Role of tegument proteins in herpesvirus assembly and egress

Morphogenesis and maturation of viral particles is an essential step of viral *** infectious herpesviral particle has a multilayered architecture,and contains a large DNA genome,a capsid shell,a tegument and an envelope spiked with *** to herpesviruses,tegument is a structure that occupies the space between the nucleocapsid and the envelope and contains many virus encoded proteins called tegument *** the tegument has been described as an amorphous structure,but increasing evidence supports the notion that there is an ordered addition of tegument during virion assembly,which is consistent with the important roles of tegument proteins in the assembly and egress of herpesviral *** this review we first give an overview of the herpesvirus assembly and egress *** then discuss the roles of selected tegument proteins in each step of the process,i.e.,primary envelopment,deenvelopment,secondary envelopment and transport of viral *** also suggest key issues that should be addressed in the near future.

Functional analysis of transcriptional regulation of herpes simplex virus type 1 tegument protein VP22

The herpes simplex virus type 1 (HSV-1) tegument proteins have important functions in the viral repli- cation process. In order to investigate the role of the HSV-1 tegument protein VP22 in viral replication, its transcriptional regulation of viral promoters was investigated using the chloramphenicol acetyl- transferase (CAT) assay. The results indicate that VP22 exerts a dose-dependent transcriptional in- hibitory effect on the HSV-1 α4, TK, and gC gene promoters. VP22 had the capacity to repress tran- scriptional activation of promoters via different viral transcription regulatory factors such as VP16 and ICP0, as evidenced by the specific repression of the TK and gC gene promoters by ICP0. In addition, VP22 was capable of inhibiting the promotion of ICP0 transcriptional activation in the presence of HAT PCAF, which is even more remarkable than the VP22 repression of ICP0 transcriptional activation. Fi- nally, the transcriptional inhibitory effect of VP22 on other viral promoters was demonstrated by the analysis of β-galactosidase activities in internal controls.

Laboratory diagnosis of schistosomiasis mansoni: Current status and future trends

Schistosomiasis is a neglected tropical disease that affects about 290 million patients worldwide. Children aged between 5 and 14 years represent 45.8% of the affected patients, in addition, schistosomiasis has been reported in Schistosoma-free areas, mostly because of tourism and immigration from endemic countries. Intestinal schistosomiasis caused by Schistosoma mansoni is mainly diagnosed via direct stool examination for egg detection. Immunological methods are favoured for disease monitoring and preliminary checking for communities in areas with low infection rates, and for patients with light and chronic infections where parasitological tests are negative. PCR-based diagnostic techniques are more sensitive, but expensive. tegument proteins and miRNAs are promising markers for diagnosis of schistosomiasis. Here we review the diagnostic methods for schistosomiasis mansoni aiming to reach a standardized technique for diagnosis of early infection to help better control of the disease.

The Role of Human Herpesvirus Type-6 (HHV-6) in Convulsions Seen in Children at Korle-Bu Teaching Hospital, Accra

Background: Since the isolation of HHV-6 in 1986, extensive investigation has revealed it to be ubiquitous and responsible for the majority of cases of a common febrile rash illness of infants known as roseola. Other clinical associations including seizure disorders, encephalitis and meningitis have also been stated in various publications. Objective: The aim of the study is to find out if there is any association between HHV-6 infection and the convulsions prevailing at the Child Health Department of the Korle-Bu Teaching Hospital, Accra-Ghana. Methods and Results: Children admitted into the Department of Child Health with episode of convulsions were recruited after informed consent had been sought from subjects. Cerebrospinal fluid (CSF) and Plasma were obtained from patients. PCR directed at the detection of the large tegument protein (LTP) gene in the SIE strain of the HHV-6 in Plasma and CSF from patients was done. The mean age of study subjects was 37.44 months with 53 (64.6%) being males. There was a significant relationship between the convulsions and fever (P < 0.05). Based on CSF characteristics gathered, viral infections may be the probable cause of the observed convulsions but not malaria or bacterial infections. None of the samples from the patients had evidence of HHV-6. Conclusion: The study was unable to establish HHV-6 infection in the CSF and Plasma of patients. What role if any HHV-6 has in convulsions seen in children or neurological diseases at large merits further studies. Other neurotropic viruses need to be investigated as possible causes for the convulsions.

Evaluation of the in vivo effect of ivermectin on Schistosoma mansoni in experimentally-infected mice

Objective:To evaluate the effect of ivermectin on mice experimentally infected with Schistosoma ***:Ivermectin was given to mice 42 days *** two treatment regimens:a single dose of 25 mg/kg or the same dose for two consecutive ***:In both regimens,there were significant reductions in female worms,hepatic tissue egg count and early immature ova,in addition to significant reductions in the count and size of hepatic *** same dose for two consecutive days resulted in significant reductions in male,total worms,and intestinal tissue egg *** both regimens,scaning electron microscopy revealed tegumental alternations as bleb formation,erosion and necrosis more evident with the double *** addition,high dose showed extensive erosions of male worms with destruction of the gynaecophoric *** females,marked destruction of the tegumental surface was extending to both ventral and oral ***:lvermnectin has promising anti--schistosomal ***,further research is needed to test the effect of ivermectin on schistosomiasis especially in combination with other antischistosomal agents,to avoid any possible resistance from monotherapy.
