To date,the overall response rate of PD-1 blockade remains unsatisfactory,partially due to limited understanding of tumor immune microenvironment(TIME).B-cell lymphoma 9(bcl9),a key transcription co-activator of the Wnt pathway,is highly expressed in cancers.By genetic depletion and pharmacological inhibition of bcl9 in tumors,we found that bcl9 suppression reduced tumor growth.

利用密度泛函B3LYP方法与耦合簇CCSD方法，结合全电子高斯基函数，计算了;bcl;和;bcl;分子的基态平衡几何和谐振频率，进而得到了分子伸缩振动模式的同位素位移.计算检验了芯-价电子关联、标量相对论效应，以及基组外推对分子几何结构、振动频率和同位素位移的影响;结果表明，高阶修正效应对同位素位移的影响很小，并分析了其原因.同时得到了不同Cl/B同位素分子体系的同位素位移.研究对激光分离同位素技术中激光中心波长的选择具有重要参考意义.

Spatio-temporate Expression of P53, bcl-2 and Fas in the Retina of RD, Rds and C3H Mice

PURPOSE: To investigate the expression status of p53, Fas and bcl-2 in the development of retinal degeneration in C3H, rd and rds mice. METHODS: Expression of p53, Fas and bcl-2 in the retina of rd, rds and C3H mice as well as in normal C3B mice in different periods was examined by immunohistochemical technique. RESULTS: The expression of Fas and p53 was not detected in normal C3B mice. Fas and p53 were expressed in the ganglionic layer in the early stage and then in the inner nuclear layer, while the scale and intense increased in the ganglionic layer. There is no difference of bcl-2 expression between the normal mice and mice with retinal degeneration. CONCLUSION: Fas and p53 may be involved in the retinal cell death in rd, rds, and C3B, but bcl-2 may not. It is unknown why p53 and Fas appeared first in ganglion layer but not in the outer nuclear layer where retinal cells death was noticed in early stage.

bcl-2 Small Interfering RNA Inhibits the Growth of Human Lymphoma Transplanted Subcutaneously in Nude Mice

OBJECTIVE To investigate whether small hairpin RNA (shRNA)targeting bcl-2 mRNA could inhibit the growth of lymphomatransplanted subcutaneously in nude mice.METHODS Recombinant bcl-2 shRNA expression vector withgreen fluorescence protein (GFP) gene was constructed andpreserved in our lab. We evaluated the antitumor effect of the bcl-2shRNA in vivo which was the model of nude mice bearing Rajicells xenografts. Human Raji cells were injected subcutaneouslyinto nude mice to establish lymphoma models. When thediameters of tumor were above 0.5 cm after Raji cells injection,the mice bearing tumor were randomly divided into four groups:saline control group, negative shRNA group, plasmid vectorgroup, bcl-2 shRNA group. The polyethylenimine (PEI) was usedto transfect shRNA into tumor. The mixed PEI and shRNA wasinjected into tumors. The growth and size of tumor were observed.Tissue was stained by H&E for its pathological morphology. Theexpression of bcl-2 mRNA in the tumor mass was detected byreverse transcription polymerase chain reaction (RT-PCR).RESULTS A significant difference in median tumor weightwas observed in mice treated with bcl-2 shRNA, compared withthose in the groups of negative shRNA or plasmid vector or salinesolution (Pbcl-2 shRNA showedapoptosis, serious necrosis of the cells and inflammatory cellsinfiltration. There was no change in the morphology of cellsamong negative shRNA, plasmid vector and saline solution group.In the group of the bcl-2 shRNA, the expression levels of bcl-2mRNA of the tumor tissue were effectively inhibited (P bcl-2 mRNAcould inhibit the growth of human lymphoma transplantedsubcutaneously in nude mice.

目的　本文研究了bcl 10基因在早期和进展期人肝细胞肝癌中的突变频率。方法　提取 4 6例新鲜肝癌及癌周组织的基因组DNA ,应用聚合酶链式反应单链构象多态性分析的方法 ,研究bcl 10基因 3个外显子的突变情况 ,并对每个外显子随即抽取 6例突变样本进行测序分析。同时分析bcl 10基因突变与血清甲胎蛋白水平以及肝癌大小的关系。结果　 4 6例肝细胞肝癌中共检测到外显子 1突变 2 6例 ,占 5 6 5 % ,6例测序样本中有 5例表现为第 5 74 4位CG颠换突变 ;外显子 2突变 2 5例 ,占 5 4 3% ,6例测序样本中有 4例表现为第 11311位T碱基缺失突变 ;外显子 3突变 2 1例 ,占 4 5 7% ,6例测序样本皆表现为系第 14116位CT转换突变。统计分析表明bcl 10突变和血清甲胎蛋白水平以及肝癌大小没有相关关系。结论　bcl 10基因在肝癌中突变频率较高。

实验用家兔共71只。乌拉坦浅麻醉,麻痹肌肉,颈_1或颈_2 横断脊髓,记录膈神经电活动。43例中,25例出现紧张性电活动,无一例自行恢复呼吸样节律放电。24例于脊髓蛛网膜下腔(i.s.s.)内给荷包牡丹碱(bcl,20μg/10μl—40μg/20μl i.s.s.),13例给印防已毒素(PIC,20μg/20μl,i.s.s.或3—5mg/kgbt i.v.),能诱发出长串的呼吸样放电(串长在0.25s以上)或短串的抽搐样放电(串长在0.1s 以下)。长串放电,按其募集与去募集特点,可分为三型: Ⅰ型,排放频率平均为23.5±2.3串/min(bcl),出现率为58.5%(bcl)或67.7%,(PIC);Ⅱ型,排放频率平均为33.8±4.7串/min(bcl),出现率为39.3%(bcl)或32.3%(PIC);Ⅱ型,排放频率平均为21.3±2.8串/min(bcl),出现率为2.2%(bcl)或3.3%(PIC)。放电持续时间平均为60.0±18.9min(bcl)或42.0±0.8min(PIG)。Ⅰ型和Ⅱ型放电均具呼吸样放电的特点,Ⅰ型和Ⅱ型的出现率共占长串放电的97.8%。本工作提示:脊髓內源性 GABA 系统对脊髓“呼吸”具有紧张性抑制作用。
