Correlation between Expression of P38 MAPK-Signaling and uPA in breast cancer

OBJECTIVE To study the expression of phosphorylated p38 mitogen-activated protein kinase （p-p38） and uPA and the correlation of their expression with breast cancer.clinicopathological characteristics, and to investigate the role of the p38MAPK-signaling pathway in regulating uPA expression in breast cancer.cells.METHODS Immunohistochemistry （S-P） was used to test the expression of p-p38 and uPA in 60 specimens of breast cancer.tissues. Western blots were adopted to detect expression of the p-p38 and uPA proteins in MDA-MB-231 and MCF-7 breast cancer.cells, and uPA expression after treatment with SB203580, a specific inhibitor of p38 MAPK.RESULTS The positive rate of the p-p38 protein and uPA protein expression in the breast cancer.tissues was 56.7% and 60.0%,respectively. The expression of p-p38 was positively related to the expression of uPA （r = 0.316, P 〈 0.05）. The expression of p-p38 and uPA was related to lymph node metastasis and the TNM stage （P 〈 0.05）, but it was not related to the patient＇s age or tumor size （P 〉 0.05）. The expression of p-p38 and uPA in MDA- MB-231 cells was higher than that in MCF-7 cells. SB203580 inhibited the p38 MAPK pathway and reduced uPA protein expression.CONCLUSION The p38 MAPK-signaling pathway promotes breast cancer.malignant progression by up-regulating uPA expression ,and it may be an important process in breast cancer.invasion and metastasis.

Facts and evidence hold out a steadily increasing prevalence of breast cancer.in the mid-1940s. In most cases, the disease is discovered when the disease has reached an advanced stage, therefore, early diagnosis can significantly reduce the burden of disease. breast cancer.has some risk factors, such as tumor size and lymph node involvement. The aim of this study was to evaluate the correlation between tumor size and axillary lymph node involvement and the prognosis for patients with breast cancer.has been surgery. The study population consisted of 100 patients with breast cancer.surgery at Shohada Hospital of Tajrish during the last 10 years. The results suggest that lymph node involvement was significantly associated with prognosis in patients with breast cancer.and in cases where the tumor size was less than 1 cm, it has no impact on the prognosis of breast cancer. Also, a family history of the disease among close relatives is an independent risk factor for poor prognosis, in patients with breast cancer.in general it can be said that involvement of lymph nodes in patients with breast cancer.is due to an unfavorable prognosis; but the number of lymph nodes involved and the size of the tumor did not have any effect on the prognosis.

CSCs （cancer.stem cells） have been involved in tumor resistance, metastasis and recurrence. In breast cancer. tumor cells are characterized by CD44＋, CD24-/low and ALDH 1 expression represents a subpopulation of BCSC （breast cancer.stem cell）. Several three-dimensional （3D） in vitro culturing cancer.cells have been used to stimulate BCSC phenotype. The present study aimed to evaluate 3D cell culture in alginate matrix and the CD44, CD24 and ALDH1 mRNA levels of BCSC markers. The 3D culture was performed using MDA-MB-231 breast cancer.cell line on alginate matrix 1.2% in RPMI medium. Expression of BCSC markers was evaluated by Real Time PCR （Polymerase Chain Reaction） comparing 3D to 2D culture. The 3D cultures increase of CD44 and CD24 mRNA levels and induce ALDH1 expression comparing to 2D culture. The data suggest that 3D alginate matrix alters the mRNA levels of genes involved in the phenotypic characteristics of BCSC.

RASSFIA Promoter Hypermethylation as a Prognosis and Diagnosis for breast cancer.in Vietnamese Population

Aberrant tumor suppressor gene promoter methylation was associated with the several cancer., including breast cancer. which was the common female deaths in most countries involved in Vietnam. The methylation in tumor suppressor genes, including RASSFIA, were the key targets of establishing the potential biomarkers for prognosis and early diagnosis of breast cancer. In present study, with the aim towards using the hypermethylation at CpG islands of promoter of RASSFIA as the biomarker for breast cancer.in Vietnamese population, MSP （methyl specific PCR） was carried out to analyze the hypermethylation status ofRASSFIA gene in 115 samples including 95 breast cancer.specimens and 20 normal breast tissues from another disease （not breast cancer.. All samples were obtained from Ho Chi Minh City Medical Hospital, Vietnam. The known predictive and prognostic factors： HER2/neu overexpression was immunohistochemistry stained as input value for breast cancer.specimens. For input value confirmed, the overexpression of p53 protein was also analyzed together with prior immunochemical assay. The results indicated that the hypermethylation of frequencies for methylation of given gene reached 42.1% （P 〈 0.05）. In addition, the DNA hypermethylation of RASSFIA gene increased the possibility to be breast cancer.with high incidence via calculated of odd ratio （P 〈 0.05）. In conclusion, the hypermethylation of candidate genes could be used as the promising biomarkers applying in Vietnamese breast cancer.patients.

Although metastatic breast cancer.is considered as an incurable disease, various biological drivers influence the outcomes. The use of trastuzumab in patients overexpressing HER（human epidermal growth factor receptor 2）-2 increases long-term survival even in those patients who developed brain metastasis. Nevertheless, special attention must be paid to the risk of cardiotoxicity. We report the case of a young woman with HER-2-positive breast cancer.with bone and lung disease who developed brain metastasis during treatment with trastuzumab. The treatment has been continued and she is alive and in complete remission after 16 years.

breast cancer.is one of the world＇s most urgent health problems. The first symptoms of mammary malignancies are manifested only at an advanced stage with significant mortality. Detecting this disease at an early stage gives the majority of patients a better chance of survival. The aim was to search for changes in gene expression of specific tumor vascular markers like death receptor 6 （Dr6） and glycoprotein M6B （Gpm6B） in the blood of patients with breast cancer. All subjects were divided into two groups. First group with patients are with different grades of breats tumors （n = 30）. Second group consists from healthy women （n = 15）. After isolation of mRNA from blood, RT-PCR was followed by gel electrophoresis. For statistical analysis one-way ANOVA was used with Student＇s T test using GraphPad InStat software. Significant changes in mRNA levels of gene Dr6 in all grades of first stage breast cancer.were detected. The mRNA levels of Dr6 showed a rising tendency from GI （116% higher value than control） to G3 （198% higher than control）. During monitoring of the mRNA level of Gpm6B, a weaker increase was observed than in Dr6. The difference in GI was only 8% higher compared with controls and 44% at G3. From our results it can be concluded that DR6 is a more suitable marker for the diagnosis of breast malignancies in the early stages than Gpm6B. In our work, a non-invasive method for more timely and precise determination of the earlier stages of breast cancer.is described, which could also contribute to monitoring the effectiveness of treatment, or regression of this disease.
