Intestinal microbes are closely associated with host health,depending on metabolic crosstalk between the microbiota and *** metabolism is one of the best examples of metabolic crosstalk between intestinal microbiota and host;however,our understanding about the influence of intestinal microbiota on host tryptophan metabolism is ***,we established germ-free(GF)pig models to systemically explore the influence of intestinal microbiota on tryptophan *** GF pigs were kept in GF conditions throughout the experiment(GF group).Six GF pigs were transplanted with fecal microbiota from donor sows to act as control *** with control pigs,the GF pigs had remarkable alterations in tryptophan *** differential metabolites(Ptryptophan metabolism varied among organs,especially for the serotonin *** GF pigs,tryptophan and kynurenine in the large intestine and 5-hydroxytryptophan in most organs were increased(Ptryptophan metabolism in GF pigs,highlighting the critical role of gut microbes in shaping host tryptophan metabolism.

To adapt to changing environments, plants have evolved elaborate regulatory mechanisms balancing their growth with stress responses. It is currently unclear whether and how the tryptophan (Trp), the growth-related hormone auxin, and the stress hormone abscisic acid (ABA) are coordinated in this trade-off. Here, we show that tryptophan synthase β subunit 1 (TSB1) is involved in the coordination of Trp and ABA, thereby affecting plant growth and abiotic stress responses. Plants experiencing high salinity or drought display reduced TSB1 expression, resulting in decreased Trp and auxin accumulation and thus reduced growth. In comparison with the wild type, amiR-TSB1 lines and TSB1 mutants exhibited repressed growth under non-stress conditions but had enhanced ABA accumulation and stress tolerance when subjected to salt or drought stress. Furthermore, we found that TSB1 interacts with and inhibits β-glucosidase 1 (BG1), which hydrolyses glucose-conjugated ABA into active ABA. Mutation of BG1 in the amiR-TSB1 lines compromised their increased ABA accumulation and enhanced stress tolerance. Moreover, stress-induced H2O2 disrupted the interaction between TSB1 and BG1 by sulfenylating cysteine-308 of TSB1, relieving the TSB1-mediated inhibition of BG1 activity. Taken together, we revealed that TSB1 serves as a key coordinator of plant growth and stress responses by balancing Trp and ABA homeostasis.

This work presents a novel electrochemical approach for detecting tryptophan through its interaction with copper nitroprusside,which is synthesized using a simple chemical co-precipitation *** utilization of the reduction reaction inherent to copper nitroprusside effectively exhibits high selectivity against common interferences present in urine,such as melatonin,lactate,cytosine,cytidine,urea,ascorbic acid,creatine,creatinine,tyrosine,glycine,alanine,arginine,and *** method demonstrates two linear ranges:0.0-0.15 mmol/L and 0.15-2.0 mmol/L with the sensitivities of 119.7±0.2μA/(mmol/L)and 9.9±0.4μA/(mmol/L),*** limit of detection(3S_(B)/m)was determined to be 5.5μmol/*** of the sensor in synthetic urine yielded the recovery of 103%±5%.

As the connecting part of diet and host physiology,intestinal microbes can convert the ingested diet into a huge number of physiologically active small *** metabolites of tryptophan are precursors or signal molecules for many biologically active substances,which are involved in serotonin and microbial catabolism *** understand the influence of tryptophan metabolism in the intestinal environment on the neurological and immune systems at the molecular level,it is important to establish a high-coverage analytical method to comprehensively analyze the metabolites involved in tryptophan ***,due to a small molecular weight and poor response during mass spectrometry analysis,as well as weak retention on the reversed-phase chromatography,determination of indole metabolites of tryptophan is ***,we proposed a method for the simultaneous determination of 20 indole metabolites of tryptophan in a single run on reversed-phase chromatography by chemical labeling coupled to liquid chromatography-tandem mass spectrometry analysis.4-(Dimethylamino)benzaldehyde(DMAB)was used for the labeling of indole metabolites of tryptophan,which could significantly improve the detection sensitivities and retention of these metabolites on reversed-phase *** the developed method,we realized the sensitive detection and comprehensive analysis of 15 endogenous indole metabolites of tryptophan in rat feces samples with functional dyspepsia intervention by *** developed method offers a useful tool for studying tryptophan metabolism-related diseases.

