On February 1, 2016, the World Health Organization declared that the cluster of microcephaly cases and other neurological disorders constitute public health emergency of international concern. Furthermore, few studies demonstrated that there was an increased evidence of causal relationship of zika virus(zikaV) infection and microcephaly, birth abnormalities and neurological disorders such as Guillain–Barre′ *** transmission occurs mainly by the bite of infected mosquitos(Aedes species), but there are also reports that infections could occur via the placenta, breast milk, saliva,blood transfusion and sex. This article reviews the global efforts, progress in scientific research to understand the pathogenesis of zikaV infection & disease, clinical presentations, congenital transmission and autoimmune neurological disorders. The paper further explores the knowledge gaps, future priority research agenda for strategic response including vector control and prevention. We conducted a systematic literature review to synthesise available evidence on zikaV infection and its vector and host interaction from electronic databases.

zika virus （ZIKV） is a mosquito-borne pathogen from the family Flaviviridae, and infection of humans with ZIKV through mosquito bites may result in a disease known as zika fever. Most zika fever cases are asymptomatic and thus go unreported, but if symptoms do appear, they are usually mild in nature （fever, joint pain, headache and macu- Iopapular rash）, and self-resolve within one week （Chen and Hamer 2016）. No deaths have ever been attributed to zika fever; however, when a woman is infected with ZIKV during pregnancy, vertical transmission of the virus may result in microcephaly in the offspring （Rasmussen, Jamieson et al., 2016）. ZIKV infections in adults are associated with episodes of Guillain-Barr6 syndrome, an autoimmune disorder of the peripheral nervous system, resulting in rapid-onset muscle weakness （Cao-Lormeau, Blake et al. 2016）.

As we know more about zika virus(ZIKV), as well as its linkage to birth defects(microcephaly) and autoimmune neurological syndromes, we realize the importance of developing an efficient vaccine against it. zika virus disease has affected many countries and is becoming a major public health concern. To deal with the infection of ZIKV, plenty of experiments have been done on selection of neutralizing antibodies that can target the envelope(E) protein on the surface of the virion. However, the existence of antibody-dependent enhancement(ADE) effect might limit the use of them as therapeutic candidates. In this review, we classify the neutralizing antibodies against ZIKV based on the epitopes and summarize the resolved structural information on antibody/antigen complex from X-ray crystallography and cryo-electron microscopy(cryo-EM), which might be useful for further development of potent neutralizing antibodies and vaccines toward clinical use.

Objective: To investigate the genetic diversity of zika virus(ZIKV) and the relationships existing among these circulating viruses worldwide. To evaluate the genetic polymorphisms harboured from ZIKV that can have an influence on the virus circulation. Methods: Three different ZIKV dataset were built. The first dataset included 63 E gene sequences, the second one 22 NS3 sequences and the third dataset was composed of 108 NS5 gene sequences. Phylogenetic and selective pressure analysis was performed. The edited nucleic acid alignment from the Envelope dataset was used to generate a conceptual translation to the corresponding peptide sequences through UGene software. Results: The phylogeographic reconstruction was able to discriminate unambiguously that the Brazilian strains are belonged to the Asian lineage. The structural analysis reveals instead the presence of the Ser residue in the Brazilian sequences(however already observed in other previously reported ZIKV infections) that could suggest the presence of a neutralization-resistant population of viruses. Conclusions: Phylogenetic, evolutionary and selective pressure analysis contributed to improve the knowledge on the circulation of ZIKV.

zika virus(ZIKV) causes rash, moderate fever, conjunctivitis, and arthralgia, and has serious connection with neurological complications;therefore, it is a major threat to public health. A rapid and supersensitive method for detecting anti-ZIKV antibodies in humans and animals is thus urgently required. Here, we report an NS1-based luciferase immunosorbent assay(LISA), developed to detect ZIKV-specific IgG. Fusion proteins including a reporter Nano-luciferase(NLuc) and various fragments of ZIKV NS1 protein were expressed in 293 T cells. LISA was performed using the above cell lysates containing the expressed fusion proteins. Sample panels of humans and animals infected with ZIKV were examined for sensitivity of LISA, relative to those of ZIKV RT-PCR, commercial NS1-based ELISA, and micro-neutralization(MN) *** and potential cross-reactivity were also evaluated using various convalescent serum samples derived from patients infected with dengue virus(DENV), Japanese encephalitis virus(JEV), and hepatitis C virus(HCV). Results indicated the optimal antigenic domain for anti-ZIKV IgG detection was located within 172–352 amino acids(aa) of ZIKV NS1 protein. NS1-based LISA performs better than commercial ELISA in anti-ZIKV Ig G detection. LISA was shown to be at least fourfold more sensitive than commercial ELISA, and could detect anti-ZIKV Ig G in various animal hosts without the need of species-specific labeled antibody. This novel assay is potentially useful for the rapid and sensitive detection of anti-ZIKV IgG in human and animal samples.

The Aedes aegypti mosquito which transmits zika virus(as well as dengue, chikungunya and yellow fever) represents a high risk for global transmission. This virus comes from Africa, the zika forest in Uganda, where it was discovered in 1947 in a rhesus monkey. In May 2015, the first local cases were recorded in Brazil, surpassing 1.5 million cases in December of the same year. By March 2016, local transmission of zika was recognized in 34 countries. Its clinical condition is similar to dengue febrile illness, although *** final geographical distribution area is constantly expanding. Recently, it has been associated with cases of Guillain-Barre′ syndrome in Brazil, Colombia, El Salvador,Venezuela and Suriname. Microcephaly was documented in Brazil. This article discusses some factors that contributed to the spread of zika virus in South America. Climate change associated with the events of the phenomenon of "El Ni?o" is also analyzed. The biggest concern is how quickly zika is spreading around the world and that it could be far more dangerous than previously thought. zika virus infection, by its explosive potential,has every chance of becoming a global pandemic.

zika virus infection is a new problematic virus infection that becomes the present public health problem. Now this mosquito borne infectious disease can be seen worldwide and can cause dengue-like infection. In addition, it can also induce transplacental infection and result in congenital neurological defect. To prevent this infection, there is still no specific vaccine. To find a new vaccine, finding epitope is the first step. Here, the authors report the study to find epitope within zika virus *** to this study, the appropriate epitopes can be seen. This is the fi rst world report on epitope finding for zika virus. The data can be useful for further vaccine development.

zika virus(ZIKV) was discovered more than half a century ago,recently it has gained unprecedented attention by the global health *** 2007,only 14 cases of human ZIKV infections were reported around the globe,while during the current outbreak,estimated cases mounted to approximately 1.5 million in Brazil alone,the virus was disseminated to wider South-American territories and travel-associated ZIKV infections were reported in USA,Europe and recently in *** infections remain asymptomatic in approximately 80% of the individuals,and no anti-viral treatments were ***,neurological complications associated with the infections,such as infant microcephaly and Guillain-Barré syndrome are major cause of the ***,based on small numbers of cases,existing evidence strongly supports an exclusive link of viral infection and observed neurological ***,much work remains to assign exact numbers of complications caused by *** its structural attributes ZIKV shows remarkable resemblance with dengue virus and West-Nile ***,genomes of different ZIKV strains have already been decoded;role of the viral components in infection process and particularly pathogenesis of the disease remain widely *** vulnerable areas,most viable strategy to ensure public health safety is vector control and enhanced public awareness about the transmission of the disease.
