Scutellaria Baicalensis Georgi is one of the most widely used traditional Chinese medicine [1]. Its roots display antioxidant, antitumor, as well as cardiovascular activity [2]. In this study, we used an in vitro model to evaluate the α-glucosidase inhibitory activity of Scutellariae Radix extract（SRE） [3], and then ultrafiltration lc-q/tof-ms/ms was applied to screen α-glucosidase inhibitors from flavonoids in SRE [4-5]. As a result, the SRE showed a stronger α-glucosidase inhibition activity with IC50 of 0.551 mg/mL（IC50 of acarbose was 1.079 mg/mL）. Thirteen compounds showed α-glucosidase inhibitory effects, which were further identified by on line lc-DAD-ms/ms compared to standards. These compounds included 2’,3,5,6’,7-pentahydroxyflavanone（1）, 2’,5,6’,7-tetrahydroxyflavane（2）, chrysin-7-O-β-D-glucopyranoside（3）, viscidulin III（4）, 2’,6’,7-trihydroxy-5-methoxyflavanone（5）, baicalein-7-O-β-D-glucopyranoside（6）, oroxylin A 7-O-β-D-glucuronide methyl ester（7）, 5,8,2’-trihydroxy-7-methoxyflavone（8）, skullcapflavone II（9）, wogonin（10）, chrysin（11）, oroxylin（12）, tenaxin I（13）. Chemical structures of these thirteen compounds were revealed at Figure 1. This study demonstrates that ultrafiltration lc-q/tof-ms/ms is a feasible and efficient technique to screen α-glucosidase inhibitors from natural products.
