Background： angiotensin ii （Ang ll） is a major contributor to the development of heart failure. However, the molecular and cellular mechanisms that underlie this process remain elusive. Inadequate angiogenesis in the myocardium leads to a transition from cardiac hypertrophy to dysfunction, and our previous study showed that Ang ii significantly impaired the angiogenesis response. The current study was designed to examine the role of Jaggedl-Notch signaling in the effect of Ang ii during impaired angiogenesis and cardiac hypertrophy. Methods： Ang ii was subcutaneously infused into 8-week-old male C57BL/6 mice at a dose of 200 ng·kg^-1·min^-1 for 2 weeks using Alzet micro-osmotic pumps. N-[N-（3, 5-difluorophenacetyl）-L-alanyl]-S-phenylglycine tert-butyl ester （DAPT）, a y-secretase inhibitor, was injected subcutaneously during Ang ii infusion at a dose of 10.0 mg.kg^-1·d^-1. Forty mice were divided into four groups （n = 10 per group）： control group; Ang 11 group, treated with Ang I1; DAPT group, treated with DAPT; and Ang ii ＋ DAPT group, treated with both Ang I1 and DAPT. At the end of experiments, myocardial （left ventricle [LV]） tissue from each experimental group was evaluated using immunohistochemistry, Western blotting, and real-time polymerase chain reaction, Data were analyzed using one-way analysis of variance test followed by the least significant difference method or independent samples t-test. Results： Ang ii treatment significantly induced cardiac hypertrophy and impaired the angiogenesis response compared to controls, as shown by hematoxylin and eosin （HE） staining and immunohistochemistry fbr CD31, a vascular marker （P 〈 0.05 for both）. Meanwhile, Jaggedl protein was significantly increased, but gene expression for both Jagl and Heyl was decreased in the LV following Ang ii treatment, compared to that in controls （relative ratio for Jag1 gene： 0.45 ± 0.13 vs. 0.84± 0.15; relative ratio for Heyl gene： 0.51 ± 0.08 vs. 0.91 ± 0.09; P 〈 0.05）. Effects of chronic peripheral pretreatment with an angiotensin ii type-1 receptor blocker on apoptosis-related molecules in rats with cerebral ischemia/reperfusion injury

Chronic systemic treatment with blockers of angiotensin ii type-1 （AT1） receptors inhibits ischemia-induced apoptosis and reduces ischemic neuronal damage. However, the molecular mechanisms of AT1 receptor blockers in modulating neuronal apoptosis remain poorly understood. Pretreatment with irbesartan significantly suppressed cell apoptosis at 1-7 days following cerebral ischemia/reperfusion, increased levels of brain-derived neurotrophic factor, and elevated the ratios of Bcl-2/Bax and phosphorylated cyclic adenosine monophosphate response element-binding protein （pCREB）/CREB in the ischemic cortex at 1 day after reperfusion, as well as suppressing caspase-3 activation. Cerebral ischemia increased the mRNA expression of AT1 and AT2 receptors in the ischemic cortex, whereas irbesartan blocked this increase in AT1 expression but potentiated the expression of AT2. Therefore, this AT1 receptor blocker was neuroprotective by increasing the ratios of Bcl-2/Bax and pCREB/CREB, increasing brain-derived neurotrophic factor levels, inhibiting caspase-3 activation, and modulating AT receptor expression.

