To the Editor:The risk of de novo cancer increases by 2 to 7-fold after liver transplantation[1].The most common de novo cancers are both solid(non-melanoma skin cancers,colorectal carcinoma,genito-urinary and head-neck cancer)[2,3]as well as hematologic malignancies,in particular the so-called post-transplant lymphoproliferative disorder[4].

Target distribution of magnetic albumin nanoparticles containing adriamycin in transplanted rat liver cancer model

BACKGROUND: Liver cancer is one of the most common diseases around the world. The aim of this study was to verify the effect of magnetic field application on target distribution of nanoparticles in transplanted rat liver cancer model and to find out a new method for the treatment of malignant liver tumor.METHODS: Seven days after the establishment of the model, the abdomen of the rat was exposed through a midline abdominal incision. A cannula was inserted into the gastro-duodenal artery. In the experimental group (12 rats), the tumor tissue was exposed to the magnetic field for 30 minutes. Magnetic albumin nanoparticles containing adriamycin or at an equal dose of free adriamycin (0.5 mg/kg) were injected into the hepatic artery. After the magnetic field was removed, the rat was immediately sacrificed. An equal dose of nanoparticles in absence of the magnetic field served as control (12 rats). Tissues of tumor, nontargeted sites of the liver, heart, kidney, lung, spleen, stomach and small intestine were analyzed for gamma-counts and examined histologically.RESULTS: In the experimental group, the radioactivity of tumor tissue was 8.7 times that of liver tissue. In the control group, the radioactivity of tumor tissue was 2.8 times that of normal liver tissue. The radioactivity of the lung was reduced more significantly in the experimental group than in the control group. No significant difference in the kidney, heart, spleen, small intestine and stomach was observed between the experimental group and control group. And over 80% of the injected nanoparticles distributed in the liver.CONCLUSIONS: In the presence of magnetic field, magnetic albumin nanoparticles may accumulate in tumor tissues, of which the radioactivity can increase to 8.7 times that of normal liver. Even if the magnetic field is not applied, magnetic albumin nanoparticles in tumor tissues still increase to 2.8 times that of normal liver tissues. Gas exchange of Rhodiola sachalinensis transplanted from different habitats in Changbai Mountain

Rhodiola sachalinensis growing in the different habitats, Xiaotianchi plot (altitude 1800 m), Tree line plot (altitude 2000 m) and Tianwenfeng plot (altitude 2325 m), of Changbai Moutain (northern slop) were transplanted to Harbin Experimental Forest Farm for measuring its characteristic of gas exchange. The study results indicated that the growth state and gas exchange characteristics ofRh. sachalinensis growing in different habitats varied markedly. The plants transplanted from Tianwenfeng plot had the highest values in net photosynthesis rate (Pn) and transpiration rate (Tr), those transplanted from Tree line plot shows the second, and those transplanted rom Xiaotianchi plot had the lowest values. The variance existed in transplanted plants was the same as shown in the field. From the result it can be extrapolated that the difference of Pn and Tr existed inRh. sachalinensis transplanted from different habitats is depend not only on the environmental factors, but on the variance of physiological characteristic of plant itself.

Influence on radiosensitivity of lung glandular cancer cells when ERCC1 gene silenced by targeted siRNA

Objective:To identify the influence on radiosensitivity of lung glandular cancer cells when excisions repair cross-complementing group1(ERCC1) gene was silenced by targeted siR ***:siR NA which targeting to ERCC1 and control siR NA was designed and *** human lung glandular cancer SPC-A-1 cells was transfected.A total of 56 nude mice were divided into two groups,and two kinds of SPC-A-1 cells were transplanted to armpit of right forelimb,to establish the nude mice subcutaneous xenotransplanted tumor model of human lung glandular cancer *** the tumor was developed,the nude mice were randomly divided into four groups and accepted different doses of X-Ray radiation,then the change of tumor volume,survival time of mice in every group were recorded and the average lifetime was ***-one days later of X-ray experiment,two mice were taken and sacrificed in each group and the tumors organizations were *** cell apoptosis rate and cell cycle distributions were obtained by FCM(flow cytometry).Results:The volume of tumor which ERCC1 gene was silenced was less than single irradiation group after X-ray irradiation,and the growth speed was slower and the lifetime of mice was lengthened as well(P<0.05).The cells apoptosis rate and the rate of G2/M cells which ERCC1 gene was silenced were higher than the same dose control group and the rate of G_1 cells were lower,which indicated that the cells could be stopped at G_2/M point,the cell proliferation was inhibited,the cell apoptosis was promoted and the radiation sensitivity was improved after the ERCC1 was ***:The radiation sensitivity of lung glandular tumor could be improved after the ERCC1 gene was silenced by siR NA.

Study on the influence of curcumin on chemosensitivity of nephroblastoma cells

Non-random tissue distribution of human na ve umbilical cord matrix stem cells

AIM: To determine the tissue and temporal distribution of human umbilical cord matrix stem (hUCMS) cells in severe combined immunodeficiency (SCID) mice. METHODS: For studying the localization of hUCMS cells, tritiated thymidine-labeled hUCMS cells were injected in SCID mice and tissue distribution was quantitatively determined using a liquid scintillation counter at days 1, 3, 7 and 14. Furthermore, an immunofluorescence detection technique was employed in which anti-human mitochondrial antibody was used to identify hUCMS cells in mouse tissues. In order to visualize the distribution of transplanted hUCMS cells in H&E stained tissue sections, India Black ink 4415 was used to label the hUCMS cells. RESULTS: When tritiated thymidine-labeled hUCMS cells were injected systemically (iv) in female SCID mice, the lung was the major site of accumulation at 24 h after transplantation. With time, the cells migrated to other tissues, and on day three, the spleen, stomach, and small and large intestines were the major accumulation sites. On day seven, a relatively large amount of radioactivity was detected in the adrenal gland, uterus, spleen, lung, and digestive tract. In addition, labeled cells had crossed the blood brain barrier by day 1. CONCLUSION: These results indicate that peripherally injected hUCMS cells distribute quantitatively in a tissue-specific manner throughout the body.
