Hepatoprotective activity of Macrothelypteris torresiana (Gaudich.) aerial parts against CCl4-induced hepatotoxicity in rodents and analysis of polyphenolic compounds by HPTLC

Macrothelypteris torresiana is a fern species belonging to the family Thelypteridaceae. The present study was conducted to evaluate hepatoprotective potential of ethanol extract from M. torresiana aerial parts(EEMTAP)and detect the polyphenolic compounds present in the extract using high performance thin layer chromatography(HPTLC). Hepatoprotective potential of EEMTAP were tested at doses of 300 and 600 mg/kg, per os(p.o.), on Wistar albino rats. The extract and silymarin treated animal groups showed significant decrease in activities of different biochemical parameters like serum glutamic oxaloacetic transaminase(SGOT), serum glutamate-pyruvate transaminase(SGPT), and alkaline phosphatase(ALP), which were elevated by carbon tetrachloride(CCl_4) intoxication. The levels of total bilirubin and total protein along with the liver weight were also restored to normalcy by EEMTAP and silymarin treatment. After CCl_4 administration, the levels of hepatic antioxidant enzymes such as glutathione(GSH) and catalase(CAT) were decreased whereas the level of hepatic lipid peroxidation(LPO) was elevated. The levels of these hepatic antioxidant enzymes were also brought to normalcy by EEMTAP and silymarin treatment. Histological studies supported the biochemical findings, and treatment with EEMTAP at doses of 300 and 600 mg/kg, p.o. was found to be effective in restoring CCl_4-induced hepatotoxicity in rats. A simple HPTLC analysis was conducted for the detection of polyphenolic compounds in EEMTAP, and the result revealed the presence of caffeic acid as phenolic acid and quercetin as flavonoid. The proposed HPTLC method is simple and concise and provides a good resolution of caffeic acid and quercetin from other constituents present in EEMTAP.

Heavy metal availability and impact on activity of soil microorganisms along a Cu/Zn contamination gradient

All the regulations that define a maximum concentration of metals in the receiving soil are based on total soil metal concentration. However, the potential toxicity of a heavy metal in the soil depends on its speciation and availability. We studied the effects of heavy metal speciation and availability on soil microorganism activities along a Cu/Zn contamination gradient. Microbial biomass and enzyme activity of soil contaminated with both Cu and Zn were investigated. The results showed that microbial biomass was negatively affected by the elevated metal levels. The microbial biomass-C （Cmic）/organic C （Corg） ratio was closely correlated to heavy metal stress. There were negative correlations between soil microbial biomass, phosphatase activity and NH4NO3 extractable heavy metals. The soil microorganism activity could be predicted using empirical models with the availability of Cu and Zn. We observed that 72% of the variation in phosphatase activity could be explained by the NH4NO3-extractable and total heavy metal concentration. By considering different monitoring approaches and different viewpoints, this set of methods applied in this study seemed sensitive to site differences and contributed to a better understanding of the effects of heavy metals on the size and activity of microorganisms in soils. The data presented demonstrate the relationship between heavy metals availability and heavy metal toxicity to soil microorganism along a contamination gradient.

No difference in activity of Sod-1 genotypes of Poa annua to short-term treatment of ambient gaseous organic pollution

Our previous studies indicated that genotypes at locus Sod-1 of Poa annua changed clinally along a gradient of gaseous organic pollution. In the present study, we aimed to know whether there were differential responses of activities of different superoxide dismutase （SOD） genotypes to short-term treatment of ambient gaseous organic pollution. Significant bias from Hardy-Weinberg equibrium was observed on locus Sod-1, and no genotype Sod- l-BB was found. Significantly increased activities were observed in most treatments for genotype Sod-1-AA and in one treatment for genotype Sod-1-AB. However, no significant difference in SOD activities was found between the two genotypes. It was interpreted that fitness difference between the two genotypes was small and treatment duration was too short or extent of ambient organic pollution was too low to lead to differential responses. Other environmental factor effects on activities of superoxide dismutases can also explain the results.

Anti-proliferation activity of terpenoids isolated from Euphorbia kansui in human cancer cells and their structure-activity relationship

Euphorbia kansui is a commonly used traditional Chinese medicine for the treatment of edema,pleural effusion,and asthma,etc.According to the previous researches,terpenoids in E.kansui possess various biological activities,e.g.,anti-virus,anti-allergy,antitumor effects.In this work,twenty five terpenoids were isolated from E.kansui,including thirteen ingenane-and eight jatrophane-type diterpenoids(with two new compounds,kansuinin P and Q)and four triterpenoids.Eighteen of them were analyzed by MTS assay for in vitro anticancer activity in five human cancer cell lines.Structure-activity relationship for 12 ingenane-type diterpenoids in colorectal cancer Colo205 cells were preliminary studied.Significant anti-proliferation activities were observed in human melanoma cells breast cancer MDA-MB-435 cells and Colo205 cells.More than half of the isolated ingenane-type diterpenoids showed inhibitory activities in MDA-MB-435 cells.Eight ingenane-and one jatrophane-type diterpenoids possessed much lower IC_(50) values in MDA-MB-435 cells than positive control staurosporine.Preliminary structure-activity relationship analysis showed that substituent on position 20 was important for the activity of ingenane-type diterpenoids in Colo205 cells and substituent on position 3 contributed more significant biological activity of the compounds than that on position 5 in both MDA-MB-435 and Colo205 cells.

