Background Felids are the only definitive hosts oftoxoplasma gondii. However, the biological features of the feline small intestine followingT. gondii infection are poorly understood. We investigated the changes in the expression of RNAs (including mRNAs, long non-coding RNAs and circular RNAs) in the small intestinal epithelia of cats followingT. gondii infection to improve our understanding of the life cycle ofT. gondii and cat responses toT. gondii *** Fifteen cats were randomly assigned to five groups, and the infection groups were inoculated with 600 tissue cysts of theT. gondii Pru strain by gavage. The small intestinal epithelia of cats were collected at 6, 10, 14, and 30 days post infection (DPI). Using high-throughput RNA sequencing (RNA-seq), we investigated the changes in RNA expression. The expression levels of differentially expressed (DE) genes and non-coding RNAs (ncRNAs) identified by RNA-seq were validated by quantitative reverse transcription PCR (qRT-PCR). Differential expression was determined using the DESeq R *** In total, 207 annotated lncRNAs, 20,552 novel lncRNAs, 3342 novel circRNAs and 19,409 mRNAs were identified. Among these, 70 to 344 DE mRNAs, lncRNAs and circRNAs were detected, and the post-cleavage binding sites between 725 ncRNAs and 2082 miRNAs were predicted. Using the co-location method, we predicted that a total of 235 lncRNAs target 1044 protein-coding genes, while the results of co-expression analysis revealed that 174 lncRNAs target 2097 mRNAs. Pathway enrichment analyses of the genes targeted by ncRNAs suggested that most ncRNAs were significantly enriched in immune or diseases-related pathways. NcRNA regulatory networks revealed that a single ncRNA could be directly or indirectly regulated by multiple genes or ncRNAs that could influence the immune response of cats. AIM： To analyze the biological role of the surface antigen of toxoplasma gondii（Tgondii） in development of vaccine. METHODS： The surface antigen of Tgondii （SAG1） was expressed in vitro. The immune response of the host to the antigen was investigated by detection of specific antibody reaction to SAG1 and production of cytokines. Mice were immunized with recombinant SAG1 and challenged with lethal strain of Tgondii RH. The monoclonal antibody to r-SAG1 was prepared and used to study the effects of SAG1 on Tgondii tachyzoites under electromicroscope. RESULTS： The mice immunized with recombinant SAG1 delayed death for 60 h compared to the control group. The recombinant SAG1 induced specific high titer of IgG and IgM antibodies as well as IFN-y, IL-2 and IL-4 cytokines in mice. In contrast, IL-12, IL-6 and TNF-α were undetectable. When T gondii tachyzoites were treated with the monoclonal antibody to r-SAG1, the parasites were gathered together, destroyed, deformed, swollen, and holes and gaps formed on the surface. CONCLUSION： SAG1 may be an excellent vaccine candidate against T gondii. The immune protection induced by SAG1 against Tgondii may be regulated by both hormone- and cell-mediated immune response.

toxoplasma gondii is an obligate intracellular protozoan parasite infecting a large variety of domestic and wild animals and human *** are involved in *** invasion and host cell interaction,and have been implicated as important virulence *** this study,we constructed a DNA vaccine expressing rhoptry protein 18(ROP18)of ***,and evaluated the immune response it induced in Kunming *** gene sequence encoding ROP18 was inserted into the eukaryotic expression vector p VAX *** immunized Kunming mice *** immunization,we evaluated the immune response using lymphoproliferative assay,cytokine and antibody measurements,and the survival times of mice challenged *** results showed that the group immunized with p VAX-ROP18 developed a high level of specific antibody responses against *** ROP18 expressed in ***,a strong lymphoproliferative response,and significant levels of IFN-γ,IL-2,IL-4 and IL-10 production,compared with the other groups immunized with either empty plasmid or phosphate-buffered saline,*** results demonstrate that p VAX-ROP18 induces significant humoral and cellular Th1 immune *** lethal challenge,the mice immunized with the p VAX-ROP18 showed a significantly increased survival time(27.9±15.1 days)compared with control mice which died within7 days of *** data demonstrate,for the first time,that ROP18 triggered a strong humoral and cellular response against ***,and that ROP18 is a promising vaccine candidate against toxoplasmosis,worth further development.

