Ticks are external parasitic arthropods that can transmit a variety of pathogens by sucking blood.Low-temperature tolerance is essential for Ticks to survive during the cold winter.Exploring the protein regulation mechanism of low-temperature tolerance of Haemaphysalis longicornis could help to explain how Ticks survive in winter.In this study,the quantitative proteomics of several tissues of H.longicornis exposed to low temperature were studied by data independent acquisition technology.Totals of 3699,3422,and 1958 proteins were identified in the salivary gland,midgut,and ovary,respectively.The proteins involved in energy metabolism,cell signal transduction,protein synthesis and repair,and cytoskeleton synthesis changed under low-temperature stress.The comprehensive analysis of the protein regulation of multiple tissues of female Ticks exposed to low temperature showed that maintaining cell homeostasis,maintaining cell viability,and enhancing cell tolerance were the most important means for Ticks to maintain vital signs under low temperature.The expression of proteins involved in and regulating the above cell activities was the key to the survival of Ticks under low temperatures.Through the analysis of a large amount of data,we found that the expression levels of arylamine N-acetyltransferase,inositol polyphosphate multikinase,and dual-specificity phosphatase were up-regulated under low temperature.We speculated that they might have important significance in low-temperature tolerance.Then,we performed RNA interference on the mRNA of these 3 proteins,and the results showed that the ability of female Ticks to tolerate low temperatures decreased significantly.

Crimean-Congo hemorrhagic fever(CCHF),caused by Crimean-Congo hemorrhagic fever virus(CCHFV),is endemic in Africa,Asia,and Europe,but CCHF epidemiology and epizootiology is only rudimentarily defined for most regions.Here we summarize what is known about CCHF in Central,Eastern,and South-eastern Asia.Searching multiple international and country-specific databases using a One Health approach,we defined disease risk and burden through identification of CCHF cases,anti-CCHFV antibody prevalence,and CCHFV isolation from vector Ticks.We identified 2313 CCHF cases that occurred in 1944–2021 in the three examined regions.Central Asian countries reported the majority of cases(2,026).In Eastern Asia,China was the only country that reported CCHF cases(287).In South-eastern Asia,no cases were reported.Next,we leveraged our previously established classification scheme to assign countries to five CCHF evidence levels.Six countries(China,Kazakhstan,Kyrgyzstan,Tajikistan,Turkmenistan,and Uzbekistan)were assigned to level 1 or level 2 based on CCHF case reports and the maturity of the countries’surveillance systems.Two countries(Mongolia and Myanmar)were assigned to level 3 due to evidence of CCHFV circulation in the absence of reported CCHF cases.Thirteen countries in Eastern and South-eastern Asia were categorized in levels 4 and 5 based on prevalence of CCHFV vector Ticks.Collectively,this paper describes the past and present status of CCHF reporting to inform international and local public-health agencies to strengthen or establish CCHFV surveillance systems and address shortcomings.

Objective:To green synthesize and characterize copper nanoparticles(Cu NPs)using Astragalus sinicus,as well as evaluate the acaricidal,larvacidal,and repellent activities of Cu NPs against Hyalomma anatolicum(H.anatolicum),one of the most prevalent Ticks infesting cattle in Saudi Arabia.Methods:Cu NPs were green synthesized by adding the Astragalus sinicus extract to a copper sulfate solution.The acaricidal,larvicidal,and repellent activities of Cu NPs against H.anatolicum were assessed via the adult immersion test,the larval packet test,and the vertical movement behavior of Tick larvae,respectively.The effects of Cu NPs on acetylcholinesterase as well as oxidative enzyme activities were examined.Results:The green synthesized Cu NPs displayed a spherical form with a size range of 15-75 nm.After exposure of adult H.anatolicum to different concentrations of Cu NPs,the viability rate of adult H.anatolicum and the mean number,weight,and hatchability of eggs were noticeably reduced,in comparison to the control group(P<0.001).In addition,the viability rate of larvae considerably declined(P<0.001)with the LC_(50)and LC_(90)values of 11.30 and 20.34μg/m L,respectively.The maximum repellent activity of Cu NPs was observed at 50,100,and 200μg/m L with complete repellent activity after 60,120,and 180 min of exposure,respectively.Cu NPs,mainly at 1/2 LC_(50)and LC_(50)concentrations,markedly suppressed the acetylcholinesterase activity of the larval stage of H.anatolicum(P<0.001).Moreover,Cu NPs,mainly at LC_(50)dose,significantly elevated malondialdehyde level while declining glutathione-S-transferase level in H.anatolicum larvae(P<0.001).Conclusions:Cu NPs show potent acaricidal,larvicidal,and repellent activities against adults and larvae of H.anatolicum.However,further studies must be performed to clarify the precise mechanisms and the efficacy of Cu NPs in practical use.