Background:Metabolic syndrome(MetS) is a global health problem.Physical activity(PA) is a known modifiable risk factor for MetS and individual MetS components.However,the role of PA could differ between sub-populations due to differences in the variability of PA and other MetS risk factors.To examine these differences,multi-country studies with standardized outcome measurement methods across cohorts are needed.Methods:Cross-sectional PA levels(total and domain specific) in healthy middle-aged(44-56 years) men in the Risk Factor Assessment among Japanese and U.S.Men in the Post-World War Ⅱ Birth Cohort(ERA-JUMP) Study(n=730;American:n=417;Japanese:n=313;from population-representative samples in Pittsburgh,Pennsylvania,USA,and Kusatsu,Shiga,Japan) were compared.The relationships between PA levels and MetS(overall and specific components) in/across the American and Japanese sub-cohorts(adjusting for age,smoking,and alcohol consumption)were also assessed using the same instruments(pedometer and validated questionnaire) to measure PA in both cohorts.Results:A total of 510 individuals provided complete data on PA(American:n=265;Japanese:n=245).The American cohort had significantly lower mean±SD steps/day(7878±3399 steps/day) vs.the Japanese cohort(9055±3797 steps/day)(p <0.001) but had significantly higher self-reported moderate-vigorous leisure PA(American:15.9(7.4-30.3) metabolic task equivalent hours per week(MET-b/week) vs.Japanese:4.0(0-11.3) METh/week,p <0.0001).In both sub-cohorts,each 1000 steps/day increase was associated with lower odds of having MetS(American:OR=0.90,95%CI:0.83-0.98;Japanese:OR=0.87,95%CI:0.79-0.95) and the individual MetS component of high waist circumference(American:OR=0.86,95%CI:0.79-0.94;Japanese:OR=0.87,95%CI:0.80-0.95).In the American cohort only,higher self-reported leisure PA(Met-b/week) was associated with lower odds of MetS and high waist circumference(OR=0.98,95%CI:0.97-0.99 for MetS and waist circumference,respectively).Conclusion:Higher total step counts/day had an important pr

Myotropic activity and immunolocalization of selected neuropeptides of the burying beetle Nicrophorus vespilloides (Coleoptera: Silphidae)

Burying beetles (Nicrophorus sp.) are necrophagous insects with developed parental care. Genome of Nicrophorus vespilloides has been recently sequenced, which makes them interesting model organism in behavioral ecology. However, we know very little about their physiology, including the functioning of their neuroendocrine system. In this study, one of the physiological activities of proctolin, myosuppressin (Nieve? MS), myoinhibitory peptide (Trica-MIP-5) and the short neuropeptide F (Nicve-sNPF) in N. vespilloides have been investigated. The tested neuropeptides were myoactive on N. vespilloides hindgut. After application of the proctolin increased hindgut contraction frequency was observed (EC50 value was 5.47 x 10-8 mol/L). The other tested neuropeptides led to inhibition of N. vespilloides hindgut contractions (Nicve-MS: IC50 = 5.20 x 10~5 mol/L;Trica-MIP-5: IC50 = 5.95 x 10-6 mol/L;Nicvc-sNPF: IC50 = 4.08 x 10-5 mol/L). Moreover, the tested neuropeptides were immunolocalized in the nervous system of N. vespilloides. Neurons containing sNPF and MIP in brain and ventral nerve cord (VNC) were identified. Proctolin-immunolabeled neurons only in VNC were observed. Moreover, MIP-immunolabeled varicosities and fibers in retrocerebral complex were observed. In addition, our results have been supplemented with alignments of amino acid sequences of these neuropeptides in beetle species. This alignment analysis clearly showed amino acid sequence similarities between neuropeptides. Moreover, this allowed to deduce amino acid sequence of N. vespilloides proctolin (RYLPTa), Nicve-MS (QDVDHVFLRFa) and six isoforms ofNicve-MIP (Nicve-MIP-1一 DWNRNLHSWa;Nicve-MIP-2—AWQNLQGGWa;Nicve-MIP-3—AWQNLQGGWa;Nicve-MlP-4—AWKNLNNAGWa;Nicve-MIP-5—SEWGNFRGSWa;Nicve-MIP-6— DPAWTNLKGIWa;and Nicve-sNPF—SGRSPSLRLRFa).