Seroprevalence of toxoplasma gondii among AIDS Patients in Saudi Arabia

Background: toxoplasma gondii (T. gondii) is an intracellular parasite mainly found in the central nervous system CNS, however, it can persist in multiple tissues in the body. Moreover, T. gondii is the commonest protozoans causing infections among individuals with acquired immunodeficiency syndrome (AIDS). Thus, the aim of this study was to investigate the frequency of T. gondii infection among AIDS patients in Makkah at Saudi Arabia. Methods: Fifty patients with AIDS proved to be positive by ULTRA HIV Ag-Ab Enzyme Immunoassay, and thirty healthy volunteers negative for AIDS by ULTRA HIV Ag-Ab Enzyme Immunoassay were subjected to determination of anti T. gondii immunoglobulin M (IgM) antibody seropositivity and anti T. gondii immunoglobulin G (IgG) antibody seropositivity using commercially available enzyme-linked immunosorbent assay kits. Results: The results showed that the seropositivity rate of anti T. gondii IgM antibodies among AIDS patients (18%) was significantly higher than in the healthy volunteers group (3.33%). Regarding the serum level of anti T. gondii IgG antibodies among AIDS patients, it was 30% significantly higher compared with those of the seropositive healthy volunteers (6.67%). Conclusions: These statistically significant results support the association between T. gondii infection and AIDS and suggest the usefulness of providing data for an educational program that will be designed to prevent T. gondii infection among AIDS patients.

Analysis of Proteotranscriptomics Landscape Reveals Differentially Regulated Pathways in toxoplasma gondii Infected Mouse Liver

toxoplasma gondii (T. gondii) an intracellular protozoan parasite, infects mammals including human population world-wide. Upon primary infection, the parasite contributes to mild flu like symptoms in immune competent host, but life threatening complication is seen in immune compromised patients and in pregnant women. Understanding the host-parasite interaction is critical for understanding the pathogenesis and biology parasite reactivation in the host. In this study, we used proteotrasncriptomics analyses by integrating the transcriptomics and proteomics data of T. gondii infected mouse liver to uncover the effector molecules responsible for disease pathogenesis that can be used as candidate markers for diagnosis and drug target. With this aim, we systematically integrated transcriptomicand proteomic data, representing the parasite infected mouse liver. Out of 2758 differentially expressed genes (DEGs) and 301 differentially expressed proteins (DEPs), 159 overlapping genes were identified. Among them, 86 genes were upregulated and 72 were downregulated in their respective mRNA and protein levels in the infected condition. Gene Ontology (GO) analysis revealed that the upregulated genes were mostly associated with immune system processes whereas the downregulated genes were involved in oxidation-reduction process and metabolism of lipid, and fatty acids. Protein-protein interaction (PPI) network analysis uncovered an interaction-hub including, Psmb8, Psmb9 and Tap1 for upregulated proteins and Cyp1A2, Cyp4A10 and Cyp3A11 for down-regulated proteins. Further studies are needed to validating these effector molecules. These molecules are likely to play a vital role in disease pathogenesis, as well as can be used as potential diagnostic marker and drug target candidates.

