A recent article in the Opto-Electronic Advances(OEA)journal from Prof.Qian Chen and Prof.Chao Zuo’s group introduced a new and efficient 3D imaging system that captures high-speed images using deep learning-enabled fringe projection profilometry(FPP).In this News&Views article,we explore potential avenues for future advancements,including expanding the measurement range through an extended number-theoretical approach,enhancing quality through the incorporation of horizontal fringes,and integrating data from other modalities to broaden the system's applications.

Gargantulides B and C,two new and highly complex 52-membered glycosylated macrolactones,were isolated from Amycolatopsis sp.strain CA-230715 during an antibacterial screening campaign.The structures of these giant macrolides were elucidated by 2D NMR spectroscopy and shown to be related to gargantulide A,although containing additional β-glucopyranose and/orα-arabinofuranose monosaccharides separately attached to their backbones.Genome sequencing allowed the identification of a strikingly large 216 kbp biosynthetic gene cluster,among the largest type Ⅰ PKS clusters described so far,and the proposal of a previously unreported biosynthetic pathway for gargantulides A-C.The absolute configurations of gargantulides B and C were assigned based on a combination of NMR and bioinformatics analysis of ketoreductase and enoylreductase domains within the multimodular type Ⅰ PKS.In addition,the absolute stereochemistry of gargantulide A has now been revised and completed.Gargantulides B and C display potent antibacterial activity against a set of drug-resistant Gram-positive bacteria and moderate activity against the clinically relevant Gram-negative pathogen Acinetobacter baumannii.

A tri-color quad-beam digital shearography is proposed to achieve the measurement of absolute three-dimensional(3D) displacement gradients.Four laser beams with three different center wavelengths are symmetrically irradiated to the object surface from the upper and lower left and right directions.Four phase maps are then extracted from the two interferograms obtained from two shots.Based on these four phase maps,the absolute 3D displacement gradients are determined.This means of absolute 3D displacement gradient measurement effectively improves the measurement capability of digital shearography and expands its application range.

1.Introduction Gravity data provide significant insights on ice mass balance and geological structure studies in Antarctica,which is the largest ice sheet on the earth(Xu,2001;Alley et al.,2005).Since the 1950s,the gravity ties have been introduced to Antarctica from Australia and South America(Sledzinski,2001;Amalvict,2004),which adopted the approach of relative gravity measurements with pendulum or spring gravimeters.absolute gravity measurements were later conducted in Antarctica by several countries to provide gravity bases and to derive its dynamic change with the technique advance in gravimeter instrumentation.By 2009,25 absolute gravity surveys had been carried out at 14 stations in Antarctica(Mäkinen et al.,2007;E et al.,2007,2011;Kazama et al.,2013).

Kai-Xin-San consists of Ginseng Radix, Polygalae Radix, Acori Tatarinowii Rhizoma, and Poria at a ratio of 3:3:2:2. Kai-Xin-San has been widely used for the treatment of emotional disorders in China. However, no studies have identified the key proteins implicated in response to Kai-Xin-San treatment. In this study, rat models of chronic mild stress were established using different stress methods over 28 days. After 14 days of stress stimulation, rats received daily intragastric administrations of 600 mg/kg Kai-Xin-San. The sucrose preference test was used to determine depression-like behavior in rats, while isobaric tags were used for relative and absolute quantitation-based proteomics to identify altered proteins following Kai-Xin-San treatment. Kai-Xin-San treatment for 2 weeks noticeably improved depression-like behaviors in rats with chronic mild stress. We identified 33 differentially expressed proteins: 7 were upregulated and 26 were downregulated. Functional analysis showed that these differentially expressed proteins participate in synaptic plasticity, neurodevelopment, and neurogenesis. Our results indicate that Kai-Xin-San has an important role in regulating the key node proteins in the synaptic signaling network, and are helpful to better understand the mechanism of the antidepressive effects of Kai-Xin-San and to provide objective theoretical support for its clinical application. The study was approved by the Ethics Committee for Animal Research from the Chinese PLA General Hospital(approval No. X5-2016-07) on March 5, 2016.