Background: Ticks are hematophagous parasites that transmit an extensive range of pathogens to their vertebrate hosts.Ticks can destroy invading microorganisms or alleviate infection via their rudimentary but orchestrated innate immune system.Antimicrobial peptides(AMPs)are important components of Tick innate immunity.Among these humoral effector molecules,defensins are well-studied and widely identified in various species of Ixodidae(hard Ticks)and Argasidae(soft Ticks).This review was aimed at presenting the characterization of Tick defensins from structure-based taxonomic status to antimicrobial function.Main text: All published papers written in English from 2001 to May 2022 were searched through PubMed and Web of Science databases with the combination of relevant terms on Tick defensins.Reports on identification and characterization of Tick defensins were included.Of the 329 entries retrieved,57 articles were finally eligible for our scoping review.Tick defensins mainly belong to the antibacterial ancient invertebrate-type defensins of the cis-defensins superfamily.They are generally small,cationic,and amphipathic,with six cysteine residues forming three intra-molecular disulfide bonds.Tick defensins primarily target membranes of a variety of pathogens,including Gram-positive and Gram-negative bacteria,fungi,viruses,and protozoa.Since Tick defensins have a high degree of variability,we summarize their common biological properties and enumerate representative peptides.Along with the various and potent antimicrobial activities,the role of Tick defensins in determining vector competence is discussed.Conclusions: Due to their broad-spectrum antimicrobial activities,Tick defensins are considered novel candidates or targets for controlling infectious diseases.

Objective The purpose of this study was to investigate the bacterial communities of biting midges and Ticks collected from three sites in the Poyang Lake area,namely,Qunlu Practice Base,Peach Blossom Garden,and Huangtong Animal Husbandry,and whether vectors carry any bacterial pathogens that may cause diseases to humans,to provide scientific basis for prospective pathogen discovery and disease prevention and control.Methods Using a metataxonomics approach in concert with full-length 16S rRNA gene sequencing and operational phylogenetic unit(OPU)analysis,we characterized the species-level microbial community structure of two important vector species,biting midges and Ticks,including 33 arthropod samples comprising 3,885 individuals,collected around Poyang Lake.Results A total of 662 OPUs were classified in biting midges,including 195 known species and 373 potentially new species,and 618 OPUs were classified in Ticks,including 217 known species and 326 potentially new species.Surprisingly,OPUs with potentially pathogenicity were detected in both arthropod vectors,with 66 known species of biting midges reported to carry potential pathogens,including Asaia lannensis and Rickettsia bellii,compared to 50 in Ticks,such as Acinetobacter lwoffii and Staphylococcus sciuri.We found that Proteobacteria was the most dominant group in both midges and Ticks.Furthermore,the outcomes demonstrated that the microbiota of midges and Ticks tend to be governed by a few highly abundant bacteria.Pantoea sp7 was predominant in biting midges,while Coxiella sp1 was enriched in Ticks.Meanwhile,Coxiella spp.,which may be essential for the survival of Haemaphysalis longicornis Neumann,were detected in all Tick samples.The identification of dominant species and pathogens of biting midges and Ticks in this study serves to broaden our knowledge associated to microbes of arthropod vectors.Conclusion Biting midges and Ticks carry large numbers of known and potentially novel bacteria,and carry a wide range of potentially pathogenic bacteria,which m

