The position of Tree shrews in the mammalian tree:Comparing multi-gene analyses with phylogenomic results leaves monophyly of Euarchonta doubtful

The well-accepted Euarchonta grandorder is a pruned version of Archonta nested within the Euarchontoglires(or Supraprimates)clade.At present,it includes Tree shrews(Scandentia),flying lemurs(Dermoptera)and primates(Primates).Here,a phylogenomic dataset containing 1912 exons from 22 representative mammals was compiled to investigate the phylogenetic relationships within this group.Phylogenetic analyses and hypothesis testing suggested that Tree shrews can be classified as a sister group to Primates or to Glires or even as a basal clade within Euarchontoglires.Further analyses of both modified and original previously published datasets found that the phylogenetic position of Tree shrews is unstable.We also found that two of three exonic indels reported as synapomorphies of Euarchonta in a previous study do not unambiguously support the monophyly of such a clade.Therefore,the monophyly of both Euarchonta and Sundatheria(Dermoptera+Scandentia)are suspect.Molecular dating and divergence rate analyses suggested that the ancestor of Euarchontoglires experienced a rapid divergence,which may cause the unresolved position of Tree shrews even using the whole genomic data.

Improving effect of Ginkgolide B on mitochondrial respiration of ischemic neuron after cerebral thrombosis in Tree shrews

Background It has been known that platelet activating factor receptors （PAFR） may mediate many acute pathological responses and that PAFR antagonist Ginkgolide B （GB） possesses multiple effects, but the actions of GB on PAFR affinity and mitochondrial respiration in the ischemic neuron were unclear until now. This study explored the possible effects of GB on PAFR and the mitochondrial respiration of the neuron in the ischemic microenvironment. Methods Thrombotic cerebral ischemia in Tree shrews was induced by a photochemical reaction; changes in the regional cerebral blood flow （rCBF, using ^99rnTc tracer technique ）, the brain water content （specific gravimetric method）, PAFR （3H-labelled PAF assay）, the respiratory control rate （RCR）, the phosphorus-oxygen （P/O） ratio of mitochondrial respiration （Clark oxygen electrode）, mitochonddal permeability transition （MPT） pore, and the mitochondrial ultrastructure in the ischemic neurons were also observed. Data were compared between the two groups （the ischemia group vs the sham group, and the ischemia group vs the GB group）. Results There were high affinity and low affinity sites for PAFR on the tree threws＇ brain cell membranes. The varying-affinity PAFR binding sites, the respiration state Ⅲ, the state Ⅳ, RCR, the P/O ratio of the mitochondria, and the rCBF all decreased markedly （respectively, P〈0.01 and P〈0.05）, but the water content increased （P〈0.01） in the ischemia group after the application of cerebral thrombosis. In Tree shrews treated with GB （5 mg/kg i.v.） 6 hours after photochemical reaction, their PAFR binding sites and respiratory state increased markedly. The rCBF gradually increased and the brain edema ameliorated （P〈0.01） at 24h after cerebral ischemia. There were significant differences between the ischemJa group and sham group （P〈0.01）. In GB treated isolated neurons＇ mitochondria, with or without cerebral ischemia, the energy metabolism of the mitochondria had not been changed. Objective We are developing a model of depression in Tree shrews,a newly recognized nonhuman primate,using learned helplessness paradigm.Learned helplessness（LH） is widely used for screening new treat targets and the pathophysiological mechanism as an animal model of depression.The present study was to test the face and predict validity of this model.Method Male Tree shrews were subjected to 40 footshocks for 7 days,uncontrollable and inescapable（UI） or controllable and escapable（CE）.Then fluoxetine（15 mg/kg） was orally administrated for 2 weeks.On the day after stress and the day after drug treatment,depression-like behaviors were determined,including escape latency,escape failure and the breakpoint for 5%sucrose under a progressive-ratio（PR） schedule.Results One third of Tree shrews subjected to UI footshocks showed increase number of escape failure,prolonged escape latency and reduced breakpoint for 5%sucrose.Fluoxetine treatment reduced the number of escape failure and shortened the escape latency,but didn’t affect the breakpoint for sucrose.Conclusion The learned helplessness in Tree shrews shows some depression-like behaviors with individual difference,some of which reversed by a typical antidepressant.Although it requires further validation,the learned helplessness model in Tree shrews is a promising primate model for research on the etiology and pathophysiology of depression.