Sensory and motor nerve fibers of peripheral nerves have different anatomies and regeneration functions after injury. To gain a clear understanding of the biological processes behind these differences, we used a labeling technique termed isobaric tags for relative and absolute quantitation to investigate the protein profiles of spinal nerve tissues from Sprague-Dawley rats. In response to Wallerian degeneration, a total of 626 proteins were screened in sensory nerves, of which 368 were upregulated and 258 were downregulated. In addition, 637 proteins were screened in motor nerves, of which 372 were upregulated and 265 were downregulated. All identified proteins were analyzed using the Gene Ontology and Kyoto Encyclopedia of Genes and Genomes analysis of bioinformatics, and the presence of several key proteins closely related to Wallerian degeneration were tested and verified using quantitative real-time polymerase chain reaction analyses. The differentially expressed proteins only identified in the sensory nerves were mainly relevant to various biological processes that included cell-cell adhesion, carbohydrate metabolic processes and cell adhesion, whereas differentially expressed proteins only identified in the motor nerves were mainly relevant to biological processes associated with the glycolytic process, cell redox homeostasis, and protein folding. In the aspect of the cellular component, the differentially expressed proteins in the sensory and motor nerves were commonly related to extracellular exosomes, the myelin sheath, and focal adhesion. According to the Kyoto Encyclopedia of Genes and Genomes, the differentially expressed proteins identified are primarily related to various types of metabolic pathways. In conclusion, the present study screened differentially expressed proteins to reveal more about the differences and similarities between sensory and motor nerves during Wallerian degeneration. AIM:To measure the prognostic significance of absolute monocyte count/absolute lymphocyte count prognostic score(AMLPS) in patients with gastric cancer.METHODS:We retrospectively examined the combination of absolute monocyte count(AMC) and absolute lymphocyte count(ALC) as prognostic variables in a cohort of 299 gastric cancer patients who underwent surgical resection between 2006 and 2013 and were followed at a single institution.Both AMC and ALC were dichotomized into two groups using cut-off points determined by receiving operator characteristic curve analysis.An AMLPS was generated,which stratified patients into three risk groups:low risk(both low AMC and high ALC),intermediate risk(either high AMC or low ALC),and high risk(both high AMC and low ALC).The primary objective of the study was to validate the impact of AMLPS on both disease-free survival(DFS) and overall survival(OS),and the second objective was to assess the AMLPS as an independent prognostic factor for survival in comparison with known prognostic factors.RESULTS:Using data from the entire cohort,the most discriminative cut-off values of AMC and ALC selected on the receiver operating characteristic curve were 672.4/μL and 1734/μL for DFS and OS.AMLPS risk groups included 158(52.8%) patients in the lowrisk,128(42.8%) in the intermediate-risk,and 13(4.3%) in the high-risk group.With a median followup of 37.2 mo(range:1.7-91.4 mo),five-year DFS rates in the low-,intermediate-,and high-risk groups were 83.4%,78.7%,and 19.8%,respectively.And fiveyear OS rates in the low-,intermediate-,and high-risk groups were 89.3%,81.1%,and 14.4%,respectively.On multivariate analysis performed with patient- and tumor-related factors,we identified AMLPS,age,and pathologic tumor-node-metastasis stage as the most valuable prognostic factors impacting DFS and OS.CONCLUSION:AMLPS identified patients with a poor DFS and OS,and it was independent of age,pathologic stage,and various inflammatory markers.

Circulating tumor DNA(ctDNA)detection has found widespread applications in tumor diagnostics and treatment,where the key is to obtain accurate quantification of ctDNA.However,this remains challenging due to the issue of background noise associated with existing assays.In this work,we developed a synthetic gene circuit-based assay with multilevel switch(termed CATCH)for background-free and absolute quantification of ctDNA.The multilevel switch combining a small transcription activating RNA and a toehold switch was designed to simultaneously regulate transcription and translation processes in gene circuit to eliminate background noise.Moreover,such a multilevel switch-based gene circuit was integrated with a Cas9 nickase H840A(Cas9n)recognizer and a molecular beacon reporter to form CATCH for ctDNA detection.The CATCH can be implemented in one-pot reaction at 35°C with virtually no background noise,and achieve robust absolute quantification of ctDNA when integrated with a digital chip(i.e.,digital CATCH).Finally,we validated the clinical capability of CATCH by detecting drug-resistant ctDNA mutations from the plasma of 76 non-small cell lung cancer(NSCLC)patients,showing satisfying clinical sensitivity and specificity.We envision that the simple and robust CATCH would be a powerful tool for next-generation ctDNA detection.

Passively mode-locked fiber lasers emit femtosecond pulse trains with excellent short-term stability. The quantum-limited timing jitter of a free running femtosecond erbium-doped fiber laser working at room temperature is considerably below one femtosecond at high Fourier frequency. The ultrashort pulse train with ultralow timing jitter enables absolute time-of-flight measurements based on a dual-comb implementation, which is typically composed of a pair of optical frequency combs generated by femtosecond lasers. Dead-zone-free absolute distance measurement with sub-micrometer precision and kHz update rate has been routinely achieved with a dual-comb configuration, which is promising for a number of precision manufacturing applications, from large step-structure measurements prevalent in microelectronic profilometry to three coordinate measurements in large-scale aerospace manufacturing and shipbuilding. In this paper, we first review the sub-femtosecond precision timing jitter characterization methods and approaches for ultralow timing jitter mode-locked fiber laser design. Then, we provide an overview of the state-of-the-art dual-comb absolute ranging technology in terms of working principles, experimental implementations, and measurement precisions. Finally, we discuss the impact of quantum-limited timing jitter on the dual-comb ranging precision at a high update rate. The route to highprecision dual-comb range finder design based on ultralow jitter femtosecond fiber lasers is proposed.

An absolute cryogenic radiometer(ACR) with a detachable optical window was designed and built for high accuracy optical radiant power measurement and photodetector spectral responsivity calibration. The ACR receiver is an electroplated pure copper cavity with a 50-μm-thick wall and inner surface coated with a specular black polymer material mixed with highly dispersible carbon nanotubes. The absorptivity of the cavity receivers was evaluated to be ≥0.9999 in the 250 nm–16 μm wavelength range and ≥0.99995 in 500 nm–16 μm. The cavity receiver works at the temperature of ~5.2 K with nanowatt-level noise-equivalent power. The relative standard uncertainty is 0.041% for the measurement of ~100 μW optical radiant power(250 nm–16 μm) and 0.015% for^1 m W(500 nm–16 μm).