activity and Formation of Sand Waves on Northern South China Sea Shelf

Sand waves on the northern South China Sea shelf had been considered as stable relict bed form. For the industry use of sea bed between stations LF13-2 and LF13-1, a new round of explorations were conducted. The newly obtained data show that both spacings and amplitudes of sand waves are all systematically changing with water depth. Repeated observations since 2003 to 2004 showed that the sea bed is currently active. Due to strong erosion of surface sediment since Dongsha （东沙） uplifting, there are almost no modern sediments on the shelf of Dongsha area. Sand materials in the study area mainly originate from the erosion of the bed sediment formation. The water depth increment revealed by repeated echo sounder data is mainly due to erosion. Bottom currents are quite complex in the area of Dongsha underwater plateaus. At site 9MKH, the southward ebb current is stronger than the north- ward flood current, while at site AEM-HR, the WNW-ward flood current is slightly stronger than the ESE-ward ebb current. At site 9MKII, the maximum bottom current speed is 48 cm/s, and 22% of the observed bottom current speeds are larger than 20 cm/s, which meet the minimum bottom current speed required for the creation of sand wave. This article points out that present-day oceanographic condition couples well with the sand-wave morphologies, and that the sand waves are to a great extent in equilibrium with the ongoing present-day oceanographic bottom current condition and active.

Integrated analysis of gene expression and methylation profiles of novel pancreatic cancer cell lines with highly metastatic activity

Pancreatic cancer is one of the most lethal human malignancies, partly because of its propensity for metastasis. However, highly metastatic human pancreatic cancer cell lines suitable for studies of metastasis are currently lacking. Here we established two highly metastatic human pancreatic cancer cell lines, MIA PaCa-2 In8 and Panc-1 In8, by Matrigel induction assay. The cell lines were further characterized both in vitro and in vivo. MIA PaCa-2 In8 and Panc-1 In8 cells demonstrated increased migration and invasion compared with their respective parental cells. Following injection into nude mice, MIA PaCa-2 In8 and Panc-1 In8 cells resulted in more pulmonary metastases compared with the parental cells. Furthermore, analyses of m RNA, long non-coding RNA, micro RNA, and methylation profiling revealed that these factors were aberrantly regulated in the highly metastatic cells,indicating that they probably affected metastasis. We thus established and characterized two highly metastatic human pancreatic cell lines that could be used as valuable tools for future investigations into the pathogenesis, metastasis, and potential treatment of human pancreatic cancer.

Improved anti-tumor activity and safety profile of a paclitaxel-loaded glycyrrhetinic acid-graft-hyaluronic acid conjugate as a synergistically targeted drug delivery system

The present study was designed to develop and evaluate glycyrrhetinic acid-graft-hyaluronic acid(HGA) conjugate for intravenous paclitaxel(PTX) delivery. Lyophilized PTX-loaded self-assembled HGA nanoparticles(PTX/HGAs) were prepared and characterized by dynamic light scattering measurements. Hemolysis test, intravenous irritation assessment, and in vitro and in vivo pharmacodynamic studies were carried out. B16F10 and HepG2 cells were used in the cell apoptosis analysis. The mouse MDA-MB-231 xenograft model was used for the evaluation of in vivo anticancer activity of the drugs, by the analysis of tumor growth and side effects on other tissues. PTX/HGAs showed high stability and good biocompability. Compared with PTX plus GA plus HA solution, PTX/HGAs displayed obvious superiority in inducing the apoptosis of the cancer cells. Following systemic administration, PTX/HGAs efficiently suppressed tumor growth, with mean tumor inhibition ratio(TIR) being 65.08%, which was significantly higher than that of PTX plus GA plus HA treatment. In conclusion, PTX/HGAs demonstrated inhibitory effects tumor growth without unwanted side effects, suggesting that HGA conjugates hold a great potential as a delivery carrier for cancer chemotherapeutics to improve therapeutic efficacy and minimize adverse effects.

Reliability and validity of the French version of the Global Physical activity Questionnaire

Background:The Global Physical activity Questionnaire(GPAQ)has been used to measure physical activity(PA)and sedentary time in France,but no study has assessed its psychometric properties.This study aimed to compare the reliability as well as criterion and concurrent validity of the French version of the GPAQ with the French International Physical activity Questionnaire long form(IPAQ-LF)and use of an accelerometer in a general adult population.Methods:We included 92 participants(students or staff)from the Medicine Campus at the University of Lorraine,Nancy(north-eastern France).The French GPAQ was completed twice,7 days apart,to study test-retest reliability.The IPAQ-LF was used to assess concurrent validity of the GPAQ,and participants wore an accelerometer(ActiGraph GT3X+)for 7 days to study criterion validity.Reliability as well as concurrent and criterion validity of the GPAQ were tested by the intraclass correlation coefficient(ICC),Spearman correlation coefficient for quantitative variables,and Kappa and Phi coefficients for qualitative variables.Both concurrent and criterion validity of GPAQ were assessed by Bland-Altman plots.Results:The GPAQ showed poor to good reliability(ICC=0.37-0.94;Kappa=0.50-0.62)and concurrent validity(Spearman r=0.41-0.86),but only poor criterion validity(Spearman r=-0.22-0.42).Limits of agreement for the GPAQ and accelerometer were wide,with differences between 286.5 min/week and 601.3 min/week.Conclusion:The French version of the GPAQ provides limited but acceptable reliability and validity for the measurement of PA and sedentary time.It may be used for assessing PA and sedentary time in a French adult population.
