Frequency-Resolved Connectome Hubs and Their test-retest reliability in the Resting Human Brain

Functional hubs with disproportionately extensive connectivities play a crucial role in global information integration in human brain networks.However,most resting-state functional magnetic resonance imaging(R-fMRI)studies have identified functional hubs by examining spontaneous fluctuations of the blood oxygen level-dependent signal within a typical low-frequency band(e.g.,0.01–0.08 Hz or 0.01–0.1 Hz).Little is known about how the spatial distributions of functional hubs depend on frequency bands of interest.Here,we used repeatedly measured R-fMRI data from 53 healthy young adults and a degree centrality analysis to identify voxelwise frequency-resolved functional hubs and further examined their test-retest reliability across two sessions.We showed that a wide-range frequency band(0.01–0.24 Hz)accessible with a typical sampling rate(fsample=0.5 Hz)could be classified into three frequency bands with distinct patterns,namely,low-frequency(LF,0.01–0.06 Hz),middle-frequency(MF,0.06–0.16 Hz),and high-frequency(HF,0.16–0.24 Hz)bands.The functional hubs were mainly located in the medial and lateral frontal and parietal cortices in the LF band,and in the medial prefrontal cortex,superior temporal gyrus,parahippocampal gyrus,amygdala,and several cerebellar regions in the MF and HF bands.These hub regions exhibited fair to good test-retest reliability,regardless of the frequency band.The presence of the three frequency bands was well replicated using an independent R-fMRI dataset from 45 healthy young adults.Our findings demonstrate reliable frequency-resolved functional connectivity hubs in three categories,thus providing insights into the frequency-specific connectome organization in healthy and disordered brains.

Aging effect and test-retest reliability of the sinusoidal harmonic acceleration test and velocity step test using nanotorque rotatory chair

Introduction:Rotatory chair testing has been used to evaluate horizontal canal function.Frequently used tests include sinusoidal harmonic acceleration test(SHAT)and velocity step test(VST).Objectives:Assessment of age effect on the SHAT and VST and assessment of test-retest reliability of the parameters of those two tests.Methods:A prospective study was performed on 100 subjects with no ear or vestibular complaints and normal vestibular evaluation.They were divided into two groups;Group A:below 50 years of age and Group B:50 years of age or above.SHAT was presented at frequencies 0.02,0.04,0.08,0.16,0.32,0.64 Hz with a peak velocity of 60°/s.VST was performed using a maximum velocity of 100°/s with acceleration and deceleration of 200°/s2.Thirty subjects were tested twice to assess reliability.Results:Study participants ranged in age from 20 to 67 years.Regarding group A,the mean age was30.92±7.31 and 55.36±4.61 for group B.No significant differences were found in SHAT parameters between the two groups.As well,there was no significant difference in VST per-rotatory time constant,however,post-rotatory time constant was significantly longer for Group B(P valuereliability(ICC 0.5800.818)for SHAT parameters for the lower frequencies and indicated moderate reliability for VST time constant(ICC 0.5090.652).Conclusions:Age has no significant effect on the parameters of SHAT and VST.test-retest reliability is generally good for both tests.

Vergence eye movements are the inward and outward rotation of the eyes responsible for binocular coordination.While studies have mapped and investigated the neural substrates of vergence,it is not well understood whether vergence eye movements evoke the blood oxygen level-dependent signal reliably in separate experimental visits.The test-retest reliability of stimulus-induced vergence eye movement tasks during a functional magnetic resonance imaging(fMRI)experiment is important for future randomized clinical trials(RCTs).In this study,we established region of interest(ROI)masks for the vergence neural circuit.Twenty-seven binocularly normal young adults participated in two functional imaging sessions measured on different days on the same 3 T Siemens scanner.The fMRI experiments used a block design of sustained visual fixation and rest blocks interleaved between task blocks that stimulated eight or four vergence eye movements.The test-retest reliability of task-activation was assessed using the intraclass correlation coefficient(ICC),and that of spatial extent was assessed using the Dice coefficient.Functional activation during the vergence eye movement task of eight movements compared to rest was repeatable within the primary visual cortex(ICC=0.8),parietal eye fields(ICC=0.6),supplementary eye field(ICC=0.5),frontal eye fields(ICC=0.5),and oculomotor vermis(ICC=0.6).The results demonstrate significant test-retest reliability in the ROIs of the vergence neural substrates for functional activation magnitude and spatial extent using the stimulus protocol of a task block stimulating eight vergence eye movements compared to sustained fixation.These ROIs can be used in future longitudinal RCTs to study patient populations with vergence dysfunctions.