Objective:To estimate the infection of Ticks to Anaplasma,Ehrlichia,Babesia,Theileria,and Brucellaceae using molecular methods in borderline of Iran,Azerbaijan,and Armenia.Methods:Totally,2022 Ticks were collected from different livestock.Then,species were diagnosed under stereomicroscope according to valid morphological keys.Tick DNA was extracted followed by PCR to detect Anaplasma,Ehrlichia,Theileria,Babesia and Brucellaceae infection in Ticks.Results:A total of 498 males[24.62%(95%CI 22.76%-26.57%)],741 females[36.64%(95%CI 34.54%-38.79%)],782 nymphs[38.67%(95%CI 36.55%-40.84%)]and 1 larva[0.04%(95%CI 0.00%-0.28%)]were identified.Among identified samples,we found four genera including Hyalomma,Rhipicephalus,Haemaphysalis,and Dermacentor.Molecular assay revealed that the prevalence of Ticks to Anaplasma or Ehrlichia,and Brucellaceae was 22.02%(95%CI 16.01%-29.06%)and 15.03%(95%CI 9.43%-22.26%),respectively.Phylogenetic analysis showed that the identified Anaplasma sp.had the most similarity with Anaplasma centrale,Anaplasma platys,Anaplasma camelii,and Anaplasma phagocytophilum,submitted in GenBank.Furthermore,the detected Ehrlichia sp.and Brucellaceae bacterium had the most similarity with Ehrlichia ruminantium and Mycoplana peli,respectively.However,no sign of the presence of Theileria and Babesia spp.was seen in the studied samples.Conclusions:Anaplasmosis,ehrlichiosis and brucellosis should be considered as important health threats in northwestern Iran and consistent monitoring on infection of Ticks and livestock should be performed regularly.

To evaluate the antinociceptive and anti-inflammatory activities and the toxic effects of Rlhipicephalus microplus saliva for elucidating the modulation mechanism between arthropod saliva and host.Methods:For saliva collection,engorged Ticks were obtained from a controlled bovine infestation and collected by natural fall.The Ticks were fixed and injected pilocarpine 0.2%for induction of salivation.Saliva was collected,lyophilized and stored at-80℃.Cytotoxic activity was assessed by the hemolysis method(25,50,100,200 and 300μg/mL)and MTT cell viability assay(2.5,5,10,20 and 40μg/mL)for 24,48 and 72 h.Anti-inflammatory activity was evaluated using the method of neutrophil migration to the peritoneal cavity of mice at doses of l0,15 and 20 mg/kg;antinociceptive activity was assessed using the acetic acid-induced writhing test,and formalin-induced paw-licking in mice at dose of 15 mg/kg.Results:Saliva did not cause erythrocytes hemolysis at any concentration tested,as well as did not decrease cell viability in the MTT assay.Saliva inhibited neutrophil migration by 87%and 73%at doses of l5 and 20 mg/kg,respectively.In the nociceptive tests,saliva presented analgesic activity of69.96%in the abdominal writhing test,and of 84.41%in the formalin test.The study proves that Rlhipiceplhalus microplus saliva has significant in vivo anti-inflammatory and antinociceptive activities.The data presented herein support the development of further studies to elucidate the active principles of Rhipicephalus microplus saliva and its mechanism of action and,in future,to develop novel anti-inflammatory and analgesic drugs.

Objective:To investigate the precise species of Tick vector and the Borrelia spirochete pathogen at the Heilongjiang Province international border with Russia.Methods:In dus study,Ticks were collected from 12 Heilongjiang border crossings(including grasslands,shrublands,forests,and plantantions) to determine the rate and species type of spirochete-infected Ticks and the most prevalent spirochete genotypes.Results:The Ticks represented three genera and four species of the Ixodidae family[Ixodes persidcatus,Dermacentor silvarum,Haemapkysalis concinna and Haemaphysalis japonica].Ixodes persulcatus had the highest amount of Borrelia burgdorferi sensu lata infection of 25.6%and the most common species of Borrelia isolated from Ixodes persulcatus mas Borrelia garinii,strain PD91.Conclusions:Our results suggest that Borrelia garinii PD91- infected Ixodes persulcatus may be the principal cause of Lyme disease in the border crossing areas of Heilongjiang Province.
