A batch microfabrication of a self-cleaning,ultradurable electrochemical sensor employing a BDD film for the online monitoring of free chlorine in tap water

Free chlorine is one of the key water quality parameters in tap water.However,a free chlorine sensor with the characteristics of batch processing,durability,antibiofouling/antiorganic passivation and in situ monitoring of free chlorine in tap water continues to be a challenging issue.In this paper,a novel silicon-based electrochemical sensor for free chlorine that can self-clean and be mass produced via microfabrication technique/MEMS(Micro-Electro-Mechanical System)is proposed.A liquid-conjugated Ag/AgCI reference electrode is fabricated,and electrochemically stable BDD/Pt is employed as the working/counter electrode to verify the effectiveness of the as-fabricated sensor for free chlorine detection.The sensor demonstrates an acceptable limit of detection(0.056 mg/L)and desirable linearity(R^(2)=0.998).Particularly,at a potential of+2.5 V,hydroxyl radicals are generated on the BBD electrode by electrolyzing water,which then remove the organic matter attached to the surface of the sensor though an electrochemical digestion process.The performance of the fouled sensor recovers from 50.2 to 94.1%compared with the initial state after self-cleaning for 30 min.In addition,by employing the MEMS technique,favorable response consistency and high reproducibility(RSD<4.05%)are observed,offering the opportunity to mass produce the proposed sensor in the future.A desirable linear dependency between the pH,temperature,and flow rate and the detection of free chlorine is observed,ensuring the accuracy of the sensor with any hydrologic parameter.The interesting sensing and selfcleaning behavior of the as-proposed sensor indicate that this study of the mass production of free chlorine sensors by MEMS is successful in developing a competitive device for the online monitoring of free chlorine in tap water.

Flexible intracerebral probes for neural recording and electrical stimulation have been the focus of many research works to achieve better compliance with the surrounding tissue while minimizing rejection.Strategies have been explored to find the best way to insert flexible probes into the brain while maintaining their flexibility once positioned.Here,we present a novel and versatile scalable batch fabrication approach to deliver ultrathin and flexible probes consisting of a silk-parylene bilayer.The biodegradable silk layer,whose degradation time is programmable,provides a temporary and programmable stiffener to allow the insertion of ultrathin parylene-based flexible devices.Our innovative and robust batch fabrication technology allows complete freedom over probe design in terms of materials,size,shape,and thickness.We demonstrate successful ex vivo insertion of the probe with acute high-fidelity recordings of epileptic seizures in field potentials as well as single-unit action potentials in mouse brain slices.Our novel technological solution for implanting ultraflexible devices in the brain while minimizing rejection risks shows high potential for use in both brain research and clinical therapies.

Two types of inoculated sludges, granular sludge that had been stored at-20°C and activated sludge, were investigated for the domestication of aerobic granular sludges(AGSs in sequencing batch reactors(SBRs). The results showed that using the stored granular sludge as inoculation sludge could effectively shorten the domestication time of AGS and yielded mature granular sludge after 22 days of operation. The AGS domesticated by stored granular sludge had better biomass and sedimentation properties; its MLSS and SVI reached8.55 g/L and 35.27 mL/g, respectively. The removal efficiencies for chemical oxygen demand(COD), ammonium nitrogen(NH_4^+-N) and total phosphorus(TP) reached 90.76%, 97.39% and 96.40%, respectively. By contrast, 54 days were needed to obtain mature granules using activated sludge. The microbial community structure was probed by using scanning electron microscopy(SEM) and high-throughput sequencing. The results showed that the diversity of the microbial community in mature granules was reduced when stored granular sludge rather than activated sludge was employed as inoculation sludge, and the dominant microbes were changed. The dominant species in mature granules domesticated using stored granular sludge were Zoogloea, Acidovorax and Tolumonas at the genus classification level, while the dominant species were Zoogloea and TM7-genera in granules developed from activated sludge.

Hybrid growth microorganisms in sequencing batch reactors have proven effective for treating the toxic compound phenol, but the toxicity effect under different toxicity conditions has rarely been discussed. Therefore, the performance of the HG-SBR under toxic, acute and chronic organic loading can provide the overall operating conditions of the system. Toxic organic loading(TOL) was monitored during the first 7 hr while introducing50 mg/L phenol to the system. The system was adversely affected with the sudden introduction of phenol to the virgin activated sludge, which caused a low degradation rate and high dissolved oxygen consumption during TOL. Acute organic loading(AOL) had significant effects at high phenol concentrations(600, 800 1000 mg/L). The specific oxygen uptake rate(SOUR) gradually decreased to 4.9 mg O_2/(g MLVSS·hr) at 1000 mg/L of phenol compared to 12.74 mg O_2/(g MLVSS·hr) for 200 mg/L of phenol. The HG-SBR was further monitored during chronic organic loading(COL) over 67 days. The effects of organic loading were more apparent at 800 mg/L and 1000 mg/L phenol concentrations, as the removal range was between 22%–30% and 18%–46% respectively, which indicated the severe effects of COL.

In many batch processes, there are related or independence relationships among process variables. The traditional monitoring method usually carries out a single statistical model according to the related or independent method, and in the feature extraction there is not fully taken into account the characterization of fault information, it will make the process monitoring ineffective, so a fault monitoring method based on WGNPE(weighted global neighborhood preserving embedding)–GSVDD(greedy support vector data description) related and independent variables is proposed. First, mutual information method is used to separate the related variables and independent variables. Secondly, WGNPE method is used to extract the local and global structures of the related variables in batch process and highlight the fault information, GSVDD method is used to extract the process information of the independent variables quickly and effectively. Finally, the statistical monitoring model is established to achieve process monitoring based on WGNPE and GSVDD. The effectiveness of the proposed method was verified by the penicillin fermentation process.

batch effects are technical sources of variation and can confound analysis.While many performance ranking exercises have been conducted to establish the best batch effect-correction algorithm(BECA),we hold the viewpoint that the notion of best is context-dependent.Moreover,alternative questions beyond the simplistic notion of "best" are also interesting:are BECAs robust against various degrees of confounding and if so,what is the limit?Using two different methods for simulating class(phenotype) and batch effects and taking various representative datasets across both genomics(RNA-Seq) and proteomics platforms,we demonstrate that under situations where sample classes and batch factors are moderately confounded,most BECAs are remarkably robust and only weakly affected by upstream normalization procedures.This observation is consistently supported across the multitude of test datasets.BECAs do have limits:When sample classes and batch factors are strongly confounded,BECA performance declines,with variable performance in precision,recall and also batch correction.We also report that while conventional normalization methods have minimal impact on batch effect correction,they do not affect downstream statistical feature selection,and in strongly confounded scenarios,may even outperform BECAs.In other words,removing batch effects is no guarantee of optimal functional analysis.Overall,this study suggests that simplistic performance ranking exercises are quite trivial,and all BECAs are compromises in some context or another.