Effect of gestational protein restriction on left ventricle hypertrophy and heart angiotensin ii signaling pathway in adult offspring rats

Maternal protein restriction may be a risk factor for cardiovascular disorders in adulthood. The RAS (renin-angiotensin-system) plays a pivotal role in cardiac remodeling. Components of the RAS, including angiotensin ii (Angii) and its receptors type 1 (AT1R) and 2 (AT2R) are expressed in the heart. This study investigates whether gestational protein restriction alters the expression and localization of AT1R and AT2R and RAS signaling pathway proteins in parallel with left ventricle hypertrophy and systemic hypertension in male offspring. Dams were kept on normal (NP, 17% protein) or low (LP, 6% protein) protein diet during pregnancy. Systolic blood pressure (SBP) of male offspring was measured from the 8th to 16th week and left ventricles of 16-wk-old rats were processed for histology, morphometric, immunoblotting and immunohistochemistry. LP offspring showed a significant reduction in birth body weight and SBP increased significantly from the 8th week. Left ventricle mass and cardiomyocytes area were also significantly higher in LP animals. Widespread perivascular fibrosis was not detected in the heart tissue. Analysis by immunoblotting and immunohistochemistry demonstrated a significant enhance in cardiomyocyte expression of AT1R and ERK1 in LP offspring. Expression of PI3K in LP was significantly reduced in cardiomyocytes and in the intramural coronary wall, while AT2R expression was unchanged in the NP group. We also found reduced LP expression of JAK2 and STAT3. In conclusion, our data also suggest that changes in the RAS may play a role in the ventricular growth through upregulation of the AT1-mediated ERK1/2 response, despite unchanged AT2R expression.

Aim:angiotensin Ⅱ is believed to play an important role in tissue repair andremodeling in lungs by the angiotensin type I(AT1)receptor via a number ofpotential mechanisms.However,the role of the AT1 receptor in early lung injuryhas not been characterized.Methods:Bleomycin-induced pulmonary fibrosis(PF)in rats was utilized to value the treatment with valsartan,an AT1 receptorantagonist,by measurement of body weight,wet weight of the left lung,hydroxy-proline content,mRNA expression of collagen Ⅰ/Ⅲ,and the degree of fibrosis inlung tissues on d 21.Tissue injury in the early phase was assessed on d 1,3 and7 by apoptosis,malondialdehyde content,myeloperoxidase activity,inflamma-tory cell count and protein content.angiotensin converting enzyme(ACE)activ-ity and the AT1 receptor in lung tissues were analyzed by biochemistry methodand Western blotting,respectively.Results:Valsartan ameliorated PF induced bybleomycin in the rats on d 21.After bleomycin was injected intratracheally,in-creases in the lung AT1 receptor and ACE activity were observed by d 1,3 and 7.Lung injury deteriorated in the early phase.Valsartan reduced the increase of theAT1 receptor,ACE activity and lung injury induced by bleomycin in the earlyphase.Conclusion:These observations suggest that angiotensin Ⅱ may play apotent role in early lung injury via the AT1 receptor.AT1 receptor antagonistsshould be assessed as potential new therapies for fibrotic lung disease.

Erythropoietin(EPO)is the main mediator of erythropoiesis and an important tissue protective hormone that appears to mediate an ancestral neuroprotective innate immune response mechanism at an early age.When the young brain is threatened-prematurity,neonatal hyperbilirubinemia,malaria-EPO is hypersecreted disproportionately to any concurrent anemic stimuli.Under eons of severe malarial selection pressure,neuroprotective EPO augmenting genetic determinants such as the various hemoglobinopathies,and the angiotensin converting enzyme(ACE)I/D polymorphism,have been positively selected.When malarial and other cerebral threats abate and the young child survives to adulthood,EPO subsides.Sustained high ACE and angiotensin ii(Ang ii)levels through the ACE D allele in adulthood may then become detrimental as witnessed by epidemiological studies.The ubiquitous renin angiotensin system(RAS)influences theα-klotho/fibroblast growth factor 23(FGF23)circuitry,and both are interconnected with EPO.Here we propose that at a young age,EPO augmenting genetic determinants through ACE D allele elevated Ang ii levels in some or HbE/beta thalassemia in others would increase EPO levels and shield against coronavirus disease 2019,akin to protection from malaria and dengue fever.Human evolution may use ACE2 as a“bait”for severe acute respiratory syndrome coronavirus-2(SARS-CoV-2)to gain cellular entry in order to trigger an ACE/ACE2 imbalance and stimulate EPO hypersecretion using tissue RAS,uncoupled from hemoglobin levels.In subjects without EPO augmenting genetic determinants at any age,ACE2 binding and internalization upon SARS-CoV-2 entry would trigger an ACE/ACE2 imbalance,and Ang ii oversecretion leading to protective EPO stimulation.In children,low nasal ACE2 Levels would beneficially augment this imbalance,especially for those without protective genetic determinants.On the other hand,in predisposed adults with the ACE D allele,ACE/ACE2 imbalance,may lead to uncontrolled RAS overactivity and an Ang ii induced proinflammatory state