Background:toxoplasma gondii is a protozoan parasite which can infect almost all warm-blooded animals and *** the diferential expression of proteins and transcripts associated with *** infection in its defnitive host(cat)may improve our knowledge of how the parasite manipulates the molecular microenvironment of its defnitive *** aim of this study was to explore the global proteomic alterations in the major organs of cats during acute *** ***:iTRAQ-based quantitative proteomic profling was performed on six organs(brain,liver,lung,spleen,heart and small intestine)of cats on day 7 post-infection by cysts of *** PRU strain(Genotype II).Mascot software was used to conduct the student’s *** with P values1.2 or<0.83 were considered as diferentially expressed proteins(DEPs).Results:A total of 32,657 proteins were identifed in the six organs,including 2556 DEPs;of which 1325 were upregulated and 1231 were *** brain,liver,lung,spleen,heart and small intestine exhibited 125 DEPs,463 DEPs,255 DEPs,283 DEPs,855 DEPs and 575 DEPs,*** Ontology(GO)annotation and Kyoto Ency‑clopedia of Genes and Genomes(KEGG)pathway enrichment analyses of all proteins and DEPs in all organs showed that many proteins were enriched in binding,cell part,cell growth and death,signal transduction,translation,sorting and degradation,extracellular matrix remodeling,tryptophan catabolism,and immune *** between diferentially expressed proteins and transcripts were detected in the liver(n=19),small intestine(n=17),heart(n=9),lung(n=9)and spleen(n=3).Conclusions:The present study identifed 2556 DEPs in six cat tissues on day 7 after infection by *** PRU strain,and functional enrichment analyses showed that these DEPs were associated with various cellular and metabolic *** fndings provide a solid base for further in-depth investigation of the complex proteotranscript

Comparison of toxoplasma gondii seropositivity in hemodialysis and peritoneal dialysis patients

Objective:To compare the seropositivity of toxoplasma gondii in a group of peritoneal dialysis patients with hemodialysis patients and a general local population as a control group in Tabriz,Northwest ***:A total of 176 individuals were participated in the present *** them,42 were peritoneal dialysis patients,84 were hemodialysis patients and 50 were healthy ***-toxoplasma immunoglobulin G and immunoglobulin M serologic study was administered on the collected serums and then the obtained data were analyzed using statistical ***:In the present research,70.2%of hemodialysis patients,66.6%of peritoneal dialysis patients and 68%of control group had positive results for anti-toxoplasma immunoglobulin G *** individuals of the groups had negative serologic results for anti-toxoplasma immunoglobulin M *** was no significant difference between toxoplasma gondii seropositivity in hemodialysis patients and peritoneal dialysis patients and general population(P>0.05).Conclusions:The findings showed that either peritoneal dialysis or hemodialysis doesn’t increase the risk of toxoplasma seropositivity in our region(Northwest Iran).It could be explained by the fact that the present research is carried out in a high seroprevalent area scale in which the majority of normal population had previous exposure to this parasitical infection.

Low Sero-Prevalence of toxoplasma gondii IgG and IgM Antibodies in HIV Sero-Positive Patients Attending National Hospital, Abuja, Nigeria

Several researchers have investigated the association of numerous opportunistic pathogens with HIV, little is documented on its association with T. gondii in our environment. We investigated the prevalence of T. gondii immunoglobulins G and M （IgG and IgM） in HIV positive individuals in relation to their cluster of differentiation 4 （CD4） cells count. IgG, IgM and CD4 were assayed using enzyme immunoassay （EIA） and flowcytometry respectively. 341 HIV positive individuals were studied in the present research, 30 （8.7%） of them had T. gondii IgG and IgM, 297 subjects had CD4 cells count a range of 200-400 cells/μL, 27 （9.7%） and 2 （0.6%） of which had T. gondii IgG and IgM respectively. Of the 44 HIV positive subjects with CD4 〉 400 cells/μL, one （2.2%） was positive for T. gondii IgG. In the control group, all the 177 had CD4 〉 400 cells/μL of which, one （0.5%） had T. gondii IgG. The prevalence of T. gondii infection was significantly higher in HIV positive individuals than in controls （P 〈 0.05）. Male subjects in the age bracket 18-30 years had significantly higher prevalence when compared to other groups （P 〈 0.05）. Although the present findings revealed a low prevalence of T. gondii antibodies in HIV infection, this suggests that a differential toxoplasmosis diagnosis is also necessary in cases of encephalitis in HIV infection.