＜正＞Tree shrews are nocturnally sleeping,primate-related animals that have been extensively studied in a welldeveloped model of social stress.Their sleep has received minimal attention.As a first step in assessing their utility as a model of the effects of social stress on sleep,we examined sleep,activity and core body temperature recorded via telemetry under baseline and chronic stress conditions.Six adult male Tree shrews（weight 120-140 g） were intraperitoneally implanted with transmitters（DataSciences TL11M2-ETA-F20） for recording EEG,EMG,activity and core body temperature. The animals were maintained on a 12-12h light-dark cycle.Chronic stress was induced by placing two males in a single cage,but separated by wire mesh.The wire mesh was removed daily for 1.0 h immediately before lights off to allow physical contact.Behavioral states were visually scored based on EEG,EMG and activity.Over 24 h,the percentage of baseline time （Mean±SEM） spent sleeping was 51±2%for total sleep,43±2%for NREM and 7.8±0.4%for REM.During the dark period, the percentage of time spent sleeping was 78±1.2%for total sleep,75±2%for NREM and 97±1.3%for REM,whereas percentage of time with recorded activity was 9±3%.The duration of dark period NREM episodes ranged from 1.0 to 25 （mean duration:4±0.2） and the duration of dark period REM episodes ranged from 0.5 to 8（mean duration:2±0.2）.Approximately 5%of REM episodes were greater than 5 min in duration.Average core body temperature（℃） was 36.5±0.05 during the dark period and 39.2±0.8 during the light period.During the dark period,average temperatures during active wakefulness, quiet wakefulness,NREM and REM were 37.3±0.25,36.8±0.2,36.4±0.1 and 36.2±0.1,respectively.Under chronic social stress,the animals exhibited reduced activity and elevated core body temperature compared to baseline.The results confirm and describe greater nocturnal sleep in Tree shrews and provide novel data on temperature changes during sleep. Tree shrews(Tupaia belangeri chinensis)share a close relationship to primates and have been widely used in biomedical research.We previously established a spermatogonial stem cell(SSC)-based gene editing platform to generate transgenic Tree shrews.However,the influences of long-term expansion on tree shrew SSC spermatogenesis potential remain unclear.Here,we examined the in vivo spermatogenesis potential of tree shrew SSCs cultured across different passages.We found that SSCs lost spermatogenesis ability after long-term expansion(>50 passages),as indicated by the failure to colonize the seminiferous epithelium and generate donor spermatogonia(SPG)-derivedspermatocytesor spermatids marking spermatogenesis.RNA sequencing(RNA-seq)analysis of undifferentiated SPGs across different passages revealed significant gene expression changes after sub-culturing primary SPG lines for more than 40 passages on feeder layers.Specifically,DNA damage response and repair genes(e.g.,MRE11,SMC3,BLM,and GEN1)were down-regulated,whereas genes associated with mitochondrial function(e.g.,NDUFA9,NDUFA8,NDUFA13,and NDUFB8)were up-regulated after expansion.The DNA damage accumulation and mitochondrial dysfunction were experimentally validated in high-passage cells.Supplementation with nicotinamide adenine dinucleotide(NAD+)precursor nicotinamide riboside(NR)exhibited beneficial effects by reducing DNA damage accumulation and mitochondrial dysfunction in SPG elicited by long-term culture.Our research presents a comprehensive analysis of the genetic and physiological attributes critical for the sustained expansion of undifferentiated SSCs in Tree shrews and proposes an effective strategy for extended in vitro maintenance.

The use of Tree shrews as experimental animals for biomedical research is a new practice.Several recent studies suggest that Tree shrews are suitable for studying cancers,including breast cancer,glioblastoma,lung cancer,and hepatocellular carcinoma.However,the telomeres and the telomerase of Tree shrews have not been studied to date.Here,we characterize telomeres and telomerase in Tree shrews.The telomere length of Tree shrews is approximately 23 kb,which is longer than that of primates and shorter than that of mice,and it is extended in breast tumor tissues according to Southern blot and flow-fluorescence in situ hybridization(FISH)analyses.tree shrew spleen,bone marrow,testis,ovary,and uterus show high telomerase activities,which are increased in breast tumor tissues by telomeric repeat amplification protocol assays.The telomere length becomes shorter,and telomerase activity decreases with age.The tree shrew TERT and TERC are more highly similar to primates than to rodents.These findings lay a solid foundation for using Tree shrews to study aging and cancers.