Malnutrition is described as a state of insufficient intake of energy,protein and other nutrients leading to changes in body composition(weight loss,reduced fat-free mass)as well as adverse functional and clinical outcomes.[1]Depending on the assessments and definition used,the prevalence in older patients ranges between 12%in communitydwelling adults to 60%of patients in geriatric care facilities.[2–5]Older populations are at high risk of nutritional deficiencies because of risk factors such as multimorbidity,polypharmacy,cognitive and physical decline,poor appetite,depressive syndromes and socioeconomic changes.[4,6].

The free exploratory paradigm has been proposed as a model of trait anxiety.By definition,trait anxiety does not vary from moment to moment,therefore any model used to evaluate it needs to give the same results in sequential trials.The aim of this study was to evaluate the test-retest reliability of free-exploratory paradigm in mice.The behavior of drug-naive,adult,male,Kunming mice was measured in the free-exploratory apparatus on two occasions,either one week apart（Experiment I） or four weeks apart（Experiment II）.The following parameters were evaluated:percentage of time spent in the novel compartment（Ntime%）;percentage of novel compartment units visited（Ncross%）;percentage of number of rears in the novel compartment（Nrear%）;and attempts to enter the novel compartment（Attempts）.Subsequently,the intraclass correlation coefficient（ICC） and the Kappa index（k） were calculated for each of these parameters.Analysis of the parameter Nrear%in Experiment I showed the acceptable values of ICC（0.755） and κ（0.520）,respectively,indicating good and fair reliability with a one-week inter-trial interval.While in Experiment n,both ICC and k of all the parameters failed to show a reasonable test/retest concordance,suggesting that with a four-week inter-trial interval,the reliability may diminish,or even disappear.However,our results still bring further support to free-exploratory paradigm as an animal model of trait anxiety.

Background:Recent developments in virtual acoustic technology has levered promising applications in the field of auditory sciences,especially in spatial perception.While conventional auditory spatial assessment using loudspeakers,interaural differences and/or questionnaires are limited by the availability and cost of instruments,the use of virtual acoustic space identification(VASI)test has widespread applications in spatial test battery as it overcomes these constraints.Purpose:The lack of test-retest reliability data of VASI test narrows its direct application in auditory spatial assessment,which is explored in the present study.Methods:Data from 75 normal-hearing young adults(mean age:25.11 y±4.65 SD)was collected in three sessions:baseline,within 15 min of baseline(intra-session),and one week after baseline session(inter-session).test-retest reliability was assessed using the intra-class correlation coefficient(ICC),coefficient of variation(CV),and cluster plots.Results:The results showed excellent reliability for both accuracy and reaction time measures of VASI,with ICC values of 0.93 and 0.87,respectively.The CV values for overall VASI accuracy and reaction time 9.66% and 11.88%,respectively.This was also complemented by the cluster plot analyses,which showed 93.33% and 96.00% of temporal stability in the accuracy and reaction time measures,indicative of high test-retest reliability of VASI test in auditory spatial assessment.Conclusions:The high temporal stability(test-retest reliability)of VASI test validates its application in spatial hearing test battery.

AIM:To assess the reliability of web-based version of ocular surface disease index in Chinese(C-OSDI)on clinically diagnosed dry eye disease(DE)patients.METHODS:A total of 254 Chinese participants(51%male,129/254;mean age:27.90±9.06 y)with DED completed paper-and web-based versions of C-OSDI questionnaires in a randomized crossover design.Ophthalmology examination and DED diagnosis were performed prior to the participants being invited to join the study.Participants were randomly designated to either group A(paper-based first and webbased second)or group B(web-based first and paper-based second).Final data analysis included participants that had successfully completed both versions of the C-OSDI.Demographic characteristics,test-retest reliability,and agreement of individual items,subscales,and total score were evaluated with intraclass correlation coefficients(ICC),Spearman rank correlation,Wilcoxon test and Rasch analysis.RESULTS:reliability indexes were adequate,Pearson correlation was greater than 0.8 and ICCs range was 0.827 to 0.982;total C-OSDI score was not statistically different between the two versions.The values of mean-squares fit statistics were very low compared to 1,indicating that the responses to the items by the model had a high degree of predictability.While comparing the favorability 72%(182/254)of the participants preferred web-based assessment.CONCLUSION:Web-based C-OSDI is reliable in assessing DED and correlation with the paper-based version is significant in all subscales and overall total score.Webbased C-OSDI can be administered to assess individuals with DED as participants predominantly favored online assessment.

