Effects of mesenchymal stem cell on dopaminergic neurons,motor and memory functions in animal models of Parkinson's disease:a systematic review and meta-analysis

Parkinson’s disease is chara cterized by the loss of dopaminergic neurons in the substantia nigra pars com pacta,and although restoring striatal dopamine levels may improve symptoms,no treatment can cure or reve rse the disease itself.Stem cell therapy has a regenerative effect and is being actively studied as a candidate for the treatment of Parkinson’s disease.Mesenchymal stem cells are considered a promising option due to fewer ethical concerns,a lower risk of immune rejection,and a lower risk of teratogenicity.We performed a meta-analysis to evaluate the therapeutic effects of mesenchymal stem cells and their derivatives on motor function,memory,and preservation of dopamine rgic neurons in a Parkinson’s disease animal model.We searched bibliographic databases(PubMed/MEDLINE,Embase,CENTRAL,Scopus,and Web of Science)to identify articles and included only pee r-reviewed in vivo interve ntional animal studies published in any language through J une 28,2023.The study utilized the random-effect model to estimate the 95%confidence intervals(CI)of the standard mean differences(SMD)between the treatment and control groups.We use the systematic review center for laboratory animal expe rimentation’s risk of bias tool and the collaborative approach to meta-analysis and review of animal studies checklist for study quality assessment.A total of 33studies with data from 840 Parkinson’s disease model animals were included in the meta-analysis.Treatment with mesenchymal stem cells significantly improved motor function as assessed by the amphetamine-induced rotational test.Among the stem cell types,the bone marrow MSCs with neurotrophic factor group showed la rgest effect size(SMD[95%CI]=-6.21[-9.50 to-2.93],P=0.0001,I^(2)=0.0%).The stem cell treatment group had significantly more tyrosine hydroxylase positive dopamine rgic neurons in the striatum([95%CI]=1.04[0.59 to 1.49],P=0.0001,I^(2)=65.1%)and substantia nigra(SMD[95%CI]=1.38[0.89 to 1.87],P=0.0001,I^(2)=75.3%),indicating a protective effect on dopaminergic neurons.Subgro

Background： Regenerative techniques help promote the formation of new attachment and bone filling in periodontal detects. However, the dimensions of intraosseous defects are a key determinant of periodontal regeneration outcomes. In this study, we evaluated the efficacy of use of anorganic bovine bone （ABB） graft in combination with collagen membrane （CM）, to facilitate healing ofnoncontained （1-wall） and contained （3-wall） critical size periodontal defects. Methods： The study began on March 2013, and was completed on May 2014. One-wall （7 mm × 4 mm） and 3-wall （5 mm × 4 mm） intrabony periodontal defects were surgically created bilaterally in the mandibular third premolars and first molars in eight beagles. The defects were treated with ABB in combination with CM （ABB ＋ CM group） or open flap debridement （OFD group）. The animals were euthanized at 8-week postsurgery for histological analysis. Two independent Student＇s t-tests （1-wall [ABB ＋ CM] vs. 1-wall [OFD] and 3-wall [ABB ＋ CM] vs. 3-wall [OFD]） were used to assess between-group differences. Results： The mean new bone height in both 1- and 3-wall intrabony defects in the ABB ＋ CM group was significantly greater than that in the OFD group （1-wall： 4.99± 0.70 mm vs. 3.01 ± 0.37 mm, P 〈 0.05; 3-wall： 3.11±0.59 mm vs. 2.08± 0.24 mm, P 〈 0.05）. The mean new cementum in 1-wall intrabony defects in the ABB ＋ CM group was significantly greater than that in their counterparts in the OFD group （5.08 ±0.68 mm vs. 1.16 ± 0.38 mm; P 〈 0.05）. Likewise, only the 1-wall intrabony defect model showed a significant difference with respect to junctional epithelium between ABB ＋ CM and OFD groups （0.67 ±0.23 mm vs. 1.12 ± 0.28 mm, P 〈 0.05）. Conclusions： One-wall intrabony defects treated with ABB and CM did not show less periodontal regeneration than that in 3-wall intrabony defect. The noncontained l-wall intrabony defect might be a more discriminative defect model for further research into periodontal regeneration.

