Surgical treatment for severe cubital tunnel syndrome with absent sensory nerve conduction

For severe cubital tunnel syndrome, patients with absent sensory nerve action potential tend to have more severe nerve damage than those without. Thus, it is speculated that such patients generally have a poor prognosis. How absent sensory nerve action potential affects surgical outcomes remains uncertain owing to a scarcity of reports and conflicting results. This retrospective study recruited one hundred and fourteen cases(88 patients with absent sensory nerve action potential and 26 patients with present sensory nerve action potential) undergoing either subcutaneous transposition or in situ decompression. The minimum follow-up was set at 2 years. Primary outcome measures of overall hand function included their McGowan grade, modified Bishop score, and Disabilities of the Arm, Shoulder, and Hand Questionnaire(DASH) score. For patients with absent sensory nerve action potential, 71 cases(80.7%) achieved at least one McGowan grade improvement, 76 hands(86.4%) got good or excellent results according to the Bishop score, and the average DASH score improved 49.5 points preoperatively to 13.1 points postoperatively. When compared with the present sensory nerve action potential group, they showed higher postoperative McGowan grades and DASH scores, but there was no statistical difference between the modified Bishop scores of the two groups. Following in situ decompression or subcutaneous transposition, great improvement in hand function was achieved for severe cubital tunnel syndrome patients with absent sensory nerve action potential. The functional outcomes after surgery for severe cubital tunnel syndrome are worse in patients with absent sensory nerve action potential than those without. This study was approved by the Ethical Committee of Huashan Hospital, Fudan University, China(approval No. 2017142).

Study on the Homologous Sequences of copia-like Retrotransposons in a Twin ovary Mutant of Rice

A twin ovary mutant derived from the doubled haploid (DH) progeny of a cross,02428/Gui 630, was presumably related to the transposition of some transposable elements. Up to date, all reported the active transposable elements in rice (Oryza sativa L.) are copia like retrotransposons. In the present study, the reverse transcriptase domains of copia like retrotransposons were amplified from the total DNA isolated from the mutant plants with the degenerated oligonucleotide primers for the domain. Three cloned insert DNAs, R33 1, R33 4 and R33 8, representing putative different copia like retrotransposons were screened out. Two of them displayed high polymorphism between “Zhaiyeqing 8” and “Jingxi 17”. Nine of the polymorphic bands were mapped on seven rice chromosomes. Sequencing analysis revealed that stop codons frequently occur in the sequence of R33 8, while both R33 1 and R33 4 contain a continuous coding region for 81 putative amino acid residues. No significant variation in hybridization patterns was found between indica and japonica rice or among 26 varieties of indica rice when R33 1 was used as a probe. Nevertheless, when R33 4 was used as a probe, high polymorphisms were detected both between indica and japonica rice and among 26 indica varieties, implying that this element might still be active in rice genomes.

Sustained,high level transgene expression in mammalian cells is desired in many cases for studying gene ***,stable transgene expression has been accomplished by using retroviral or lentiviral ***,such viral vector-mediated transgene expression is often at low levels and can be reduced over time due to low copy numbers and/or chromatin remodeling *** piggyBac transposon has emerged as a promising nonviral vector system for efficient gene transfer into mammalian *** its inherent advantages over lentiviral and retroviral systems,piggyBac system has not been widely used,at least in part due to their limited manipulation ***,we seek to optimize piggyBac-mediated transgene expression and generate a more efficient,user-friendly piggyBac *** engineering a panel of versatile piggyBac vectors and constructing recombinant adenoviruses expressing piggyBac transposase(PBase),we demonstrate that adenovirusmediated PBase expression significantly enhances the integration efficiency and expression level of transgenes in mesenchymal stem cells and osteosarcoma cells,compared to that obtained from co-transfection of the CMV-PBase *** further determine the drug selection timeline to achieve optimal stable transgene ***,we demonstrate that the transgene copy number of piggyBac-mediated integration is approximately 10 times higher than that mediated by retroviral *** the engineered tandem expression vector,we show that three transgenes can be simultaneously expressed in a single vector with high ***,these results strongly suggest that the optimized piggyBac system is a valuable tool for making stable cell lines with sustained,high transgene expression.

