AIM:To rapidly detect molecular alterations in different malignancies and investigate the possible role of tp53,C-myc,and CCND1 genes in development of tumors in human organs and their adjacent normal tissues,as well as the possible relation between well-and poorly-differentiated ***:A tissue array consisting of seven different tumors was *** tissue array included 120 points of esophagus,120 points of stomach,80 points of rectum,60 points of thyroid gland,100 points of mammary gland,80 points of liver,and 80 points of *** of tp53,C-myc,and CCND1 were determined by RNA in situ hybridization.3' terminal digoxin-labeled anti-sense single stranded oligonucleotide and locked nucleic acid modifying probe were ***:The expression level of tp53 gene was higher in six different carcinoma tissue samples than in paracancerous tissue samples with the exception in colon carcinoma tissue samples(P tp53,CCND1,and C-myc genes appears to play a role in development of human cancer by regulating the expression of ***53,CCND1 and C-myc genes are significantly correlated with the development of different carcinomas.

旨在研究斑马鱼(Danio rerio)mapk1基因对tp53基因的调控作用。通过生信网站分析斑马鱼tp53启动子序列与mapk1基因CDS序列,构建斑马鱼tp53启动子荧光素酶报告基因质粒pGL3-tp53-Luc和mapk1表达质粒pCMV-Tag2B-mapk1。利用Luciferase实验验证tp53启动子报告基因的活性,以及mapk1基因对tp53基因的活性影响。结果显示,斑马鱼tp53基因启动子区无CpG岛位点,包含Oct-1(TATGTAAAGC)、Sp-1(AGATCCCGCC)、c-Jun(CTGACGTCAC)、CREB(CTGACGTCAC)、CREBP1(CTGACGTCAC)、NF-kappa B1(AGGGGAATCC)和RAR-alpha1(TTGAACTTTT)共7个转录因子结合位点;斑马鱼与人和小鼠MAPK1氨基酸的一致性分别为91.33%和90.79%。pGL3-tp53-Luc在哺乳动物细胞系和鱼类细胞系中均具有很高的活性,分别是对照组的17倍和9倍。在HEK293T细胞中过表达pCMV-Tag2B-mapk1质粒后pGL3-tp53-Luc的luciferase活性是对照组的3倍左右。实验验证了斑马鱼mapk1基因可促进tp53基因的表达。

目的探讨人群tp53基因Arg72Pro(rs1042522)位点单核苷酸多态性与甲状腺癌的相关性。方法检索PubMed、Embase、Web of Science数据库,搜索tp53基因Arg72Pro位点多态性与甲状腺癌发病风险相关的文献,筛选出符合要求的风险关联性研究,使用Stata 14.0软件进行meta分析,并进行敏感性分析、亚组分析及发表偏倚评价。采用比值比(OR)及其相应的95%CI作为效应量来评价tp53基因Arg72Pro位点单核苷酸多态性与甲状腺癌发病风险的关系。结果共计13篇文献纳入meta分析,甲状腺癌2 112例,对照4 000例。Meta分析的总体结果显示,tp53基因Arg72Pro位点的突变纯合子(Pro/Pro)基因型能够增加总人群罹患甲状腺癌的风险(隐性模型OR=1.78,95% CI 1.24~2.56,P=0.002),在甲状腺癌患者中该位点Pro突变比例较高(等位基因模型OR=1.35,95% CI 1.12~1.63,P=0.002)。亚组分析显示,该位点的突变纯合子(Pro/Pro)基因型能够增加亚洲人罹患甲状腺癌的风险,但与欧洲人及南美洲人罹患甲状腺癌无关;该位点的突变纯合子(Pro/Pro)基因型能够增加总人群罹患甲状腺乳头状癌的风险,但与总人群罹患甲状腺髓样癌无关。结论tp53基因Arg72Pro位点突变与甲状腺癌的发生相关,尤其在亚洲人群中,该位点的突变纯合子基因型Pro/Pro可能为甲状腺癌发生的危险因素。