Recently,we read with great interest an article reporting a relationship between dietary tryptophan and the risk of obesity and type 2 diabetes(T2D).However,baseline characteristics differed among tertiles of cumulative dietary tryptophan intake in that study,which may be a confounding factor for the relationship between dietary tryptophan and the risk of obesity and T2D.

Background: tryptophan metabolites such as serotonin, kynurenine, or kynurenic acids are considered to be the most important metabolites of gut microbiota. We wanted to know about changes in tryptophan metabolites in various diseases in which the etiology gut microbiota are considered to participate. Methods: Ultra-high speed liquid chromatography/mass spectroscopy (LC/MS) has been used to analyze simultaneously all the tryptophan metabolites, which we have explored for the first time in the world. Results: We analyzed plasma levels of tryptophan metabolites in patients with depression, autism, diabetes mellitus ‘DM’), and acute coronary syndrome (ACS). Of all the metabolites serotonin and kynurenine levels of these patients were higher than those of controls. Conclusion: Measurements of tryptophan metabolites in plasma of various diseases are important to know roles of gut microbiota in etiology, further therapeutic measures.

Laser flash photolysis was used to investigate the photoinduced reactions of excited triplet bioquinone molecule duroquinone(DQ)with tryptophan(Trp)and tyrosine(Tyr)in acetonitrile-water(Me CN-H_(2)O)and ethylene glycol-water(EG-H_(2)O)*** reaction mechanisms were analyzed and the reaction rate constants were measured based on Stern-Volmer *** H-atom transfer reaction from Trp(Tyr)to ^(3)DQ^(*)is dominant after the formation of^(3)DQ^(*)during the laser *** DQ and Trp in MeCN-H_(2)O and EG-H_(2)O solutions,^(3)DQ^(*)captures H-atom from Trp to generate duroquinone neutral radical DQH^(·),carboncentered tryptophan neutral radical Trp^(·)/NH and nitrogen-centered tryptophan neutral radical Trp/N^(·).For DQ and Tyr in Me CN-H_(2)O and EG-H_(2)O solutions,^(3)DQ^(*)captures H-atom from Tyr to generate duroquinone neutral radical DQH^(·) and tyrosine neutral radical Tyr/O^(·).The H-atom transfer reaction rate constant of^(3)DQ^(*)with Trp(Tyr)is on the level of 109 L·mol^(-1)·s^(-1),nearly controlled by *** reaction rate constant of^(3)DQ^(*)with Trp(Tyr)in Me CN/H_(2)O solution is larger than that in EG/H2O solution,which agrees with Stokes-Einstein relationship qualitatively.

Chlorine dioxide(ClO2),an alternative disinfectant to chlorine,has a superior ability to inactivate microorganisms,in which protein damage has been considered as the main inactivation ***,the reactivity of ClO2 with amino acid residues in oligopeptides and proteins remains poorly *** this research,we studied the reaction rate constants of ClO2 with tryptophan residues in five heptapeptides and four proteins using stopped-flow or competition kinetic *** heptapeptide and protein contain only one tryptophan residue and the reactivity of tryptophan residue with ClO2 was lower than that of free tryptophan(3.88×10^4(mol/L)^-1 sec^-1 at pH 7.0).The neighboring amino acid residues affected the reaction rates through promoting inter-peptide aggregation,changing electron density,shifting pKa values or inducing electron transfer via redox reactions.A single amino acid residue difference in oligopeptides can make the reaction rate constants differ by over 60%(e.g.3.01×10^4(mol/L)^-1 sec^-1 for DDDWNDD and 1.85×10^4(mol/L)^-1 sec^-1 for DDDWDDD at pH 7.0(D:aspartic acid,W:tryptophan,N:asparagine)).The reaction rates of tryptophan-containing oligopeptides were also highly pH-dependent with higher reactivity for deprotonated tryptophan than the neutral *** residues in proteins spanned a 4-fold range reactivity toward ClO2(i.e.0.84×10^4(mol/L)^-1 sec^-1 for ribonuclease T1 and 3.21×10^4(mol/L)^-1 sec^-1 for melittin at pH 7.0)with accessibility to the oxidant as the determinating *** local environment surrounding the tryptophan residue in proteins can also accelerate the reaction rates by increasing the electron density of the indole ring of tryptophan or inhibit the reaction rates by inducing electron transfer *** results are of significance in advancing understanding of ClO2 oxidative reactions with proteins and microbial inactivation mechanisms.