Objective Structural magnetic resonance imaging（s MRI） is a widely used way to measure the cortical volume in human brain studies, and it also shows great potentials in related rat brain studies. But to date, the test-retest reliability of cortical volume measurement for the rat brain is still unknown.Methods In this study, 37 Sprague-Dawley rat were scanned twice by s MRI with an interval of six hours, and the VBM-DARTEL method was used to automatically extract the gray matter volumes based on a rat brain atlas. After that, the intraclass coefficient（ICC）, percent volume change（PVC） and Pearson correlation coefficient（PCC） were calculated to comprehensively evaluate the test-retest reliability of every subregion in the atlas. Results The results revealed that most subregions showed excellent test-retest reliabilities and several regions displayed good test-retest reliabilities, including left/right RSGb and right RSGc.Conclusion Our results demonstrated the cortical volume measurement by s MRI using VBM-DARTEL method was reliable in the rat brain, and could be served as a convincing index for future quantitative neurobiological study.

Study Design This study proposed to carry out the cross-cultural adaptation and psychometric tests in a convenience sample of patients with cervical spondylotic myelopathy(CSM).Objective This paper aimed to translate and adapt the Japanese Orthopaedic Association Cervical Myelopathy Evaluation Questionnaire(JOACMEQ)into a simplified Chinese version.Meanwhile,its internal consistency,test-retest reliability,validity,as well as ceiling and floor effects were assessed.Summary of Background Data No simplified Chinese version of JOACMEQ had been available previously.Methods The original English version of JOACMEQ was translated and adapted to the simplified Chinese version based on the cross-cultural adaptation guideline.Meanwhile,factor structure of each subscale was verified using principal component analysis(PCA).Internal consistency was evaluated by Cronbach’sα.test-retest reliability was detected by means of intraclass correlation coefficients(ICC)among stable patients.In addition,the validity of the simplified Chinese version of JOACMEQ was assessed by examining itsrelationships with Numerical Pain Rating Scale(NPRS),Japanese Orthopaedic Association Scores(JOA),Neck Disability Index(NDI),and Short Form Health Survey(SF-36).Furthermore,the presence of ceiling and floor effects was determined when over 15%respondents had achieved the lowest or highest possible total score.Results The simplified Chinese version of JOACMEQ showed excellent internal consistency,with the Cronbach’sαof 0.909.At the same time,the ICC of test-retest reliability varied from 0.926 to 0.965.The convergent validity of the simplified Chinese version of JOACMEQ was supported by its good correlation with other physical functional status measures(NDI,JOA,and SF-36 Physical functioning;r-values:-0.705 to 0.671).Notably,Q5 Mental health was significantly correlated with SF-36 items(r-values:0.404 to 0.667).Moreover,ceiling effect could be seen in Q1 Cervical spine function(38,37.62%),Q2 Upper extremity function(34,33.67%),Q3 Lower extremity functi

Increased reaction time variability(RTV) is a characteristic of sustained attention fluctuation.The amplitude of low frequency fluctuation(ALFF) of RT has been used to assess frequency-dependent RTV in studies of sustained attention fluctuation.However,the test-retest reliability of RT-ALFF remains unclear.The present study aims to assess this issue by examining the frequency-dependent reproducibility of RT-ALFF over time and across different tasks.Thirty seven healthy college students(20.7 ±2.1 years,18 females) participated in a behavioral experiment twice,with a mean interval of 56 days(ranged from 54 to 58 days).In each visit,two tasks,i.e.,Eriksen flanker task(EFT) and simple reaction task(SRT) were performed.The order of the two tasks was counter-balanced across all participants.We employed an arrow version of EFT,in which trials were presented continuously with a fixed 3-s inter-trial interval(ITI)in order to enable frequency-dependent analysis.Participants were instructed to judge the direction(left or right) of the central arrow and press corresponding key "F" or "J" of the computer keyboard with the index fingers in the computer keyboard as accurately and quickly as possible.We also employed a fixed ITI(3-s) for the SRT.Participants were instructed to press the key "J" with their right index finger of the right hand when target displayed.Missing and anticipatory responses(RT ≤100 ms) were interpolated by linear interpolation to re-construct an integrated time series.Then,RT time series of EFT and SRT for each visit were standardized via dividing mean RT,removed of the linear trend and converted into frequency domain.Then,RT-ALFF was calculated in the whole frequency band(0-0.167 Hz) and a few sub-frequency bands including Slow-6(test-retest reliability of RT-ALFF for EFT and SRT,respectively,was evaluated through intra-class correlation(ICC) between the two visits.Linear correlation and differen
