Introduction:Accumulating evidence certifies that histones acetylation and deacetylation aberrations play an important role in the pathogenesis of rheumatoid arthritis(RA).Studies on the activity of histone deacetylases(HDACs) in RA have provided conflicting results,and most of these paradoxical conclusions about histone modifications are from and as yet quite limited in RA synovial cells(RASFs).Purpose:To investigate the expression and activity of class Ⅰ HDACs(1-3.8) in peripheral blood mononuclear cells(PBMCs) of RA and its effect on histone acetylation and the correlation with disease ***:The mRNA expressions were examined by quantitative real-time *** protein expression of class Ⅰ HDACs were examined by Western *** of histone acetylases(HATs) and HDACs was measured in nuclear extracts of peripheral blood mononuclear cells(PBMCs) by Activity Assay *** total Histone H3 and H4 acetylation were measured by enzyme linked immunosorbent assay(Elisa) ***:The mRNA expression and protein level of all class Ⅰ HDACs in PBMCs from RA patients was negative correlation with CRP,ESR and *** increased nuclear HAT activities and decreased HDAC activity(especially HDAC3) were discovered in RA than in HCs(P<0.0001).The total Histone H3,but not H4,acetylation degree in PBMCs from RA patients is increased compared to HCs(p=0.0074).Conclusion:HDACs/HATs activity is unbalance and histone acetylation is intensive in RA *** acetylation was involved in the disease progression of RA.