Impact of catheter-tissue contact force on lesion size during right ventricular outflow tract ablation in a swine model

Background:The catheter-tissue contact force(CF)is one of the significant determinants of lesion size and thus has a considerable impact on the effectiveness of ablation procedures.This study aimed to evaluate the impact of CF on the lesion size during right ventricular outflow tract(RVOT)ablation in a swine model.Methods:Twelve Guangxi Bama miniature male pigs weighing 40 to 50 kg were studied.After general anesthesia,a ThermoCool SmartTouch contact-sensing ablation catheter was introduced to the RVOT via the femoral vein under the guidance of the CARTO 3 system.The local ventricular voltage amplitude and impedance were measured using different CF levels.We randomly divided the animals into the following four groups according to the different CF levels:group A(3–9 g);group B(10–19 g);group C(20–29 g);and group D(30–39 g).Radiofrequency ablations were performed at three points in the free wall and septum of the RVOT in power control mode at 30 W for 30 s while maintaining the saline irrigation rate at 17 mL/min.At the end of the procedures,the maximum depth,surface diameter,and lesion volume were measured and recorded.A linear regression analysis was performed to determine the relationship between continuous variables.Results:A total of 72 ablation lesions were created in the RVOT of the 12 Bama pigs.The maximum depth,surface diameter,and volume of the lesions measured were well correlated with the CF(free wall:b=0.105,b=0.162,b=3.355,respectively,P<0.001;septum:b=0.093,b=0.150,b=3.712,respectively,P<0.001).The regional ventricular bipolar voltage amplitude,unipolar voltage amplitude,and impedance were weakly positively associated with the CF(b=0.065,b=0.125,and b=1.054,respectively,P<0.001).There was a significant difference in the incidence of steam pops among groups A,B,C,and D(free wall:F=7.3,P=0.032;septum:F=10.5,P=0.009);and steam pops occurred only when the CF exceeded 20 g.Trans-mural lesions were observed when the CF exceeded 10 g in the free wall,while the lesions in the septum were non-trans-mural even

Experimental study on the mechanism of sex difference in the risk of torsade de pointes

Background Torsade de pointes (TdP) is a form of polymorphic ventricular tachycardia featuring prolonged QT intervals. Female gender is associated with an increased risk of TdP. However, the causes of the sex difference in risk are poorly understood. Recently, transmural dispersion of repolarization (TDR) has been implicated in the genesis of TdP. Consequently, we compared TdP incidence and TDR between male and female rabbit hearts in order to investigate the mechanism of sex difference in TdP risk in rabbits in vitro. Methods By means of monophasic action potential recording techniques, the monophasic action potential of the epicardium, midmyocardium, and endocardium were simultaneously recorded using specially designed plunge-needle electrodes placed across the left ventricular free wall of both female (n=8) and male (n=8) rabbit hearts purfused by the Langendorff method. TdP was induced by bradycardia, d-sotalol, and low-K +, Mg 2+ Tyrode solution. Results TDR measurements in all three myocardial layers of male and female rabbit hearts were (18±2) ms and (21±2) ms, respectively (n=8, P>0.05). After perfusion with d-sotalol, the 90% monophasic action potential duration was prolonged in both male and femlae rabbits. TDR in male and female rabbit hearts increased to (29±2) ms and (61±2) ms, respectively, a difference that is significant. Eight female rabbit hearts had early afterdepolarization and 7 of them developed TdP. Seven male rabbit hearts had early after depolarization, but only one of these hearts developed TdP.Conclusion Greater TDR may play an important role in the higher incidence of TdP in female rabbit hearts.

