Molecular analyses of bacterioplankton communities with highly abundant Vibrio clades: a case study in Bohai Sea coastal waters

We investigated the bacterioplankton abundance, community composition and the associated Vibrio clades of natural seawater in Bohai Sea coastal waters. Seawater samples (10 L in triplicate) were collected at 0.5, 3, and 5 m depths near the coastal aquaculture zone of the Bohai Sea on May 12, 2016. Real-time PCR and 16S rRNA gene amplicon high-throughput sequencing methods were employed by which 485 operational taxonomic units (OTUs) at a 97% sequence similarity level were generated. Gammaproteobacteria, Alphaproteobacteria, and Bacteroidetes were the most abundant groups, accounting for 49.5%, 23.5%, and 18.9% of the total assemblage, respectively. Obvious variations in Pseudoalteromonas, Vibrio, and Octadecabacter , which were the most abundant genera, could be observed among diff erent samples. Notably, the results of Vibrio -specifi c real-time PCR indicated that Vibrio had extremely high 16S rRNA gene copy numbers. The 16S rRNA gene sequencing results across all the samples also indicated that they occupied a large proportion of the total assemblage. Both the alpha diversity and major bacterioplankton group Pseudoalteromonas had significant correlations with the concentration of PO4^3-. Overall, studies on bacterioplankton communities with highly abundant Vibrio clades can provide interesting insight into the microbial function and health assessment of the Bohai Sea coastal ecosystem.

利用琼胶酶生产琼胶寡糖具有良好的应用前景,然而目前产琼胶酶菌株的性状不稳定、酶活力低等因素限制了琼胶酶的工业化应用。从龙须菜表面筛选出1株具有高琼胶酶活力的菌株A8,利用VITEK 2 GN微生物鉴定系统和16S rRNA基因序列分析,分别进行生理生化和分子生物学鉴定,确定该菌为Vibrio fluvialis A8。通过单因素试验和正交试验确定该菌的最适产酶培养基为:在人工海水中添加2 g/L琼脂、3 g/L半乳糖、3 g/L酵母浸粉、5 g/L Na Cl;最适培养条件为:接种量2%、温度20℃、p H 7.0。优化后发酵液琼胶酶的活力为21.80U/m L,是优化前的4.05倍。Pearson相关性分析表明,菌株生长对其产酶有正相关影响(P<0.01)。采用(NH4)2SO4分级沉淀和DEAE阴离子交换层析对琼胶酶进行分离纯化,琼胶酶的纯化倍数为3.26,比活力为141.52 U/mg,回收率为15.21%。SDS-PAGE凝胶电泳分析显示,琼胶酶的分子质量约为37 k Da。

Objective:To investigate the isolation of enterobacteria associated with Macrobrachium amazonicum(M.amazonicum) farming and evaluate the in vitro antimicrobial susceptibility of Vibrio strains.Methods:Strains were isolated from female M.amazonicum prawns and environmental and hatchery water.Biochemical assays were used to identify bacterial genera and those belonging to the genus Vibrio were submitted to further analyses for species identification,through Vitek 2 automated system and scrotyping.Susceptibility test was performed according to Clinical Laboratory Standards Institute.Results:The following genera of enlerobacteria were recovered:Enterobacter(n=11),Citrobacter(n=10),Proteus(n=2),Serratia(n=2),Kluyvera(n=2),Providencia(n=2),Cedecea(n=1),Escherichia(n=1).Edwardsiella(n=1) and BulliauxeUa(n=1).As for Vibrio,three species were identified:Vibrio cholerae non-O1/non-O139(n=4).Vibrio vulnificus(V.vulnificus)(n=1) and Vibrio mimicus(n=1).Vibrio spp.showed minimum inhibitory concentrations values within the susceptibility range established by Clinical Laboratory Standards Institute for almost all antibiotics,except for V.vulnificus,which presented intermediate profile to ampicillin.Conclusions:Enterobacteria do not seem to be the most important pathogens associated with M.amazonicum farming,whereas the recovery of Vibrio spp.from larviculture,with emphasis on Vibrio cholerae and V.vulnificus,deserves special attention due to their role as potentially zoonotic aquaculture-associated pathogens.Furthermore,the intermediate susceptibility of V.vulnificus to ampicillin reflects the importance of monitoring drug use in prawn farming.

The lysogenic phage CTXΦ of Vibrio cholerae can transfer the cholera toxin gene both horizontally(inter-strain) and vertically(cell proliferation). Due to its diversity in form and species, the complexity of regulatory mechanisms, and the important role of the infection mechanism in the production of new virulent strains of V.cholerae, the study of the lysogenic phage CTXΦ has attracted much attention. Based on the progress of current research, the genomic features and their arrangement, the host-dependent regulatory mechanisms of CTXΦ phage survival, proliferation and propagation were reviewed to further understand the phage's role in the evolutionary and epidemiological mechanisms of V. cholerae.

