Characterization of a Novel 1Ay Gene and Its Expression Protein in triticum urartu

High molecular weight glutenin subunit （HMW-GS） plays an important role in determining dough property and breadmaking quality, and the exploration of novel genes for HMW-GS will be crucial for quality improvement program. A gene coding the y type HMW-GS at Glu-A1 locus in triticum urartu （AA, 2n=2×= 14） with an electrophoretic mobility similar to that of 1Dyl2, was cloned, sequenced, and heterologously expressed. This novel active lay gene FJ404595 was confirmed by structure analyses of nucleotide and deduced amino acid sequences combining with phylogenetic analysis. The open reading frame （ORF） of this gene was 1 830 bp, encoded a protein of 608 amino acid residues containing 46 hexapeptides and 14 nonapeptides, which was mostly similar to the lAy gene AM183223 at a high identity of 99.62% with the two substitutions of both leucine/proline and valine/glutamate, obviously different from the lAy gene EU984504 with 587 residues containing 44 hexapeptides and 13 nonapeptides in T. urartu. The amino acid （leucine） at 446 differed from that （proline） of all the eight compared active lAy subunits. The predicted secondary protein structure implied that this lAy subunit might also have positive impact on flour processing quality.

triticum urartu is the progenitor of the A subgenome in tetraploid and hexaploid *** the landscape of genetic variations in *** will help us understand the evolutionary and polyploid characteristics of ***,we investigated the population genomics of *** by genome-wide sequencing of 59 representative accessions collected around the world.A total of 42.2 million highquality single-nucleotide polymorphisms and 3 million insertions and deletions were obtained by mapping reads to the reference *** ancient *** population experienced a significant reduction in effective population size(Ne)from3000000 to140000 and subsequently split into eastern Mediterranean coastal and Mesopotamian-Transcaucasian populations during the Younger Dryas period.A map of allelic drift paths displayed splits and mixtures between different geographic groups,and a strong genetic drift towards hexaploid wheat was also observed,indicating that the direct donor of the A subgenome originated from northwestern *** changes were revealed between the eastern Mediterranean coastal and Mesopotamian-Transcaucasian populations in genes orthologous to those regulating plant development and stress responses.A genome-wide association study identified two single-nucleotide polymorphisms in the exonic regions of the SEMI-DWARF 37 ortholog that corresponded to the different *** ecotype *** study provides novel insights into the origin and genetic legacy of the A subgenome in polyploid wheat and contributes a gene repertoire for genomicsenabled improvements in wheat breeding.

MADS-box genes encode a large family of transcription factors that play crucial roles in plant growth and ***,till date,study on stress resistance-related functions of MADS-box genes is very limited,and identification and function of MADS-box genes remain largely unclear in triticum *** gain insight into this multigene family in ***,a whole-genome survey was employed and retrieved 68 non-redundant MADS-box *** on protein motif and phybgenetic analyses,these genes were categorized into type I(11 Mα,10 Mβ,and 5 Mγ) and type II(34 MIKC and 8 MIKC).The type II genes which mainly comprised 6-7 exons were more complex than the type I genes(mostly 1-2 exons).Cis-acting elements of the nucleotide sequences about 2000 bp before the initiation codon suggested that MADS-box genes in *** might be participated in the regulation of multiple phyto-hormones and environmental signaling ***,the expression analysis in various tissues and under low-temperature,salt,and drought conditions indicated that MADS-box genes had diverse functions in the development of ***,and most of the MADS-box genes may be involved in abiotic stress *** together,this study adds to our understanding of the roles that the MADS-box genes played in plant growth and development and stress resistance,provides a basis for further functional analyses of MADS-box gene family in cereal crops,especially wheat.

triticum urartu(2n =2x =14;AA) is the A-genome donor of cultivated tetraploid(2n=2x=28;AABB) and hexaploid wheat(2n =2x =42;AABBDD),investigating and utilizing the wild gennplasm resource could accelerate the breeding of common *** this research,to investigate the genetic diversity,population structure and evolution in triticum urartu,247 accessions,originated from seven countries and covering most of the population,were subjected to SSR and HMW-GS analysis.A set of62 highly polymorphic SSRs,located in the A genome of common wheat were screened and 1366 alleles were obtained,giving an average of 21.70 alleles per *** Fof all these polymorphic markers ranged from 0.183 for Xgwm247 to 0.794 for Cfd88 with an average of *** HMW-GS polymorphism,revealed by SDS-PAGE,resulted in 25 band patterns comprising of 4 alleles of 1Ax and 7 alleles of 1Ay *** analysis demonstrated that these accessions grouped into two subpopulations:Group Ⅰmainly collected from Lebanon,Jordan and a part of Syria,and Group Ⅱ comprising accessions from the rest of regions,which indicated that the triticum urartu accessions from Lebanon were divergent from other regions,due to the special coastal climate of Lebanon *** Coordinates Analysis(PCoA) was also performed based on the whole dataset to test the effectiveness of the discrimination between *** first two principal coordinated explained 42.3%and 17.3%of the total genetic variation respectively,with more than 59.6%of the variation in total,which revealed two main groups in the whole population.
