All-trans retinoic acid(ATRA)inhibits insufficient radiofrequency ablation(IRFA)-induced enrichment of tumor-initiating cells in hepatocellular carcinoma

Objective:Local recurrence of hepatocellular carcinoma(HCC)after radiofrequency ablation(RFA)treatment remains a serious ***-initiating cells(TICs)are thought to be responsible for tumor ***,we investigated the effect of the TIC differentiation inducer,all-trans retinoic acid(ATRA),on RFA and explored the potential molecular ***:The proportions of CD133+and epithelial cell adhesion molecule(Ep CAM);TICs in recurrent HCC after RFA and primary HCC were first determined in ***,the effect of heat intervention or insufficient RFA(IRFA)on the malignant potential of HCC cells,including cell migration,sphere formation ability,tumor growth,the proportion of CD133+and Ep CAM+TICs and expression of stem cell-related genes,was evaluated in vitro and in ***,the effect of ATRA on the tumor growth and the proportion of TICs was ***:In clinical data,a higher proportion of CD133+and Ep CAM+TICs was found in recurrent tumors than in primary *** vitro heat intervention promoted the cell migration and sphere formation ***,it increased the proportion of CD133+and Ep CAM+TICs and the expression of stem cell-related *** addition,after IRFA the residual tumors in xenografts grew faster and had more TICs than untreated *** remarkably inhibited residual tumor growth after IRFA by elimination of TICs though the PI3 K/AKT *** treatment with ATRA resulted in longer survival outcomes in mouse xenografts than RFA ***:ATRA,as a TIC differentiation inducer,could help to improve the effect of RFA treatment,which was partially attributed to its effect against *** data indicated its potential as an alternative drug in the development of better therapeutic strategies for use in combination with RFA.

Carcinogenesis is a multistep process that requires the accumulation of various genetic and epigenetic aberrations to drive the progressive malignant transformation of normal human *** major hallmarks of carcinogenesis that have been described are angiogenesis and the stem cell characteristic of limitless replicative *** properties have been targeted over the past decade in the development of therapeutic treatments for colorectal cancer(CRC),one of the most commonly diagnosed and lethal cancers *** treatment of solid tumor cancers such as CRC has been challenging due to the heterogeneity of the tumor itself and the chemoresistance of the malignant ***,the same microenvironment that maintains the pool of intestinal stem cells that contribute to the continuous renewal of the intestinal epithelia also provides the necessary conditions for proliferative growth of cancer stem-like *** cancer stem-like cells are responsible for the resistance to therapy and cancer recurrence,though they represent less than 2.5%of the tumor *** stromal environment surrounding the tumor cells,referred to as the tumor niche,also supports angiogenesis,which supplies the oxygen and nutrients needed for tumor ***-angiogenic therapy,such as with bevacizumab,a monoclonal antibody against vascular-endothelial growth factor,significantly prolongs the survival of metastatic CRC ***,such treatments are not completely curative,and a large proportion of patient tumors retain chemoresistance or show *** article reviews the current knowledge regarding the molecular phenotype of CRC cancer cells,as well as discusses the mechanisms contributing to their *** personalized therapeutic approaches that are based on the interaction of the carcinogenic hallmarks,namely angiogenic and proliferative attributes,could improve survival and decrease adverse effects induced by unnecessary chemotherapy.

The proliferative ability of transit-amplifying progenitors needs to be tightly restricted to prevent lineage reversion and stem cell-originated ***,the molecular mechanisms limiting the ability of progenitors to self-renew remain largely *** we identify a highly conserved transcription complex B98/P56 as a key regulatory machinery for mediating progenitor dedifferentiation and stem cell-derived tumorigenesis in Drosophila neural stem cell *** show that inactivation of the B98/P56 complex specifically blocks neural progenitor dediflferentiation and consequent brain tumor formation upon Notch hyperactivation.B98/P56 expression is low in mature neural progenitors or terminally-differentiated neurons but high in NSCs,where it promotes NSC self-renewal by enhancing transcription activity of Notch *** Notch overactivation,the B98/P56complex is ectopically upregulated in dedifferentiating neural progenitor and promotes progenitor cell fate reversion by sustaining high Notch ***,a delicate interplay between the B98/P56 complex and Notch signaling distinguishes neural progenitors from NSCs and ensures correct lineage *** findings provide a mechanistic insight into the process of progenitor dedifferentiation,which may ultimately help to develop more effective anti-cancer therapy.