为了探究抗生素对发光菌的Hormesis效应,选择费氏弧菌（Vibrio fischeri,V.fischeri）为受试生物,氯霉素为研究对象,测定了0~24h下氯霉素对V.fischeri的发光强度（Hv）及生长量（OD_（600））的作用,探讨氯霉素对V.fischeri的低浓度促进效应及其可能机制.结果表明,氯霉素对V.fischeri的发光强度具有明显的促进作用,且仅出现在细菌生长的延滞期.根据V.fischeri的发光机制,结合Gaussian量子化学计算结果,即氯霉素的最正氢电荷为0.244,远大于烟酰胺腺嘌呤二核苷酸（NADH）的最正氢电荷0.135,提出氯霉素通过提供质子促进荧光反应,进而产生Hormesis效应的可能机制.

以海洋弧菌(Vibrio sp.QD-5)的基因组为模板,使用设计的褐藻胶裂解酶引物进行PCR扩增。将目的基因克隆至pet-22b(+)载体后进行测序。测序结果显示,基因aly-Ⅱ的大小为2 170bp,预测编码含有723个氨基酸的蛋白质。对该蛋白质进一步进行了生物信息学分析。利用ProtParam和ProtScale对蛋白质的理化性质进行分析,结果表明,蛋白质Aly-Ⅱ的理论分子质量为85.6kDa,理论等电点为5.1,并且是一种亲水性蛋白。利用软件MEG 6.0对蛋白质Aly-Ⅱ进行系统发育分析,结果显示,蛋白质Aly-Ⅱ很可能是PL-17家族的褐藻胶裂解酶。利用同源建模法构建蛋白质Aly-Ⅱ的三维结构,Aly-Ⅱ含有2个结构域,分别为(α/α)6toroid结构域和β-sheet结构域。

Members of the Vibrionaceae family are predominantly fast-growing and halophilic microorganisms that have captured the attention of researchers owing to their potential applications in rapid biotechnology.Among them,Vibrio alginolyticus FA2 is a particularly noteworthy halophilic bacterium that exhibits superior growth capability.It has the potential to serve as a biotechnological platform for sustainable and eco-friendly open fermentation with seawater.To evaluate this hypothesis,we integrated the N-acetylglucosamine(GlcNAc)pathway into V.alginolyticus FA2.Seven nag genes were knocked out to obstruct the utilization of GlcNAc,and then 16 exogenous gna1s co-expressing with EcglmS were introduced to strengthen the flux of GlcNAc pathway,respectively.To further enhance GlcNAc production,we fine-tuned promoter strength of the two genes and inactivated two genes alsS and alsD to prevent the production of acetoin.Furthermore,unsterile open fermentation was carried out using simulated seawater and a chemically defined medium,resulting in the production of 9.2 g/L GlcNAc in 14 h.This is the first report for de-novo synthesizing GlcNAc with a Vibrio strain,facilitated by an unsterile open fermentation process employing seawater as a substitute for fresh water.This development establishes a basis for production of diverse valuable chemicals using Vibrio strains and provides insights into biomanufacture.

以Vibrio fischeri为模式生物,3类群体感应抑制剂(呋喃酮、吡咯酮和吡咯)与典型磺胺类抗生素为研究对象,测定了群体感应抑制剂与磺胺类抗生素联合急性毒性效应,通过群体感应抑制剂与信号分子竞争结合LuxR的实验和结合能Ebinding的测定,初步探讨了其联合毒性作用机制.结果显示,呋喃酮类化合物与磺胺类抗生素的二元等毒性比下的联合效应表现为协同与加和,吡咯酮类、吡咯类化合物与磺胺类抗生素的二元等毒性比下的联合效应分别表现为加和与拮抗;它们与LuxR的竞争结合力的差异,可能是它们与磺胺类抗生素等毒性比下的联合效应不同的原因.

Objective:To identify a potential bacterium which produces antimicrobial peptide(Vibriocin),and its purification,characterization and production optimization.The bacteria subjected in the study were isolated from a highly competitive ecological niche of mangrove ecosystem.Methods:The bacterium was characterized by phenotype besides 16S rRNA gene sequence analysis.The antibacterial activity was recognised by using agar well diffusion method.The Vibriocin was purified using ammonium sulphate precipitation,butanol extraction,gel filtration chromatography,ion-exchange chromatography and subsequently,by HPLC.Molecular weight of the substance identified in SDS-PAGE.Production optimization performed according to Taguchi's mathematical model using 6 different nutritional parameters as variables.Results:The objective bacterium was identified as Vibrio parahaemolyticm.The Vibriocin showed 18 KDa of molecular mass with mono peptide in nature and highest activity against pathogenic Vibrio harveyi.The peptide act stable in a wide range of pH,temperature.UV radiation,solvents and chemicals utilized.An overall^20%of Vibriocin production was improved,and was noticed that NaCl and agitation speed played a vital role in secretion of Vibriocin.Conclusion:The Vibriocin identified here would he an effective alternative for chemically synthesized drugs for the management of Vibrio infections in mariculture industry.