Clustering of Inflammatory Biomarkers and Risk of Hypertension in a Mongolian Population in China

Abstract Objective There is little knowledge on whether there is clustering of inflammatory biomarkers, such as C-reactive protein （CRP）, soluble intracellular adhesion molecule1-1 （slCAM-1）, and angiotensin It （Ang Ⅱ）, in individuals with hypertension in the Mongolian population. In the present study, we investigated this relationship in a Mongolian population in China. Methods A total of 2589 adult Mongolians, aged 20 years and older, were recruited as study participants. Data on demographics, lifestyle, family history of hypertension, blood pressure, and blood chemistry were collected, and inflammatory biomarkers were measured in all participants. Results The proportion of subjects with increased levels of two or three biomarkers was significantly higher in those with hypertension （21.0% and 6.0%, respectively） than in those with prehypertension （12.7% and 0.5%, respectively） or normotension （8.1% and 0.2%, respectively）. The multivariate adjusted odds ratios （95% confidence interval） of hypertension associated with increased levels of one, two or three biomarkers were 0.94 （0.72-1.22）, 1.42 （0.93-2.16）, and 11.08 （1.45-84.80）, respectively, compared with subjects with no increase in any biomarker. Conclusion Hypertension was associated with a cluster of inflammatory biomarkers in the Mongolian population.

Enhanced sympathetic activity and cardiac sympathetic afferent reflex in rats with heart failure induced by adriamycin

Our previous studies have shown that the cardiac sympathetic afferent reflex is enhanced in rats with chronic heart failure （CHF） induced by coronary artery ligation and contributes to the over-excitation of sympathetic ac- tivity. We sought to determine whether sympathetic activity and cardiac sympathetic afferent reflex were en- hanced in adriamycin-induced CHF and whether angiotensin ii （Ang ii） in the paraventricular nucleus （PVN） was involved in enhancing sympathetic activity and cardiac sympathetic afferent reflex. Heart failure was induced by intraperitoneal injection of adriamycin for six times during 2 weeks （15 mg/kg）. Six weeks after the first injec- tion, the rats underwent anesthesia with urethane and a-chloralose. After vagotomy and baroreceptor denervation, cardiac sympathetic afferent reflex was evaluated by renal sympathetic nerve activity and mean arterial pressure （MAP） response to epicardial application of capsaicin （1.0 nmol）. The response of MAP to ganglionic blockade with hexamethonium in conscious rats was performed to evaluate sympathetic activity. The renal sympathetic nerve activity and cardiac sympathetic afferent reflex were enhanced in adriamycin rats and the maximum depres- sor response of MAP induced by hexamethonium was significantly greater in adriamycin rats than that in control rats. Bilateral PVN microinjection of angiotensin ii （Ang ii） caused larger responses of the cardiac sympathetic afferent reflex, baseline renal sympathetic nerve activity and MAP in adriamycin rats than control rats. These re- sults indicated that both sympathetic activity and cardiac sympathetic afferent reflex were enhanced and Ang ii in the PVN was involved in the enhanced sympathetic activity and cardiac sympathetic afferent reflex in rats with adriamycin-induced heart failure.

