein changed with dynamic UHPH treatment,and the treatment at 150 MPa could significantly improve casein aqueous solubility,foaming and emulsifying *** functional improvements could be attributed to its structural changes,since the dynamic UHPH treatment could change the secondary structure,promote the interchange reaction between the disulfide bond and the sulfhydryl group,and increase the surface *** obtained results could broaden the application of casein and provide ideas for the non-thermal processing of proteins.

Expression and regulation of hFIX minigene and cDNA driven by β-casein gene in mouse mammary gland

Mammary gland specific expression vectors for human clotting factor IX (hFIX) and LacZ reporter gene driven by bovine β\|casein gene were constructed. Vectors were packaged by stearylamine (SA) liposome and were transferred to lactating mice via tail vein. Both hFIX and LacZ gene could be expressed in the mammary gland of the treated mice. The highest production of hFIX protein was 80.28 ng per mL milk, and more than 85% of hFIX protein appeared to be γ carboxylation and biologically active. The results suggested that the 2.0 kb sequence of β casein gene including promoter, exon 1 was effective to drive hFIX gene expression in mammary gland and intron 1 of β casein gene had an effect on the tissue specific expression. The expression level in mouse milk injected with hFIX minigene vector containing hFIX endogenous intron 1 was increased by above 3 times of that injected with hFIX cDNA vector.

Identification and evaluation of antioxidant activity of bioactive casein-derived peptides during in vitro digestion and transepithelial transport in Caco-2 cells

Dairy products are important sources of bioactive(antibacterial,antithrombotic,antihypertensive,immuno-modulatory,and antioxidative)peptides,that need to be released from the target *** the present study,bovine milk casein protein was hydrolyzed by pancreatin and trypsin(4 and 24 h)to evaluate the hydrolyses efficiency(ninhydrin reaction);to elucidate the sequence of casein-derived peptides(SDS-PAGE,LC-ESI-TOF-MS);and to assess the peptides antioxidant activity(DPPH and ORAC),bioactivity in silico,bioaccessibility after in vitro digestion,and bioavailablility by transepithelial transport in Caco-2 *** hydrolysis degree increased with the incubation time,with evident degradation of casein-derived proteins into low molecular weight peptides(casein,21 fromα-s1-casein,15 fromα-s2-casein,and 7 from k-casein *** of these peptides were described as antioxidant,anti-inflammatory,and *** in vitro digestion 123 peptides were found in gastric(56 peptides),duodenal(43 peptides)and colonic(24 peptides)fraction;while five peptides were bioavailable by crossing the intestinal barrier,with increased peptides’abundance after trypsin hydrolysis treatment.

In vitro and in silico analysis of dual-function peptides derived from casein hydrolysate

There is no study on food-derived peptide with both anticoagulant and angiotensin I-converting enzyme inhibitory (ACEI) activities yet. In this work, the anticoagulant and ACEI activities of the casein hydrolysates released by pepsin digestion were evaluated for the first time to the best of our knowledge. Results indicated that the casein hydrolysate exhibited potent anticoagulant activity by prolonging the thrombin time (TT) and the activated partial thromboplastin time (APTT). Compared with control samples, at 10 mg/mL, the TT and APTT of casein hydrolysate were 186.0 % ± 6.6 % and 163.5 % ± 7.4 %, respectively. The casein hydrolysate also showed a strong ACEI activity with an IC50 value of 1.775 mg/mL. The components of the bioactive casein hydrolysate were analyzed by nanoscale liquid chromatography quadrupole time-of-flight tandem mass spectrometry (NanoLC-Q-TOF-MS/MS). Total of 115 peptides were identified, among which 34, 9, 55 and 17 peptides were derived from α_(s1-), α_(s2-), β-, and κ-casein, respectively. The results of PeptideRanker and PepSite 2 analysis showed that 6 peptides (FRQFYQL, NENLLRF, NPWDQVKR, PVVVPPFLQ, PVRGPFPIIV, and ARHPHPHLSF) have both ACEI and anticoagulant activities by binding to the active sites of ACE and thrombin. This study indicated that casein is a potential functional food supplement that can be used for medical purposes.

Alginate-casein Microspheres as Bioactive Vehicles for Nutrients

The aim of this work was to develop an alginate-casein composite microsphere as a bioaetive vehicle for oral administration of nutrients by a simple extrusion dripping method. Riboflavin was selected as a model drug, and the microencapsulation efficiency was raised to 97.94% after optimizing the preparation conditions by response surface methodology. In vitro release studies showed that riboflavin was released completely from alginate-casein microspheres in simulated intestinal fluids. Meanwhile, the morphology, structure and interaction between alginate and casein were characterized by scanning electron microscopy （SEM） and Fourier transform infrared （FTIR） spectra.
