Polydopamine-modified chitin conduits with sustained release of bioactive peptides enhance peripheral nerve regeneration in rats

Potential antioxidant bioactive peptides from camel milk proteins

Camel milk is traditionally considered to have medicinal characteristics that it has potential health benefits and could help to treat several illnesses. Particularly, it is closest to human breast milk and has high levels of nutrients and bioactive components. The aim of this study was to explore the antioxidant peptides derived from protein fractions of camel milk. Camel milk proteins(CMP) were fractionated into camel casein protein(CCP) and camel whey protein(CWP), which were hydrolyzed with pepsin to produce peptic digests P-CCP and P-CWP, respectively. RP-HPLC was used for fractionation of the peptides from the P-CCP and P-CWP. The antioxidant activities were evaluated using superoxide anion generating system of xanthine oxidase(XOD)and 2,2-diphenyl-1-picrylhydrazyl(DPPH) scavenging assay. Active peptides were analyzed using matrixassisted laser desorption ionization time of flight mass spectrometry(MALDI-TOF-MS) whereas a number of antioxidant peptides, with masses ranging from 913 to 2,951 Da, derived mainly from alpha-casein, lactophorin and lactoferrin, were identified. When yeast cells are used as a system for modeling mitochondrial disease, the peptides in caseins and whey fractions significantly enhanced the tolerance of yeast cells against peroxide-induced oxidative stress. The results show that both caseins and whey proteins of camel milk possess bioactive peptides with significant radical-scavenging activities and thus herald a fascinating opportunity for their potential as nutraceuticals or therapeutic peptides for prevention and treatment of oxidative stress-associated diseases.

Identification and evaluation of antioxidant activity of bioactive casein-derived peptides during in vitro digestion and transepithelial transport in Caco-2 cells

Dairy products are important sources of bioactive(antibacterial,antithrombotic,antihypertensive,immuno-modulatory,and antioxidative)peptides,that need to be released from the target *** the present study,bovine milk casein protein was hydrolyzed by pancreatin and trypsin(4 and 24 h)to evaluate the hydrolyses efficiency(ninhydrin reaction);to elucidate the sequence of casein-derived peptides(SDS-PAGE,LC-ESI-TOF-MS);and to assess the peptides antioxidant activity(DPPH and ORAC),bioactivity in silico,bioaccessibility after in vitro digestion,and bioavailablility by transepithelial transport in Caco-2 *** hydrolysis degree increased with the incubation time,with evident degradation of casein-derived proteins into low molecular weight peptides(peptides(47%reduction of free radicals;32112μM Trolox equivalents)than pancreatin derived peptides(15%reduction;2862μM Trolox equivalents).A total of 69 ca-seins derived peptides were identified after trypsin(33 peptides)and pancreatin(36 peptides)hydrolysis for 24 h;with 26 peptides released fromβ-casein,21 fromα-s1-casein,15 fromα-s2-casein,and 7 from k-casein *** of these peptides were described as antioxidant,anti-inflammatory,and *** in vitro digestion 123 peptides were found in gastric(56 peptides),duodenal(43 peptides)and colonic(24 peptides)fraction;while five peptides were bioavailable by crossing the intestinal barrier,with increased peptides’abundance after trypsin hydrolysis treatment.

Systematic functional analysis and potential application of a serine protease from cold-adapted Planococcus bacterium

In this study, a gene encoding serine protease(PmSpr288)from cold-adapted bacterium, namely Planococcus maritimus XJ11, was cloned and overexpressed in Escherichia coli BL21(DE3). Bioinformatics analysis revealed that PmSpr288 belongs to serine protease S8 superfamily with a classical catalytic triad comprised by the Asp49, His86 and Ser251. Moreover, PmSpr288 was found to be active over broad alkaline pH and low-moderate temperature, and exhibited wide range of protein substrate specificity. In addition, PmSpr288 was able to hydrolyze the meat proteins actin and myosin, and molecular docking results suggested that the crucial interaction between PmSpr288 and actin/myosin complexes was mainly occupied by hydrogen bonds. The muscle protein hydrolysates of silver carp prepared by PmSpr288 was shown to have antioxidant activity via DPPH radical scavenging assay, which presented an IC_(50) valve of 1.309 mg/mL. In conclusion, these characteristics imply that PmSpr288 has potential biotechnological application prospect for the production of bioactive peptides.