我想在这里重提一个老问题：古典希腊文化是按照自己的形象重塑了大流散时期的犹太教和基督教，还是恰恰相反，古代犹太教和基督教在很大程度上重新塑造了希腊文学传统，使之能够保有其自身的旨要？我们可以换一种方式来问这个问题(而且人们经常问这个问题)：摩西五经独有的训导以及耶稣传达的信息，是不是因为古代希腊哲学和伦理原则的注入而打了折扣？我们如何处理这些久已有之的问题，对于我们理解希腊古典传承如何影响了最早期的犹太教和基督教的《圣经》阅读者和释经者来说，有着很重要的意义。当然，我的探究针对的是有争议的话题。在古代基督徒的时代，这个话题从一开始就争论不已：使用希腊哲学的结构和表达方式作为工具，究竟在何种程度上影响到了最原初的基督教信息？很多学者进行了卷帙浩繁的论述，19世纪晚期和20世纪早期的历史学家尤为如此。其中最著名的就是上一世纪的阿道夫·冯·哈纳克(Adolf von Harnack)，他认为耶稣最早的门徒具有原始的纯朴性，这才是基督教真正的标志性特征。最早期的一代基督教信徒倚靠的是恩赐与自由，但好景不长，希腊精神很快就使之大打折扣。希腊精神原本是异己而外在的力量，却使原初的信仰转化成了一个由信条建构的庞大体系，世人称之为“基督公教”(catholicism)。“基督公教……减损了最初的信念：直接倚靠耶稣；既简要，又属灵。”①沃尔纳·耶格尔(Werner Jaeger)也是一位研究古代历史的学者，影响很大。他所持的立场却与哈纳克截然相反：对于耶格尔来说，希腊元素从一开始就在基督教中随处可见，《新约》即是一个铁证(《新约》最初的文本全部以通用希腊文写就)。修辞学的表达、伦理学和宇宙论在公元3世纪亚历山大城的基督徒知识分子圈内最为明显。亚历山大城就是基督教和希腊文化达到“终极统一”的地方。②但是，古代的情况比上述两种立场都更为微妙。犹太教和基督教内的诠释者确实采用了希腊文献和哲学，有些作家甚至表现出了“援希入耶”的超众热情，③但他们似乎是以批判的方式去这样做的，他们非常了解《圣经》传统和希腊传统之间的统一与分歧。大流散时期的犹太人和基督徒最初使用古典文献的时候，其出发点是要使摩西五经和《新约》能够回应当时的文化。因此，我们就能够论说某种智性上的连续性，它将古代犹太教、希腊文化和基督教的视野融合起来，表述共同的形而上学和道德理念。④在这些理念之中，最为突出的就是paideia的形成以及对美德的追求。

Transposable elements in cyanobacteria are briefly reviewed. Evidence is presentedto show that transposable elements in Spirulina platensis is actually reflected on the phenotype change, i.e., helical to straight filaments. transposition intermediates of DNA were isolated from the extrachromo-some and the transposition was related to helical variations in Spirulina. Uses of transposable elements formicroalgal recombination are discussed based on the transposition mechanism.

Inverse Correlations between Daily Average Energy of Global 0.01 to 1 M Earthquakes and Solar Flux Units: Possible Source of Coupling

The potential coupling between discrete releases of solar energy within the 2.8 GHz band and increments of seismic energy could suggest alternative mechanisms for solar-terrestrial interactions. Daily measures of Solar Flux Units (SFU) and the average energy of global earthquakes between 0.01 and 1 M for a 5 year period (2009-2013) shared about 25% of the same variance (r = -0.50). Average energies from the other successive 1 M intervals did not display the strength of this effect or were not statistically significant. The slope of the association indicated that for every 10-22 W/m2Hz1 decrease in daily SFUs, the average energy per earthquake in the 0.01 to 1 M range increased over the earth’s surface area on average by ~3 × 10-12 J/m2. The discrepancy of ~1010 in energy density was accommodated by multiplication of the solar unit by the frequency equivalent (1.39 × 1010 Hz/T) of the Bohr magneton and its intrinsic magnetic field strength. Although the results and the convergent quantification suggest a coupling between the mechanisms responsible for fluctuations in 2.8 GHz power from the sun and the energies of the smallest magnitude earthquake activities as predicted, the presence of a third factor related to time or to the sun’s movement through space must still be accommodated.

Memory of Familiar Melodies： Comparison Between Original and Transposed Keys

The long-term memory for musical keys of familiar melodies was investigated. An experiment was conducted focusing on memory strength, music familiarity, and key transposition using musical pieces. Participants were eighty-one Japanese undergraduate and graduate students. Eight were absolute pitch （AP） possessors and seventy-three were non-AP possessors. Two pieces of well-known classical music were selected as stimuli. These pieces were played in seven different keys： One was an original key and the other six were transposed keys in which the linear distance and harmonic distance were varied. Participants rated their strength of long-term memory for a particular segment of well-known music by comparing it with their memory of this piece. Importantly they were not required to identify the musical key of the melody. Results indicated that the strength of memory for these musical segments depended mainly on the pitch range associated with the transposed piece and partially on its key. We discussed participants＇ memory of melodies in the light of linear distance between original and transposed keys, harmonic distance between these factors, and the possibility of absolute tonality.

TnphoA and Its Use in Transposon Mutagenesis

TnphoA construct was derived from the transposon Tn5, which contained a kanamycin resistance gene flanked by two IS50 elements. This construct contains a version of the alkaline phosphatase gene with its signal sequence and promoter deleted, which will result in a blue colony phenotype on X-phos (5-bromo-4-chloro-3-indolyl phosphate toluidine salt) containing media when secreted into the periplasm. This secretion can occur only if the TnphoA has been inserted into a gene with a signal sequence which directs that gene-product to leave the cytosol. Additionally, the transposon must be inserted into the correct orientation and the same reading frame as the gene has been inserted into. TnphoA’s transposition activity derives from that of this Tn5 transposon by a conservative mechanism in a relatively rare but highly regulated process. The phoA portion of the TnphoA construct came from Escherichia coli K12 where it coded for the periplasmic protein alkaline phosphatase. This enzyme must be secreted from the cytoplasm in order to demonstrate activity and its secretion is determined by the presence of a signal sequence at its amino-terminal end. TnphoA mutagenesis is the identification of new genes that code for transmembrane or secreted proteins. TnphoA is an extremely useful construct, which along with other derivatives of the transposon Tn5 is used extensively in transposon mutagenesis based genetic analysis. TnphoA will ensure its continued significance and prominence in the area of transposon mutagenesis. TnphoA has been used to isolate new chromosomal genes, whose products are secreted, as in the case of some virulence genes in Agrobacterium tumefaciens and symbiotic genes in Rhizobium meliloti.