Survival effect of probiotics in a rat model of colorectal cancer treated with capecitabine

BACKGROUND Probiotics are used to manage a number of gastrointestinal disorders due to their beneficial properties.Clinical reports showed that probiotics also improve the life quality of patients with colorectal cancer(CRC)subjected to oncologic treatment.In a CRC animal model,probiotics supplementation has the potential to decrease the formation of aberrant crypts and ameliorate tumor malignancy,enhancing the antitumor effect of 5-fluorouracil(5-FU)chemotherapy.Based on these data,we hypothesize that the administration of probiotics impact positively in the overall survival and life quality of rats with CRC under the treatment of capecitabine,which is the pro drug of 5-FU.AIM To evaluate the probiotics effects in a rat CRC model treated with capecitabine and followed until the end of life.METHODS 1,2-Dimethylhidrazine dihydrochloride(1,2-DMH)was employed as carcinogen inductor of CRC.Fifty male Wistar-Lewis rats were randomly assigned to one of five following groups:Control(n=5),Control+probiotics(Control-P group,n=5),1,2-DMH alone(DMH group,n=10),1,2-DMH+capecitabine(DMH-C group,n=10),1,2-DMH+probiotics(DMH-P group,n=10)and 1,2-DMH+capecitabine+probiotics(DMH-C-P group,n=10).All parametric data were expressed as the mean±SD.The statistical significance of differences was analyzed using one-way ANOVA.Data were analyzed with InfoStat software.The results were considered statistically significant at P<0.05.Overall survival was evaluated with the Kaplan-Meier estimator with the log-rank test.RESULTS The data of mean overall survival for DMH,DMH-P,DMH-C,DMH-C-P,Control and Control-P groups were 250 d[95%confidence interval(CI):242.5-253.1],268 d(95%CI:246.3-271.4),380 d(95%CI:337.8-421.9),480 d(95%CI:436.9-530.7),588 d(95%CI:565.8-609.3)and 590 d(95%CI:564.3-612.9),respectively,with a significant difference between DMH-C and DMH-C-P groups(P=0.001).Comparing all groups by Kaplan-Meier estimator,we found a significantly different in the overall survival of DMH and DMH-P groups respect to DMH-C(P=0.001)and DMH-C-P(P=

animal models are live subjects applied to translational research.They provide insights into human diseases and enhance biomedical knowledge.Livestock development has favored the pace of human social development over millennia.Today's society is more aware of animal welfare than past generations.The general public has marked objections to animal research and many species are falling into disuse.The search for an ideal methodology to replace animal use is on,but animal modeling still holds great importance to human health.Bone research,in particular,has unmet requirements that in vitro technologies cannot fully address.Standardizing novel models remains necessary and rabbits are gaining in popularity as potential bone models.Our aim here is to provide a broad overview of animal modeling and its ethical implications,followed by a narrower focus on bone research and the role rabbits are playing in the current scenario.

BACKGROUND Hybrid endoscopic submucosal dissection(ESD)that comprises mucosal incision and partial submucosal dissection followed by snaring in a planned manner,has been developed for endoscopic resection of gastrointestinal neoplasms to overcome the technical barrier of ESD.Although the superiority of hybrid ESD with SOUTEN,a single multifunctional device,over conventional ESD has been indicated,the actual effect of snaring itself remains unclear since SOUTEN could be applied to hybrid ESD group,but not to the conventional ESD group,due to ethical issue in clinical practice.AIM To determine whether and how hybrid ESD was superior to conventional ESD in the endoscopic treatment of gastric lesions in an ex vivo porcine model basic study.METHODS Sixteen endoscopists participated in this basic study in August 2020 at Kyushu University,performing 32 procedures each for hybrid ESD and conventional ESD.Mock lesions(10-15 mm,diameter)were created in the porcine stomach.The primary outcome was total procedure time and secondary outcomes were en bloc or complete resection,perforation,procedure time/speed for both,mucosal incision,and submucosal dissection.Factors associated with difficulty in ESD including longer procedure time,incomplete resection,and perforation,were also investigated.Categorical and continuous data were analyzed using the chi-square test or Fisher’s exact test and the Mann-Whitney U test,respectively.RESULTS The median total procedure time of hybrid ESD was significantly shorter than that of conventional ESD(median:8.3 min vs 16.2 min,P<0.001).Time,speed,and the amount of hyaluronic acid during submucosal dissection were more favorable in hybrid ESD than conventional ESD(time,5.2 min vs 10.4 min,P<0.001;speed,43.7 mm^(2)/min vs 23.8 mm^(2)/min,P<0.00;injection volume,1.5 mL vs 3.0 mL,P<0.001),although no significant differences in those factors were observed between both groups during mucosal incision.There was also no significant difference between both groups in the en bloc/complete resection rate an

