ABSTRACTHuman placenta-derived mononuclear cells (MNC) were isolated by a Percoll density gradient and cultured in mesen-chymal stem cell (MSC) maintenance medium. The homogenous layer of adherent cells exhibited a typical fibroblast-like morphology, a large expansive potential, and cell cycle characteristics including a subset of quiescent cells. In vitrodifferentiation assays showed the tripotential differentiation capacity of these cells toward adipogenic, osteogenic andchondrogenic lineages. Flow cytometry analyses and immunocytochemistry stain showed that placental MSC was ahomogeneous cell population devoid of hematopoietic cells, which uniformly expressed CD29, CD44, CD73, CD105,CD166, laminin, fibronectin and vimentin while being negative for expression of CD31, CD34, CD45 and α-smoothmuscle actin. Most importantly, immuno-phenotypic analyses demonstrated that these cells expressed class I majorhistocompatibility complex (MHC-I), but they did not express MHC-II molecules. Additionally these cells could sup-press umbilical cord blood (UCB) lymphocytes proliferation induced by cellular or nonspecific mitogenic stimuli. Thisstrongly implies that they may have potential application in allograft transplantation. Since placenta and UCB arehomogeneous, the MSC derived from human placenta can be transplanted combined with hematopoietic stem cells(HSC) from UCB to reduce the potential graft -versus-host disease (GVHD) in recipients.

umbilical cord blood(UCB)is an efficient and valuable source of hematopoietic stem cells(HSCs)for transplantation.In addition to HSCs it harbours low amounts of mesenchymal stem cells(MSCs).No single marker to identify cord blood-derived stem cells,or to indicate their multipotent phenotype,has been characterized so far.SSEA-3 and-4 are cell surface globoseries glycosphingolipid epitopes that are commonly used as markers for human embryonic stem cells,where SSEA-3 rapidly disappears when the cells start to differentiate.Lately SSEA-3 and-4 have also been observed in MSCs.As there is an ongoing discussion and variation of stem-cell markers between laboratories,we have now comprehensively characterized the expression of these epitopes in both the multipotent stem-cell types derived from UCB.We have performed complementary analysis using gene expression analysis,mass spectrometry and immunochemical methods,including both flow cytometry and immunofluoresence microscopy.SSEA-4,but not SSEA-3,was expressed on MSCs but absent from HSCs.Our findings indicate that SSEA-3 and/or-4 may not be optimal markers for multipotency in the case of stem cells derived from cord blood,as their expression may be altered by cell-culture conditions.

Background Pancreatic islet cell transplantation is an effective approach to treat type 1 diabetes. However, this therapy is not widely used because of the severe shortage of transplantable donor islets. This study investigated whether mesenchymal stem cells （MSCs） derived from human umbilical cord blood （UCB） could be transdifferentiated into insulin producing cells in vitro and the role of extracellular matrix （ECM） gel in this procedure. Methods Human UCB samples were collected and MSCs were isolated. MSCs specific marker proteins were analyzed by a flow cytometer, The capacities of osteoblast and adipocyte to differentiate were tested. Differentiation into islet like cell was induced by a 15-day protocol with or without ECM gel. Pancreatic characteristics were evaluated with immunofluorescence, reverse transcription polymerase chain reaction （RT-PCR） and flow cytometry. Insulin content and release in response to glucose stimulation were detected with chemiluminescent immunoassay system. Results Sixteen MSCs were isolated from 42 term human UCB units （38%）. Human UCB-MSCs expressed MSCs specific markers and could be induced in vitro into osteoblast and adipocyte. Islet like cell clusters appeared about 9 days after pancreatic differentiation in the inducing system with ECM gel. The insulin positive cells accounted for （25.2~3.4）% of the induced cells. The induced cells expressed islet related genes and hormones, but were not very responsive to glucose challenge. When MSCs were induced without ECM gel, clusters formation and secretion of functional islet proteins could not be observed, Conclusions Human UCB-MSCs can differentiate into islet like cells in vitro and ECM gel plays an important role in pancreatic endocrine cell maturation and formation of three dimensional structures.

OBJECTIVE： Human umbilical cord blood-denved mesenchymal stem cells （HUCB-derived MSCs） can differentiate into neuron-like cells, which can be used to treat some central nervous system （CNS） diseases. To investigate the factors, which can induce HUCB-derived MSCs to differentiate into neuron-like cells, so as to find effective methods for future clinical application.DATA SOURCES ： Using the key terms ＂human umbilical cord blood＂ combined with ＂mesenchymal stem cells,neuron-like cells, neural cells＂ respectively, the relevant articles in English published dunng the period from January 1999 to June 2006 were searched from the Medline database. Meanwhile, relevant Chinese articles published from January 1999 to June 2006 were searched using the same key terms.STUDY SELECTION ： All articles associated with the differentiation from human umbilical cord blood into neuron-like cells were selected firstly. Then the full texts were looked up by searching Ovid medical joumals full-text database and Elsevier Electncal Joumals Full-text Database. Articles with full experiments, enrolled in inducible factors or involved inducible mechanism were retrieved.DATA EXTRACTION ： Among 119 collected correlative articles, 29 were involved and 90 were excluded.DATA SYNTHESIS ： The inducible factors of HUCB-derived MSCs differentiating into neuron-like cells included renal endothelial growth factors, fibroblasts, β- mercaptoethanol, dimethyl sulfoxide, butyl hydroxyl anisol, brain-derived neurotrophic factor, Danshen, retinoic acid, sodium ferulate and so on, but its mechanism was unclear.CONCLUSION： Human umbilical cord blood-derived MSCs can differentiate into neuron-like cells, with varied inductors.