Effects of tetrandrine on smooth muscle contraction induced by mediators in pulmonary hypertension

AIM: In attempt to characterize tetrandrine on pulmonary hypertension, biological activities induced by a range of mediators implicated in the pathogenesis of pulmonary hypertension were investigated. METHODS: Pulmonary artery rings and tracheal segments were contracted with couples of bioactive substances in which a series experi-ments including effects of tetrandrine on calcium agonist, endothelin, thromboxane A2, angiotensin ii, neuropeptide Y, histamine, 5-methyl furmethide were performed, the influences of tetrandrine in the concentration of 1 to 30 μmol/L were investigated. RESULTS: Tetrandrine inhibited calcium agonist BayK8644, endothelin-1 and throm-boxane A2 mimetic U46619, angiotensin ii- and neuropeptide Y-induced contractile responses with depression of the maximal contraction of pulmonary artery rings in a varying extent. Involvement of Ca^(2+)-activated K^+ Channels in Receptor-Regulated Sperm Motility in Rats

Previous voltage clamp studies have demonstrated the modulation of sperm Ca 2+ activated K + (KCa) channels expressed in Xenopus oocytes by angiotensin ii (Ang ii) and extracellular ATP via AT 1 receptor and P 2U receptor, respectively. In the present study, we investigated the involvement of KCa channels in receptor regulated sperm motility of the rat using a computer aided sperm analysis system, HTM IVOS, in conjunction with Ca 2+ mobilizing agents, receptor agonists/antagonists and KCa channels blockers. The percentage of motile sperm was increased by ionomycin (0.5 μmol/L), which could be inhibited by K + channel blockers, tetraethylammonium (TEA 1 μmol/L ) or charybdotoxin (ChTX, 300 nmol/L) indicating the presence of KCa channels. Ang ii, at low concentration, 10 nmol/L, was found to increase motility, however, at higher concentration, 1 μmol/L, percentage of motility was found to be suppressed. Both stimulatory and inhibitory effects of Ang ii could be reversed by losartan, a specific antagonist of AT 1 receptors, but not AT 2 antagonist PD123177, indicating the involvement of AT 1 but not AT2 receptor in mediating both effects. ChTX also abolished both stimulatory and inhibitory effects of Ang ii, suggesting the involvement of KCa channels. The percentage of motility was also enhanced by extracellular ATP, a factor known to be involved in sperm activation. The ATP enhanced sperm motility was mimicked by UTP, and inhibited by ChTX and reactive blue, an antagonist of P 2 receptor, indicating the involvement of both P 2U and KCa channels. RT PCR study was also conducted to confirm the expression of KCa channels, AT 1 receptors and P 2U receptor, but not AT 2 receptor, in rat caudal epididymal sperm. The present findings suggest an important role of KCa channels in the regulation of sperm motility by AT 1 and P 2U receptors.

Perinatally Imposed Essential Fatty Acid Deficiency Changes Renal Function of the Adult Rat

This study was designed to investigate whether essential fatty acid deficiency early during development could change the content of phospholipids and cholesterol in whole membranes of the kidney and renal function at adult life. For this, female Wistar rats were maintained on a standard diet or on an essential fatty acid deficient diet (EFAD) from the age of 30 days, throughout the pregnancy, at age of 90 days and until the weaning, for evaluation of their offspring. Weanling rats were maintained on a standard diet until the age of 13 weeks. Systolic blood pressure (SBP), glomerular filtration rate (GFR), urinary sodium excretion (UNa+V), positive cells for angiotensin ii (Ang ii) and cholesterol and phospholipids in whole membranes of the kidney were evaluated. Cholesterol, total phospholipids and the relative content of classes of phospholipids were unaltered in the cortex and medullary kidney. SBP, GFR and UNa+V were also unaltered in the EFAD group. However, the number of positive cells for Ang ii in the tubulointerstitial area of the renal cortex was higher in the EFAD group. Therefore, these findings indicated that although cholesterol and phospholipids were unaltered and urinary sodium excretion was unchanged, Ang ii expression in the kidney was erroneously programmed and later hindering of renal function was not ruled out.
