＜正＞Spontaneous calcium transient has been reported in early development of many species. It drives many kinds of cellular processing, including cell differentiation, axon outgrowth and pathfinding and so on. These processing are important for normal maturation of neural network. The underlying mechanism is known litter. Here we used calcium-imaging technique to explore spontaneous calcium transient in dissociated, brain of Drosophila. We found that the photoreceptor in early development has spontaneous calcium transient, which is trigger by extracellular calcium. In the other hand, the frequency of spontaneous calcium activity was dramatically increased in sodium free saline, which is probably driven by reverse mode of calcium-sodium exchange. We also want to find out the channel through which extracellular calcium influx. It is not Ih channel since cesium cannot influence this kind of calcium activity. trp and trpl channels are important calcium channel in photoreceptor of the fruitfly. We will test these channels by using trp or trpl mutant flies.
