vestigial suppresses apoptosis and cell migration in a manner dependent on the level of JNK-Caspase signaling in the Drosophila wing disc

The Decapentaplegic(Dpp)and Wingless(Wg)signal pathways play important roles in numerous biological processes in *** Drosophila vestigial(vg)gene is selectively required for wing imaginal disc cell proliferation,which is essential for the formation of the adult wing and halter structures,and is regulated by Dpp and Wg *** a Drosophila invasion model of wing epithelium,we showed herein that inhibition of Dpp or Wg signaling promoted cells to migrate across the cell lineage restrictive anterior/posterior(A/P)compartment *** downstream of both Dpp and Wg signaling,vg can block cell migration induced by loss of either *** addition,suppression of vg is sufficient to induce cell migration across the A/P *** analysis revealed potential downstream genes involved in the cell migration after suppressing vg in the wing *** further demonstrated that the c-Jun N-terminal kinase(JNK)signaling promoted cell migration induced by vg suppression by upregulating Caspase *** together,our results revealed the requirement ofVg for suppressing cell migration and clarified how developmental signals collaborate to stabilize cells along the compartment boundary.

BACKGROUND vestigial like family member 3(VGLL3)is associated with the prognosis of epithelial ovarian cancer and soft tissue sarcoma,but its role in gastric cancer(GC)is *** To explore the expression pattern and clinical significance of VGLL3 in *** Integrative analysis was performed on the GC transcriptome profiles and survival information deposited in the ONCOMINE,GEPIA,and ONCOLNC *** expression levels of VGLL3 mRNA and protein were analyzed in the freshly resected tumor and normal gastric tissues from GC patients by quantitative RT-PCR and Western blot,*** addition,the in situ expression of VGLL3 in the GC tissues was determined by immunohistochemistry(IHC),and the patients were accordingly classified into the high and low expression *** correlation of VGLL3 expression status with patient prognosis was then determined by univariate and multivariate Cox regression *** Analysis of the ONCOMINE and GEPIA databases showed that VGLL3 was significantly up-regulated in GC tissues(P=0.003),and associated with the tumor TNM stage(P=0.0163).The high VGLL3 expression group had a significantly worse prognosis compared to the low expression group,as per both GEPIA(P=0.0057)and ONCOLNC(P=0.01).The bioinformatics results were validated by the significantly higher VGLL3 mRNA and protein levels in the GC tissues compared to the adjacent normal tissues(P<0.001)in a cohort of 30 GC ***,high in situ expression of VGLL3 protein was associated with more advanced N and TNM stages and HER2 mutation(P<0.05)in a cohort of 172 ***-Meier analysis showed that the high VGLL3 expression group had a worse prognosis compared to the low expression group(P=0.019).Multivariate analysis showed that VGLL3 expression status was an independent risk factor for *** addition,the prognostic risk model nomogram showed that VGLL3 was the most important indicator,with an area under the receiver operating character

The glume is an organ of the maize spikelet and plays important roles in anther and kernel *** glume1(Vg1)is a classic mutant associated with ligule and glume *** we report the phenotypic characterization,fine mapping,and candidate gene analysis of the Vg1 ***1 is a semi-dominant and pleiotropic gene,and also affects plant height,ear height,and tassel ***1 ligule degeneration begins at the first leaf,and the Vg1 tassel and ear can be distinguished from those of wild-type plants when their lengths reach respectively 55 mm and 51 *** a BC3 mapping population of 11,445 plants,we delimited the Vg1 functional site to an interval of 7.4 kb,flanked by the markers InDelLM and CRM6.A putative cyclopropane fatty-acid synthase gene(ZmCPA-FAS1)was hypothesized to underlie the mutant *** detected a Helitron insertion in the sixth intron of *** presence caused abnormal alternative splicing of ZmCPA-FAS1 that conferred new characteristics on the Vg1 *** findings are a basis for further discovery of the molecularmechanism underlying glume development and a potential guide formaize breeding of small-glume varieties,especially sweet corn breeding.

Fosmid library construction and screening for the maize mutant gene vestigial glume 1

The maize mutant gene vestigial glume 1(Vg1) has been fine-mapped to a narrow region by map-based cloning and the candidate gene for Vg1 spanned 19.5 kb. Here we report Vg1 genomic fosmid library construction and screening. The fosmid library of Vg1 consisted of574,000 clones with an average insert size of 36.4 kb, representing 7.9-fold coverage of the maize genome. Fosmid stability assays indicated that clones were stable during propagation in the fosmid system. Using Vg1 candidate gene-specific primers, a positive clone was successfully identified. This discovery will pave the way for identifying the function of Vg1 in maize development.

Nanocarrier-delivered dsRNA suppresses wing development of green peach aphids

RNA interference(RNAi)has developed rapidly as a potential“green”pest management *** present,most studies have focused on the screening of aphid lethal genes,whereas only a few studies have been conducted on wing development,which is crucial for aphid migration and plant-virus ***,the Myzus persicae genes vestigial(vg)and Ultrabithorax(Ubx)related to wing development,were *** two genes were expressed in various tissues of 3rd-instar winged *** mRNA level of vg was high in 3rd-instar nymphs,whereas the expression level of Ubx was high in *** nanocarrier-mediated delivery system delivered double-stranded RNAs for aphid RNAi using topical and root *** expression levels of vg and Ubx were downregulated by 44.0%and 36.5%,respectively,using the topical *** simultaneous RNAi of the two target genes caused 63.3%and 32.2%wing aberration rates using topical and root applications,*** current study provided a promising method for controlling aphid migration to alleviate the spread of insect transmitted plant diseases.