Objective: In this study, we aimed to investigate umbilical cord blood CD33 and erythropoietin (EPO) levels of pregnants with abnormal umbilical and uterine artery doppler waveforms and to compare with normal pregnancies. Materials and Methods: Total 40 pregnant women were included in this study. Of these 40 women, while 20 patients had abnormal umbilical and uterine artery doppler waveforms, the other 20 patients had normal umbilical and uterine artery doppler waveforms. After the delivery, blood samples were taken from umbilical artery of double clemped umbilical cord for blood gas parameters, EPO and CD33 levels. Sociodemographic findings, antepartum, intrapartum test results, labor and delivery characteristics and newborn examination results were recorded. blood gas parameters, EPO and CD33 levels between groups were analyzed. Mann-Whitney U test and t-test were used as statistical methods. Results: There were no differences between parity, gestational ages and newborn weights of the groups. cord blood CD33 and EPO levels of group with abnormal umbilical and uterine artery doppler waveforms were significantly higher than group with normal umbilical and uterine artery doppler waveforms (p blood CD33 levels for cord blood transplantation especially during last years can also be used with preference of cord blood with abnormal doppler findings.

[ Objective] The study aimed to understand the status of piglet diarrhea virus in large-scale farms in surrounding areas of Beijing. [ Method] By collecting 570 umbilical cord blood samples of diseased pigs, nucleic acids of porcine reproductive and respiratory syndrome virus （PRRSV） , classical swine fever virus （CSFV）, porcine pseudorabies virus （PRV）, porcine circovirus （PCV）, porcine epidemic diarrhea virus （PEDV）, transmissible gastroenteritis virus （TGEV）, and porcine rotavirus （RV） were detected by PCR or RT-PCR assay. [Result] The detection rates of PRRSV, PCV-2, PEI）V, PPV, TGEV, PRV, RV and CSFV were 38.7%, 29.6%, 21.8%, 13.03%, 5.6%, 4.2%, 3.8% and 1.7%, respectively. [ Conclusion] The major pathogens were PRRSV, PCV-2, PEDV and PPV, and mixed infection was serious.

Objective To explore the relationship of lead levels in umbilical cord blood and foremilk with the neurobehavioral development of neonates with intrauterine exposure to lead at low level. Methods Lead levels in umbilical cord blood,which could be determined as indicators for intrauterine exposure to lead, were determined for 170 randomly selected neonates delivered at Nanjing Maternity and Child Health Care Hospital. Neonatal behavioral neurological assessment(NBNA) total score were determined as effect index.A descriptive analysis was applied to evaluate their correlations.The neonates were divided into two groups of high- and low-exposure based on the cutoff lead values of 10μg/dl. Results There were significant differences in scores of NBNA and initiative muscle tension with Pumbilical cord blood,and the score of the primitive reflex showed difference with P<0.01,while no difference could be observed in the behavioral capability,passive muscle tension and general evaluation. Conclusion The lead in the foremilk could transfer to the neonates,resulting in adverse effect on the early neurobehavioral development of neonates.

umbilical cord blood(UCB) transplant is a therapeutic option for both pediatric and adult patients with a variety of hematologic diseases such as several types of blood cancers, myeloproliferative disorders, genetic diseases, and metabolic disorders. However, the level of cellular heterogeneity and diversity of nucleated cells in the UCB has not yet been assessed in an unbiased and systemic fashion. In the current study, nucleated cells collected from UCB donors were subjected to single-cell RNA sequencing, a technology enabled simultaneous profiling of the gene expression signatures of thousands of cells, generating rich resources for further functional studies. Here, we report the transcriptomic maps of 19,052 UCB cells, covering 11 major cell types. Many of these cell types are comprised of distinct subpopulations, including distinct signatures in NK and NKT cell types in the UCB. Pseudotime ordering of nucleated red blood cells(NRBC) identifies wave-like activation and suppression of transcription regulators, leading to a polarized cellular state, which may reflect the NRBC maturation. Progenitor cells in the UBC also consist two subpopulations with divergent transcription programs activated, leading to specific cell-fate commitment. Collectively, we provide this comprehensive single-cell transcriptomic landscape and show that it can uncover previously unrecognized cell types, pathways and gene expression regulations that may contribute to the efficacy and outcome of UCB transplant, broadening the scope of research and clinical innovations.
