The aim of this research was to explore the effects and signaling pathway of ultraviolet-b(UVb)irradiation on the expression of hypoxia-inducible factor 1a(HIF-1α)and transferrin receptor(TfR).HIF-1α protein was measured by Western blot *** of epidermal growth factor receptor(EGFR),phosphor-EGF-R and TfR after UVb irradiation were determined with flow *** UVb irradiation,mRNA levels of HIF-1α and TfR were detected by real *** showed that compared with control groups,UVb was able to induce HIF1α and TfR protein expression in a dose-and time-dependent manner in HaCat cells(Pby UVb treatment(P>0.05).The EGFR/PI3K/AKT signaling pathway was required for the induction of HIF-1α and TfR expression induced by *** induced activation of EGFR in HaCat cells and EGFR regulated expression of TfR and HIF-1α.EGFR(−/−)MEF did not increase the HIF1 expression following UVb irradiation(P>0.05).In contrast,EGFR(+/+)MEF strongly enhanced HIF1a expression after UVb irradiation(Pbitor of EGFR tyrosine kinase,inhibited the TfR protein expression in UVb-treated cells in a dose-dependent manner(Pbitors,LY294002 and wortmannin,inhibited HIF-1a and TfR expressions induced by UVb(Pb irradiation(P>0.05).In contrast,DEC1(+/+)HaCat cells strongly enhanced TfR and HIF-1α protein expression after UVb irradiation(Pb induces TfR and HIF-1α expressions via EGFR/PI3K/AKT/DEC1 signaling pathway.

Proteomics Uncovers a Role for Enhanced ultraviolet-b Radiation on Wheat Leaves

Wheat (Triticum aestivum), as a kind of important economic crop cultured in the Northern China, is affected by present-day enhanced ultraviolet-b (UV-b) radiation. To get the information of the impact by UV-b radiation on it, the proteins of wheat (Jin mai NO.8) leaves, which were divided into the normal light group (CK) and UV-b radiation group (b), were extracted and ran at SDS-PAGE at different treatment days (5, 6, 7). The proteins were also analyzed by run two-dimensional gel electrophoresis (2-DE), which allowed the identification of some significantly different gel spots. The proteins spots were further verified by Matrix-Assisted Laser Desorption/lonization-time of Flight Mass Spectrometry. The results showed: 1) the enhanced UV-b affects the growth of the wheat, as the visual changes appear on the sixth day;2) the proteins expressions between the b group and the CK group were remarkably different on the sixth day;3) the proteins of wheat leaves of the sixth day were further analyzed by 2-DE revealed that twenty-one protein points were identificated between the b group and the CK group. Among these twenty-one proteins, six proteins of them were up-regulated and twelve proteins of them were down-regulated, three new proteins were expressed only in the b group. Three proteins among six proteins, which were up-regulated, were further verified as RubisCo large subunit binding protein;SOD;Calmodulin. The result indicates wheat could improve genes encoding proteins in their leaves and protect themselves, when enhanced UV-b affects the growth of the wheat.

Ascorbic acid and curcumin alleviate abnormal estrous cycle and morphological changes in cells induced by repeated ultraviolet b radiations in female Wistar rats

~ AIM： To study the effects of curcumin on the secretion of interleukin （IL） -6 and IL-8 by corneal limbus epithelial cells. METHODS： Human corneal Iimbus epithelial cells were isolated and cultured from donor eyes and irradiated by UVb at different dosages with or without curcumin. MTT test was used for studying the effects of UVb and curcumin on the cell viability. The role of mitogen-activated protein kinase （MAPK） and nuclear factor-kappa b （NF-Kb） pathways on the UVb-induced secretion of IL.6 and IL-8 were tested by addition of their inhibitors to the culture with or without UVb-radiation. Levels of various signal pathways, IL-6 and IL-8 in the cells and in the conditioned culture medium were measured by ELISA analysis. RESULTS： UVb at 20 mJ/cm2 or less and curcumin at 20 IJmol/L or less did not affect the cell viability of culturedlimbus epithelial cells （P〉0.05）. UVb irradiation at 10 and 20 mJ/cm2 induced a significant increase of secretion of IL-6 and IL-8 and upregulated NF-Kb and phosphorylated MAPK pathways of cultured limbus epithelial cells （P〈0.05）. Various signal pathway inhibitors, including SP600125 （JNK inhibitor）, Sb203580 （p38 MAPK inhibitor） and bAY11-7082 （NF-Kb inhibitor） significantly decreased the UVb-induced secretion of IL-6 and IL-8 secretion （P〈0.05）. Curcumin at 5-20 pmol/L significantly inhibited UVb- induced secretion of IL-6 and IL-8 by limbus epithelial cells in a dose-dependent manner; while curcumin alone did not affect the secretion of IL-6 and IL-8. The upregulation of NF-Kb and MAPK pathways induced by UVb treatment was significantly inhibited by curcumin, suggesting that NF-Kb and MAPK pathways are involved in the inhibitory effect of curcumin on UVb-induced production of IL-6 and IL-8.

Using agarose gel electrophoresis and T4-endo-deoxyribonuclease-V, which is a kind of restriction endonu-clease of cyclobutyl pyrimidine dimer (CPD), the impacts of He-Ne laser (5mW·mm-2) irradiation on DNA excision repair capacity in damaged wheat cells induced by enhanced ultraviolet-b (10.08kJ·m-2·d-1) radiation were studied. The results indicated that the content of endonuclease sensitive sites (ESS) was reduced by He-Ne laser irradiation, which formed in cells irradiated by enhanced ultraviolet-b. With the irradiation of He-Ne laser, the excision of CPDs and the reduction of single strand breaks (SSb) contents which were the endonuclease sensitive sites (ESS) digested by T4-endodeoxyribonuclease-Y had been stimulated in the wheat cells.

Protecting effect of He-Ne laser on winter wheat from UV-b radiation damage by analyzing proteomic changes in leaves

The ultraviolet-b (UV-b) radiation can affect gene expression of plant in growth and development. Winter wheat (Triticum aestivum), as a kind of economic crop cultured in the Northern China, is also damaged by present-day ultraviolet-b (UV-b) radiation. It was reported that He-Ne lasers could alleviate cell damage caused by UV-b radiation in plants. To get the proteomic changes of wheat alleviated by He-Ne lasers, we studied protein expression profiles of winter wheat (Jin Mai NO.79) leaves by running 2-DE (two-dimensional gel electrophoresis), which allowed the identification of some significantly different gel spots. The spots were further verified by matrix-assisted laser desorption/ionization-time of flight mass spectrometry, in which they were confirmed to be winter wheat proteins. The results showed: 1) the proteomic changes between the ultraviolet-b radiation group and normal light group were significantly different on sixth day;2) The expression of some proteins such as catalase, DNA polymerase and reduced glutathione declines at the ultraviolet-b radiation group and increases at the UV-b + He-Ne laser group. This indicates UV-b could regulate genes encoding proteins in Winter wheat leaves and affect the growth of the Winter wheat. He-Ne lasers treatment could significantly alleviate UV-b-induced damage in wheat.