Application of biological dural graft made by meninges from porkers

BACKGROUND： Presently, over 40 kinds of dural grafts have been successively used in clinic. Among them, lyophilized human dura mater with good histocompatibility and less complications is applied most widely. But there are a few reports on cases of infected spongiform encephalopathy following application of lyodura. More ideal repair materials deserve to be further investigated. OBJECTIVE： To investigate the efficiency and safety of biological dural graft made by meninges from porkers to repair meningeal injury. DESIGN： A self-control observation. SETTING： Wuhan General Hospital of Guangzhou Military Area Command of Chinese PLA. MATERIALS： Sixteen New Zealand Rabbits, of either gender, weighing from 2 to 3 kg, of clean grade Ⅱ, with the age of 0.5 - 1 year, were involved in this experiment. The involved rabbits were provided by the animal Experimental Center of the First Military Medical University of Chinese PLA. Biological surgical patch （dural graft） was developed by Guangdong Guanhao Biotechnological Co.,Ltd. It was processed by using meninges from porkers by tissue engineering technology. METHODS： This experiment was carried out in the Experimental Center of the 157 Hospital of Chinese PLA between December 2003 and June 2004. ①The experimental rabbits were anesthetized. Dura mater was exposed from two sides ofpostmedial line of coronal suture. A rectangular dura mater about 8 mm ×8 mm in size was cut off. Then a biological surgical patch （dural graft） was sheared into insert with 8 mm diameter and sutured. The left dura mater was untouched and used as control. Scalp was sutured, and postoperative wound healing and recovery were observed. ②The anesthetized rabbits were sacrificed at postoperative 3, 14, 30 and 90 days, 4 rabbits once. The whole head was cut off, and its scalp was removed. Afterwards, the head was fixed by formalin. Complete Achilles Tenotomy:A New Improved Experimental Surgical Technique in Rats

Rupture of the Achilles tendon is one of the most frequent tendon injuries in humans. However, there is no standard surgical technique thus far to reproduce complete rupture of this tendon in rats. Surgical procedures were performed in Wistar rats under light microscopy, with posterior longitudinal incision and preservation of the plantaris tendon. The animals were sacrificed on 2nd and 8th postoperative week and we used hematoxilin and eosin, picrosirius-red and Weigert’s resorcin-fuchsin to observe general tendon healing. The mean operative time was one minute and 24 seconds. Histological studies showed cellular proliferation and fibrillogenesis at two weeks, with decreased amounts of cellularity and elastic fibers at the 8th week, besides changes in structural organization of collagen fibers. The complete Achilles tenotomy in rats, was improved under light microscopy and sparing the plantaris tendon. This surgical technique was simple and quick, showed signs of normal healing process, could be easily reproducible, and triggered translational researches in this area.

本方法所用栓堵材料为聚醚型聚氨酯弹性体,可用注射方法推入输精管内,迅速固化为栓子,堵塞输精管腔,阻断精子通过,达至避孕的目的。需要再通时,可顺利取出栓子,恢复输精管的再通。该材料经毒理检查无毒、无害、不致癌、不致畸、不致突变,是输精管堵塞的适用材料。本方法既可阻断精子通过,又不会使输精管组织因受压而坏死。病理检查表明栓堵段输精管壁内,无炎症、无粘连、无肉芽及瘢痕组织形成等病理改变。通过二年多来临床观察,其节育效果:精子消失率达到98%,并发症为1.44%,可复率100%,证实了本法是目前较为满意、安全、高效、可靠、简便、经济的男性节育措施。
