Critical roles of interleukin-27(IL-27) in autoimmune diseases and infections have been reported, however, the contribution of endogenous IL-27 to tumor progression remains *** this study, by using IL-27p28 conditional knockout mice, we demonstrated that dendritic cells(DCs) were the primary source of IL-27 in vivo in protective immune response against methyl-cholanthrene induced fibrosarcoma and transplanted B16 melanoma in NK1.1+ cells-dependent *** defined that DC-derived IL-27 was critical for modulating the tumor microenvironment through promoting the expression of CXCL-10 by MDSCs, which recruited NK1.1+ cells;and also through regulating the expression of cytokines, which were key for the effec-tor function of NK1.1+ ***, reconstitution of IL-27 in tumor site significantly inhibits tumor growth and restores the number and function of NK and NKT ***, studies in breast cancer patients show that the expression level of IL-27 is decreased in advanced tumors, and positively correlates with the levels of IFN-γ, suggesting an important role of IL-27 in human antitumor immune *** summary, our study identifies a previous unknown critical role of DC-derived IL-27 in antitumor immune surveillance through shaping tumor microenvironment, and delivering IL-27 into tumor may provide a novel therapeutic approach.

CD147 is an immunoglobulin superfamily member that is present on the surface of many tumor cells and mediates malignant cell behaviors. Cyclophilin A(Cyp A) is an intracellular protein that is proinflammatory when released by cells and serves as an natural ligand for CD147. It is widely accepted that T cells play an important role in tumor immune surveillance. In this study, a CD147 specific-short hairpin RNA(sh RNA) vector was transfected into murine hepatocellular carcinoma Hepa1-6 cells to assess the effects of CD147 on the hepatoma cells escaping from immune surveillance of T cells. The results demonstrated that extracellular Cyp A stimulated cell proliferation through CD147 by activating ERK1/2 signaling pathway,downregulating expression of CD147 on Hepa1-6 cells significantly suppressed tumor progression in vivo, and decreased cell viability when co-cultured with T cells in ***, knockdown of CD147 on Hepa1-6 cells by CD147-sh RNA resulted in significantly increased T cells chemotaxis induced by Cyp A both in vivo and in *** findings provide novel mechanisms whereby tumor cells escaping from immune surveillance of T cells, and new evidence that hepatocellular carcinoma may be treated by targeting CD147 or CD147-Cyp A interactions.