随着二代基因检测技术的发展,人们发现与单个基因突变相比,多个体细胞共突变已被证明与预后较差有关。在这些共突变中,tp53突变最常见,但被认为是不可用药的靶点。近年来对tp53突变的预测价值进行了深入研究,由于结果不一致,尚未达到临床应用。本篇综述通过对近几年国内外关于tp53抑癌基因与EGFR共突变对非小细胞肺癌(non-small cell lung cancer,NSCLC)治疗及预后的影响进行小结,为EGFR伴随tp53突变NSCLC的靶向治疗提供新的思路。

为研究牦牛肿瘤抑制蛋白tp53(Tumor protein p53)的基因序列特征及其在牦牛卵巢中的表达情况,采集不同发情时期牦牛卵巢。根据黄牛的基因序列设计5'到3'端特异性引物,RT-PCR扩增基因克隆得到tp53基因,并对其基因结构等其他生物信息进行分析。采用Real-time PCR方法分析牦牛不同发情时期卵巢中tp53基因的表达差异。结果显示:牦牛tp53基因序列的编码区为1 161 bp,编码386个氨基酸。相似性与进化分析显示,与瘤牛tp53基因的相似性最高,达到98.4%,与家猫的相似性最低,为80.7%,表明tp53基因在进化中具有高度保守性;Real-time PCR检测结果显示,tp53基因在各个发情时期卵巢中均有表达,且表达量差异显著(Ptp53基因的表达量出现差异可能是由于在不同发情时期卵巢细胞DNA损伤程度及内分泌等因素的不同所致。牦牛tp53基因的成功克隆及生物信息学分析为该基因的进一步研究奠定了一定基础。

＜正＞[Objective]To investigate the status of tp53 mutation in six esophageal cancer cell lines and its relation with the induced expression of MGMT and MMR system,moreover to evaluate the biological behaviour of the cell lines exposed to N-methyl-N’-nitro-N-nitrosoguanidine（MNNG）.[Methods]Direct PCR product Sequencing and quantitative real-time reverse transcription PCR（RT-PCR） were applied to detect the status of tp53 mutation and the induced mRNA expression level of MGMT and MMR;simultaneously,MTT was employed to calculate the survival rate of the cells.[Results]Two esophageal cancer cell lines,EC9706 and CaEs-17,had a higher level of induced expression of MMR system and MGMT mRNA, respectively,and tolerated a lower dose of MNNG cytotoxicity compared to the other four cell lines.[Conclusion]In esophageal cancer cell lines,tp53 was not involved in the induced expression of MGMT and MMR system,and the status of which may collectively decide the sensitivity of cells to the cytotoxicity of MNNG.

目的：用cDNA Array 比较鼻咽癌组织及正常组织基因表达谱，寻找鼻咽癌组织tp53积聚的可能原因。方法：（1）Atlas Human Cancer cDNA Expression Array 7742-1滤膜杂交。（2）AtlasImage 1.01a分析滤膜杂交结果。（3）RT-PCR反应验证滤膜杂交结果。（4）免疫组化证实基因在蛋白质水平的表达改变。结果：（1）588个肿瘤相关基因中，共有134个基因表达上调，88个基因表达下调。（2）膜上有tp53调节基因32种。其中13种显示差异表达，有11个表达上调，2个表达下调。（3）tp53上游的蛋白激酶基因ATM和JNK2表达呈上调趋势。（4）泛素降解系统的两个基因ubiquitin-conjugating enzyme E2（M74524）ubiquitin-conjugating enzyme E2 （L22005）mRNA在鼻咽癌和正常鼻咽组织内的表达没有明显改变。结论：（1）在鼻咽癌组织内，tp53的功能失控。（2）ATM和JNK极有可能是tp53积聚的重要原因。

Period2(Per2) gene is an indispensable component of mammalian circadian clock *** the former study,we reported Per1 and Per2 expression abnormalities can be associated with the occurrence of *** Per1 and Per2 expression increase the efficacy of radiotherapy against glioma by promoting *** our present study,we examined what role of Per2 played under X-ray-induced DNA damage in U343 glioma cell and animal *** interference technology was used to downregulate the expression of *** to low dose of X-ray irradiation,we observed that low-level expression of Per2 can reduce DNA damage and cell death in U343 cell and glioma *** results indicated that Per2 was associated with the tp53 pathway,contributed to increasing activity of tp53,and involved DNA damaged cell in tp53-mediated *** that suggest that Per2 knockdown can inhibit the apoptosis of U343 glioma caused by X-ray irradiation.
