The emergency of anthocyanin biosynthetic pathway has played vital roles in plant development,MYB regulators involved has been reported to arise early during land plant evolution.After the division of monocots and dicots,they might function divergently suffered from diverse selection pressures.Divergent regulation mechanisms were found in dicot and monocot plants represented by Arabidopsis thaliana and Zea mays,respectively.So far,amounts of anthocyanin-promoting R2 R3-MYBs have been identified and well elucidated in core dicot plants,whereas they were studied randomly in monocot plants,which are of crucial importance to decipher functional diversities among angiosperms during evolution.In this study,one R2 R3-MYB gene FhPAP1 leading to the accumulation of anthocyanin was isolated from flowers of one monocot plant,Freesia hybrida.Temporal and spatial expression profiles revealed FhPAP1 showed positive correlations with anthocyanin biosynthetic genes especially the late biosynthetic genes.Further analysis indicated that it could form sets of MBW（MYB-bHLH-WD40） complexes with FhTT8 L,FhGL3 L and FhTTG1 L.Gain-of-function experiments in Arabidopsis and Nicotiana and transient protoplasts transfection assays in Freesia further proved that FhPAP1 was sufficient to interacted with the bHLH proteins to regulate the expression of anthocyanin biosynthetic genes.Moreover,other partners in the MBW complex,i.e.,WD40 regulators,could strengthen the transcription activation efficiency.However,FhPAP1 showed higher transactivation property when compared with AtPAP1 and NtAN2 from dicot plants Arabidopsis and tobacco,which resulted in different increments of target gene expression levels.Therefore,it can be deduced that the MYB regulator from Freesia hybrida might have similar target genes and function conservatively compared with core eudicot plants,but its significant transactivation property will privilege it in crop improvement and also shed light on the mechanisms of anthocyanin-promoting MYB regulators in angiosperms during

There has been increasing interest in anthocyanins due to anthocyanins potential nutritionalbenefits. Although the basic reactions of the anthocyanin biosynthetic pathways inplants have been analysed intensively, the regulation of their accumulation and fluxthrough the pathway has not been particularly well established. The aim of this study wasto use Arabidopsis thaliana overexpressing the radish peroxidase gene RsPRX1 to investigategene expression patterns associated with the accumulation of anthocyanins in Arabidopsisseedlings. Plants exposed to white light and a set of metabolic sugars(sucrose,glucose and fructose) showed an increase in anthocyanins content and number of compoundsin RsPRX1-overexpressing lines compared to the wild-type line when they weretreated with sucrose. There is superimposing effective in sucrose+glucose+fructose treatmentgroup compared with sucrose or glucose+fructose treatment groups. Peroxidaseactivity is ten times higher in transgenic Arabidopsis than in wild-type plants. Overexpressionof RsPRX1 strongly stimulates expression of anthocyanin regulatory genes(MYB75and bHLH(TT8)) and anthocyanin structural genes(CHS(chalcone synthase), LDOX(leucoanthocyanidin dioxygenase), DFR(dihydroflavonol 4-reductase)), and in the downregulatedexpression of endogenous FLS(flavonol synthase) and ANR(anthocyanidin reductase),but does not affect expression of the gene encoding sucrose transporter-1. Twobranches in the anthocyanin biosynthesis pathway have been proposed: FLS catalyses thereaction of dihydroquercetin to flavonols, and ANR catalyses the reaction of cyanidin toprocyanidin oligomers. It was concluded that a specific member of the peroxidase familyhas a role in regulating anthocyanin biosynthesis in response to sugar.
