PD-L1^(+)exosomes from bone marrow-derived cells of tumor-bearing mice inhibit antitumor immunity

Pre-degenerated peripheral nerves co-cultured with bone marrow-derived cells: a new technique for harvesting high-purity Schwann cells

Schwann cells play an important role in the peripheral nervous system, especially in nerve repair following injury, so artificial nerve regen- eration requires an effective technique for obtaining purified Schwann cells. In vivo and in vitro pre-degeneration of peripheral nerves have been shown to obtain high-purity Schwann cells. We believed that in vitro pre-degeneration was simple and controllable, and available for the clinic. Thus, we co-cultured the crushed sciatic nerves with bone marrow-derived cells in vitro. Results demonstrated that, 3 hours after injury, a large number of mononuclear cells moved to the crushed nerves and a large number of bone marrow-derived cells infiltrated the nerve segments. These changes promoted the degradation of the nerve segments, and the dedifferentiation and proliferation of Schwann cells. Neural cell adhesion molecule and glial fibrillary acidic protein expression were detected in the crushed nerves. Schwann cell yield was 9.08 ± 2.01 ×104/mg. The purity of primary cultured Schwann cells was 88.4 ± 5.79%. These indicate a successful new method for ob- taining Schwann cells of high purity and yield from adult crushed sciatic nerve using bone marrow-derived cells.

LXHY Formula Inhibits Choroidal Neovascularization Development via Inhibiting the Recruitment and Adhesion of BMCs to the Retina

Objective:The objective of this study is to investigate the inhibitory effect of LXHY,a Chinese medicine compound formula,on choroidal neovascularization(CNV) and to find the possible working ***:CNV was induced in C57 BL/6 mice by krypton laser and bone marrow-derived cells(BMCs) isolated from enhanced green fluorescent protein(EGFP) transgenic mice were injected through tail vein 0.5–1 h after the laser *** BMC-treated mice were randomly divided into two groups gavaged with either distilled water(DW group) or LXHY formula solution from day 1 after laser *** days 7,14,and 28 after treatment,histopathologic examination,fundus fluorescein angiography,and choroidal flatmount assay were performed to measure the CNV severity and BMC ***4 levels in peripheral blood were measured by enzyme-linked immunosorbent *** cell-derived factor-1α(SDF?1α),vascular cell adhesion molecule-1(VCAM-1),and intercellular adhesion molecule-1(ICAM-1) were detected by immunofluorescent ***:On days 7 and 14 after treatment,CNV lesions in the LXHY-treated mice showed less recruitment of BMCs and were smaller in size compared to DW-treated *** examination also confirmed less severe CNV lesions in the LXHY ***4 levels in peripheral blood in the LXHY group were less than that of DW group on days 7 and ***,the expression levels of SDF-1α,ICAM?1,and VCAM?1 at the lesion sites in the LXHY group were lower compared with the DW ***:This experiment indicated that LXHY formula could inhibit CNV formation and development,probably by inhibiting the recruitment and attachment of BMCs into CNV area.

Autologous bone marrow stem cell transplantation in critical limb ischemia： a meta-analysis of randomized controlled trials

Background Amputation-free survival （AFS） has been recommended as the gold standard for evaluating No-Option Critical Limb Ischemia （NO-CLI） therapy. Early-phase clinical trials suggest that autologous bone-marrow derived cells （BMCs） transplantation may have a positive effect on patients with NO-CLI, especially decreasing the incidence of amputation. However, the BMCs therapeutic efficacy remains controversial and whether BMCs therapy is suitable for all CLI patients is unclear. Methods We conducted a meta-analysis using data from randomized controlled trials （RCTs） by comparing autologous BMCs therapy with controls in patients with critical limb ischemia, and the primary endpoint is the incidence of amputation. Pubmed, EBSCO and the Cochrane Central Register of Controlled Trials （to approximately July 25, 2012） were searched. Results Seven RCTs with 373 patients were enrolled in the meta-analysis. Because serious disease was the main reason leading to amputation in one trial, six studies with 333 patients were finally included in the meta-analysis. Pooling the data of the final six studies, we found that BMCs therapy significantly decreased the incidence of amputation in patients with CLI （odds ratio （OR）, 0.37; 95% confidence interval （CI）, 0.22 to 0.62; P=-0.0002）, and the efficacy had not significantly declined within 6 months after BMCs were transplanted; OR, 0.33; 95% CI, 0.16 to 0.70; P=0.004 within 6 months and OR, 0.30; 95% CI, 0.11 to 0.79; P=0.01 within 3 months. The rate of AFS after BMCs therapy was significantly increased in patients with Rutherford class 5 CLI （OR 3.28; 95% CI, 1.12 to 9.65; P=0.03）, while there was no significant improvement in patients with Rutherford class 4 （OR 0.35; 95% CI, 0.05 to 2.33; P=0.28） compared with controls. The BMCs therapy also improved ulcer healing （OR, 5.83; 95% CI, 2.37 to 14.29; P=-0.0001）.
