Thermogenic protein ucp1 and ucp3 expression in non- small cell lung cancer： relation with glycolysis and anaerobic metabolism

Uncoupling protein 1(ucp1)is a proton transporter/channel residing on the inner mitochondrial membrane and is involved in cellular heat *** immunohistochemistry,we investigated the expression of ucp1 and ucp3 in a series of 98 patients with non-small cell lung cancer(NSCLC)treated with *** patterns were correlated with histopathological variables,prognosis,and the expression of enzymes/proteins related to cell *** epithelium did not express ucp1 or ucp3,while alveolar cells strongly expressed *** tumors,strong expression of ucp1 and ucp3 was recorded in43/98(43.8%)and 27/98(27.6%)cases,***1 was significantly associated with squamous cell histology(P=0.05),whilst ucp3 was more frequently overexpressed in large cell carcinomas(P=0.08),and was inversely related to necrosis(P=0.009).In linear regression analysis,ucp1 was directly related to markers of glycolysis[hexokinase(HXKII)and phosphofructokinase(PFK1)]and anaerobic glucose metabolism[pyruvate dehydrogenase kinase(PDK1)and lactate dehydrogenase(LDH5)].ucp3 was directly linked with a glucose transporter(GLUT2),monocarboxylate transporter(MCT2),glycolysis markers(PFK1 and aldolase),and with the phosphorylation of pyruvate dehydrogenase(p PDH).Kaplan-Meier survival analysis showed that ucp3 was significantly related to poor prognosis in squamous cell carcinomas(P=0.04).ucp1 and ucp3 are overexpressed in a large subgroup of non-small cell lung tumors and their expression coincides with increased glucose absorption,intensified glycolysis,and anaerobic glucose *** ucps are targets for therapeutic interventions in lung cancer is a hypothesis that demands further investigation.

Supplementation of Fermented Barley Extracts with Lactobacillus Plantarum dy-1 Inhibits Obesity via a ucp1-dependent Mechanism

Objective We aimed to explore how fermented barley extracts with Lactobacillus plantarum dy-1(LFBE) affected the browning in adipocytes and obese *** In vitro, 3T3-L1 cells were induced by LFBE, raw barley extraction(RBE) and polyphenol compounds(PC) from LFBE to evaluate the adipocyte *** vivo, obese SD rats induced by high fat diet(HFD) were randomly divided into three groups treated with oral gavage:(a) normal control diet with distilled water,(b) HFD with distilled water,(c) HFD with 800 mg LFBE/kg body weight(bw).Results In vitro, LFBE and the PC in the extraction significantly inhibited adipogenesis and potentiated browning of 3T3-L1 preadipocytes, rather than *** vivo, we observed remarkable decreases in the body weight, serum lipid levels, white adipose tissue(WAT) weights and cell sizes of brown adipose tissues(BAT) in the LFBE group after 10 *** group could gain more mass of interscapular BAT(IBAT) and promote the dehydrogenase activity in the *** LFBE may potentiate process of the IBAT thermogenesis and epididymis adipose tissue(EAT) browning via activating the uncoupling protein 1(ucp1)-dependent mechanism to suppress the *** These results demonstrated that LFBE decreased obesity partly by increasing the BAT mass and the energy expenditure by activating BAT thermogenesis and WAT browning in a ucp1-dependent mechanism.

【目的】探究ucp1基因的多态性与牛耐寒性之间的关联性。【方法】采集3个品种,共计180头牛的耳组织样品,利用PCR扩增和产物直接测序的方法,获得牛ucp1基因6个外显子的核苷酸序列,采用生物分析软件探讨了其遗传多态性与牛耐寒性的关联性。【结果】牛ucp1基因编码区存在8个变异位点,其中有5个错义位点,相关分析结果表明,5个错义位点多态性与牛抗寒性存在显著相关性。443位点C等位基因频率和6414位点的G等位基因频率随牛群体生活海拔高度的降低而降低;牦牛在3593位点和4873有生存优势的基因型分别为AG和AA型;4973位点CC基因型是高原牛生存的必要基因型。【结论】ucp1可能是牛抗寒性状基因之一。

PAQR9对棕色脂肪产热及ucp1表达的影响

目的初步探究孕激素和脂联素受体家族成员9(progestin and adipoQ receptor family member 9,PAQR9)对棕色脂肪产热及解偶联蛋白1(uncoupling protein 1,ucp1)表达的影响。方法选取C57BL/6J小鼠的白色脂肪组织(white adipose tissue,WAT)、棕色脂肪组织(brown adipose tissue,BAT)及前棕色脂肪细胞系DE2-3和间充质干细胞C3H10T1/2细胞系进行研究。实时荧光定量PCR技术检测不同条件下PAQR9及ucp1的mRNA水平,蛋白免疫印迹法检测ucp1的蛋白质水平。实时荧光定量PCR技术和油红O染色法检测脂肪细胞的成脂分化情况。结果 C57BL/6J小鼠BAT中PAQR9的mRNA水平显著高于WAT。冷刺激诱导下,BAT和腹股沟皮下WAT中PAQR9的mRNA水平显著升高。DE2-3细胞诱导分化成熟即第6天时,PAQR9的mRNA水平显著高表达。在DE2-3细胞中过表达PAQR9,ucp1的mRNA水平显著上调,细胞耗氧量显著增加,而DE2-3细胞成脂分化功能无显著性差异。在DE2-3细胞中敲低PAQR9,ucp1的mRNA水平显著下调。在C3H10T1/2细胞第4~6天脂肪细胞分化成熟时,PAQR9的mRNA水平显著上调,在敲低PAQR9后ucp1蛋白质水平下调,而C3H10T1/2细胞成脂分化功能无显著性差异。结论 PAQR9能够促进棕色脂肪细胞产热,调控产热基因ucp1的表达。

