Increased heat shock protein 70 expression in the pancreas of rats with endotoxic shock

AIM： To investigate the ultra-structural changes and heat shock protein 70 （HSP70） expression in the pancreas of rats with endotoxic shock and to detect their possible relationship. METHODS： A total of 33 Wistar rats were randomly divided into three groups： control group （given normal saline）, small dose lipopolysaccharide （LPS） group （given LPS 5 mg/kg） and large dose LPS group （given LPS 10 mg/kg）. Pancreas was explanted to detect the ultrastructural changes by TEM and the HSP70 expression by immunohistochemistry and Western blot. RESULTS： Rats given small doses of LPS showed swelling and loss of mitochondrial cristae of acinar cells and increased number of autophagic vacuoles in the cytoplasm of acinar cells. Rats given large doses of LPS showed swelling, vacuolization, and obvious myeloid changes of mitochondrial cristae of acinar cells, increased number of autophagic vacuoles in the cytoplasm of acinar cells. HSPT0 expression was increased compared to the control group （P〈0.05）. CONCLUSION： Small doses of LPS may induce stronger expression of HSP70, promote autophagocytosis and ameliorate ultra-structural injuries.

Electron Microscopic Studies on the Silver-stained Nucleolar Cycle of Physarum polycephalum

The dynamic changes of nucleolar ultrastructure in the cell cycle of Physarum polycephalum Schw. were studied by an en bloc silver-staining method. The results showed that the nucleolus was large in size and situated in the center of the nucleus in late G(2)-phase, and the fibrillar centers, dense fibrillar components and granular components could be observed in the nucleolus. During prophase, the nucleolus moved towards the periphery of the nucleus and in late prophase disintegrated near the nuclear envelope. in metaphase, the disintegrated nucleolar components were dispersed in masses and located at the periphery of the chromosomal region of the nucleus. No specifically silver-stained area and argentophilic protein sheath were observed on the chromosomes, but there were some big dispersed silver particles within the chromosomes. During telophase the nucleolar components moved towards the two poles along with the chromosomes and co-existed with the decondensing chromatin in daughter nuclei. The nucleolar components then gradually converged with one another and separated from the chromatin. A big nucleolus was formed in the nucleus about 120 min after the completion of mitosis.

作者观察1例尸检,2例活检的心肌组织,发现围产期心肌病的组织学改变为灶状肌溶性坏死,病灶呈全心分布,以左室内侧密集且趋向融合,间质未见炎性渗出反应。取右室心肌活检电镜观察,可见心肌细胞有两种变化:一为肌原纤维的灶状溶解脱失,一为肌纤维内絮斑状电子密度物质沉积。闰盘呈各种变异及间隙加宽。作者未发现心肌细胞质内有任何其他沉积物,电镜下也未见任何异常的微丝聚集及高电子密度团块。通过观察,作者同意本病属多因素综合症的说法,其病变可能为机体免疫状态发生变化所导致的变应性心肌病。

The morphogenesis of NGVEV and the characteristic ultrastructural changes of DEF were studied by ultrathin sectioning and transmission electron microscopy after monolayer DEF was infected experimentally with NGVEV virulent *** investigation showed that the typical NGVEV particles were round-shaped with diameter of 75-90 nm and could be found in both the nucleus and the cytoplasm of *** mature virions contained nucleocapsid and nucleic *** virion penetrated into the DEF, replicated and matured in nucleus,and finally was released to the extracellular space via the budding and the disruption of cytoplasmic *** the appearance of progeny NGVEV,some densely electron stained virus-related structures,which were circle-shaped,U-shaped or irregular appearance,could be observed in the cytoplasm of DEF,Three kinds of cytoplasmic inclusion bodies were observed in the infected ***,it was found that NGVEV could induce DEF undergo *** morphological changes of apoptosis included the apoptotic cells shrank,chromatin condensation and margination,as well as the appearance of vacuoles on the cytomembrane aad the formation of apoptotic *** became ultracondensed and aggregated into compact clusters during apoptosis.