目的观察肥胖2型糖尿病模型OLETF鼠体质量变化和解偶联蛋白(ucps)转录基因或蛋白表达的相关性。方法测量OLETF大鼠和对照组LETO大鼠不同周龄阶段体质量,在7、10和25周时用Western blot方法测定棕色脂肪组织(BAT)中的ucp1蛋白,用Northern blot方法测定骨骼肌中ucp2和ucp3mRNA量。结果 OLETF大鼠体质量增加比LETO大鼠快,9周前后两组体质量增加差异最明显。10周时LETO组ucp1是OLETF组的1.9倍(Pucp3mRNA量约为OLETF组的5.5倍(Pucp1蛋白和ucp3基因表达的减少可能是引起OLETF大鼠肥胖的病因之一。

A better understanding of the molecular mechanisms that control the ucp1 expression in brown and beige adipocytes is essential for us to modulate adipose cell fate and promote thermogenesis,which may provide a therapeutic view for the treatment of obesity and obesity-related *** systematically identify cis-element(s)that transcriptionally regulates ucp1,we here took advantage of the high-throughput CRIPSR-Cas9 screening system,and performed an in situ saturating mutagenesis screen,by using a customized sgRNA library targeting the~20 kb genomic region near *** the screening,we have identified several genomic loci that may contain key regulatory element for ucp1 expression in cultured brown and white adipocytes in vitro,and in inguinal white adipose tissue in *** study highlights a broadly useful approach for studying cis-regulatory elements in a high-throughput manner.

为探讨中缅树鼩(Tupaia belangeri)白色脂肪组织(white adipose tissue,WAT)应对环境温度变化的适应机制,以荧光标记的ucp1和Cd137抗体分别对中缅树鼩WAT进行标记,利用流式细胞术分选分析出对照组、冷驯化28 d组、复温28 d组阳性的白色脂肪细胞。结果发现:WAT中表达ucp1、Cd137阳性的白色脂肪细胞百分数,在冷驯化组均显著高于对照组,而复温组均显著低于冷驯化组,恢复至对照组水平。表明:冷驯化诱导中缅树鼩WAT中表达ucp1、Cd137的阳性细胞群增加,显示冷驯化能诱导中缅树鼩的WAT发生褐变;复温后表达ucp1、Cd137阳性的白色脂肪细胞百分数恢复到对照水平,反映出WAT的可塑性。因此,WAT的可塑性调节也是中缅树鼩适应环境温度变化的重要产热机制。

Dear Editor,Genome-wide association studies(GWAS)revealed a large number of common variants in the human genome associated with an increased risk of *** lead signals of common obesity-associated variants are located within the first intron of the FTO(fat mass and obesity-associated)gene,being the most significant and impactive single-nucleotide polymorphism(SNP)clusters in obesity inheritance[1].Although a strong positive association between these FTO“risk SNPs”(including rs1421085 T>C)and obesity is reported across human populations of diverse ancestry,relatively weak(even none)significant associations have been observed in African populations[2].As known,the frequency of the FTO SNPs varies among different ancestral groups,with the“risk allele”being prevalent in individuals of European ancestry and less common in Asian and African ***,the linkage disequilibrium(LD)of the“risk SNPs”in African populations was much lower than in non-African populations[3].The reason for the discrepancy remains elusive thus far;however,it could be redefined in the context of the unexpected biological function of the FTO SNPs in vivo[4].

试验旨在探索前体脂肪细胞受到冷应激后的响应机制,并初步鉴定以单不饱和脂肪酸中的油酸(OA)、肉豆蔻油酸(MOA)对解偶联蛋白1(ucp1)、解偶联蛋白3(ucp3)基因表达的影响,探索以OA、MOA为代表的单不饱和脂肪酸与细胞冷应激之间是否存在关联。以OA、MOA以及延边牛前体脂肪细胞作为试验材料,分别用OA、MOA处理前体脂肪细胞96 h,通过油红O染色和三酰甘油含量的测定来评估前体脂肪细胞内脂肪生成情况,利用qPCR测定ucp1、ucp3基因的相对表达量。再分别对未经处理、OA处理96 h、MOA处理96 h的前体脂肪细胞进行4℃冷应激处理0、6、12、24 h后,分别检测其ucp1、ucp3基因的相对表达量。结果显示:OA组与MOA组均可显著降低ucp1和ucp3基因的表达水平(Pucp1基因表达量发生显著性上升(Pucp3基因表达量发生显著性降低(Pucp1基因表达量上升,而ucp3的表达量随着冷应激处理时间的延长而降低,并且试验初步验证了OA及MOA对两种基因具有抑制作用。
